ERK1 and ERK2 are involved in recruitment and maturation of human mesenchymal stem cells induced to adipogenic differentiation

Adipocytes’biology and the mechanisms that control adipogenesis have gained importance because of the need to develop therapeutic strategies to control obesity and the related *** mesenchymal stem cells(hMSCs),undifferentiated stem cells present in the bone marrow that are physiological precursors of adipocytes,were induced to adipogenic *** molecular mechanisms on the basis of the adipogenesis were evaluated,focusing on the MAPKinases ERK1 and ERK2,which are involved in many biological and cellular ***1 and ERK2 phosphorylation was reduced with different timing and intensity for the two isoforms in treated hMSCs in comparison with control cells until day 10 and then at 14–28 days,it reached the level of untreated *** total amount of ERK1 was also decreased up to day 10 and then was induced to the level of untreated cultures,whereas the expression of ERK2 was not changed following adipogenic *** with the specific ERK1/2 inhibitor U0126 during the whole differentiation period hampered hMSCs’adipogenic differentiation,as lipid droplets appeared in very few cells and were reduced in number and *** U0126 was administered only during the initial phase of differentiation,the number of hMSCs recruited to adipogenesis was reduced while,when it was administered later,hMSCs did not acquire a mature adipocytic ***1 and ERK2 are important for hMSC adipogenic differentiation since any alteration to the correct timing of their phosphorylation affects either the recruitment into the differentiation program and the extent of their maturation.

The association between spondyloarthropathy and in flammatory bowel disease(IBD) is largely established, although prevalence is variable because of different population selection and diagnostic *** studies indicate that as many as 10%15% of cases of IBD are complicated by ankylosing spondylitis(AS) or other forms of spondylarthritis(SpA).Of note, ileal inflammation resembling IBD has been reported in up to two thirds of cases of SpA, and it has been suggested that the presence of ileitis is associated with the chronic ity of articular *** this observation is of interest to unravel the pathophysiology of the disease, systematic screening of patients with SpA by ileocolonos copy is not indicated in the absence of gut symptoms, as only a small proportion of patients with subclinical gut inflammation will develop overt IBD over *** existence of familial clustering of both IBD and AS, the coexistence of both conditions in a patient, the evidence of an increased risk ratio among f irstand seconddegree relatives of affected AS or IBD patients and f inally, the increased crossrisk ratios between AS and IBD, strongly suggest a shared genetic *** far, however, IL23R is the only identified susceptibility gene shared by both IBD and *** functional studies are still needed to better understand its pathogenic role, great ef fort is being spent therapeutically targeting this pathway that may prove effective for both disorders.

Role of autophagy in cholangiocarcinoma:Pathophysiology and implications for therapy

Cholangiocarcinoma(CCA)is a malignant tumour of the biliary system that originates from the neoplastic transformation of *** is characterized by late diagnosis and poor outcome,with surgery considered as the last option for *** is a physiological lysosomal degradation process,essential for cellular homeostasis and ubiquitous in all eukaryotic *** studies have reported a potential involvement of autophagy in cancer,but it remains unclear whether activation of this process represents a survival mechanism of cancer *** the present review,we examine the autophagic process and summarize the current knowledge about the involvement of autophagy in the progression of *** link between autophagy and chemoresistance and the use of autophagic markers in diagnosis are also considered in *** evidence shows that the combination of autophagy modulators(activators or inhibitors)with conventional chemotherapeutic agents offers a possible treatment option against signalling pathways that are hyperactivated or altered in *** vitro evidence suggests that combination of chemotherapy agents,such as cisplatin,under activation or inhibition of autophagic processes,in two different CCA cell lines,may improve chemosensitivity and reduce cell survival,respectively.A deeper understanding of these pathways,in both cancer and non-cancer cells,could unveil possible therapeutic targets to treat CCA patients.

Novel technique of extracorporeal intrauterine morcellation after total laparoscopic hysterectomy: Three emblematic case reports

BACKGROUND In the presence of a large uterus,total laparoscopic hysterectomy(TLH),always requires morcellation to allow removal of the tissues from the abdominal ***,uncontained morcellation has been scrutinized because of the possible spread of occult ***,in-bag extracorporeal morcellation has been ***,tissue containment and extraction are extremely challenging,especially when considering the increasing uterine size to be removed through minimally invasive *** SUMMARY Herein,we describe a novel technique for extracorporeal intrauterine morcellation using the uterus outermost layer as a bag to achieve tissue extraction of very large uteri with suspected occult leiomyosarcoma after *** study enrolled patients who were planned for TLH for large uteri(weight>500 g).TLH was performed following the procedure reported in our previous *** novel technique has been described step-by-step in a video,which representatively describes the preoperative imaging and morcellation procedure of three very large uteri weighing 1500 g,1700 g,and 3700 g,*** procedures were performed without any *** patients had an uneventful postoperative course,and in all cases,the pathology was benign *** Extracorporeal intrauterine morcellation using the uterus outmost layer as a bag was found to be a feasible technique that allows a careful diagnosis and safe removal of suspected occult *** technique herein presented may be adopted in surgical practice,by adding it to the other available techniques of contained *** may represent a valid and feasible alternative,especially useful in cases of very large uteri exceeding the capacity of specimen retrieval bags.

Microtubule Depolymerization Affects Endocytosis and Exocytosis in the Tip and Influences Endosome Movement in Tobacco Pollen Tubes

Polarized organization of the cytoplasm of growing pollen tubes is maintained by coordinated function of actin filaments （AFs） and microtubules （MTs）. AFs convey post-Golgi secretory vesicles to the tip where some fuse with specific domains of the plasma membrane （PM）. Secretory activity is balanced by PM retrieval that maintains cell mem- brane economy and regulates the polarized composition of the PM, by dividing lipids/proteins between the shank and the tip. Although AFs play a key role in PM internalization in the shank, the role of MTs in exo-endocytosis needs to be characterized. The present results show that integrity of the MT cytoskeleton is necessary to control exo-endocytosis events in the tip. MT polymerization plays a role in promoting PM invagination in the apex of tobacco pollen tubes since nocodazole affected PM internalization in the tip and subsequent migration of endocytic vesicles from the apex for degradation. MT depolymerization in the apex and shank was associated with misallocation of a significantly greater amount of internalized PM to the Golgi apparatus and its early recycling to the secretory pathway. Fluorescence Recovery After Photobleaching （FRAP） experiments also showed that MT depolymerization in the tip region influenced the rate of exocytosis in the central domain of the apical PM.
