Inhibition of guanosine triphosphate（GTP） and cytidine triphosphate（CTP） biosynthetic pathways induces cells to assemble rod/ring（RR） structures,also named cytoophidia,which consist of the enzymes cytidine triphosphate synthase（CTPS） and inosine-50-monophosphate dehydrogenase 2（IMPDH2）.We aim to explore the interaction of CTPS and IMPDH2 in the generation of RR *** la and COS-7 cells were cultured in normal conditions or in the presence of 6-diazo-5-oxo-l-norleucine（DON）,ribavirin,or mycophenolic acid（MPA）.Over 90% of DON-treated cells presented RR *** He la cells,35% of the RR structures were positive for IMPDH2 alone,26% were CTPS alone,and 31% were IMPDH2/CTPS mixed,while in COS-7 cells,42% of RR were IMPDH2 alone,41% were CTPS alone,and 10% were IMPDH2/CTPS *** and MPA treatments induced only IMPDH2-based *** were also transfected with an N-terminal hemagglutinin（NHA）-tagged CTPS1 *** 95% of NHA-CTPS1 transfected cells with DON treatment presented IMPDH2-based RR and almost 100% presented CTPS1-based RR;when treated with ribavirin,over 94% of transfected cells presented IMPDH2-based RR and 37% presented CTPS1-based RR,whereas 2% of untreated transfected cells presented IMPDH2-based RR and 28% presented CTPS1-based *** results may help in understanding the relationship between CTP and GTP biosynthetic pathways,especially concerning the formation of filamentous RR structures.

Nitrogen(N)and potassium(K)are the most required nutrients for corn and wheat *** the N application rate usually boosts crop ***,many uncertainties remain for K *** deficiency results in yield losses,but K application rate based on the percentage of K+in the cation exchange capacity(CEC)is doubtful,especially in soil with high CEC.A field trial was conducted to examine the effects of KCl application before sowing corn and wheat,by raising the percentage of K+in CEC at pH 7.0(CECpH7.0)to approximately 2.5%,3.5%,and 4.5%,and adding N as a topdressing(75,150,and 225 kg ha-1 to corn and 40,80,and 120 kg ha-1 to wheat)on the nutrition and yield of corn and wheat under a continuous no-till system(30 years).Exchangeable K+content increased in the topsoil(0–20 cm depth)up to 7.2 mmolc dm-3 after K application at the highest rate,which,however,did not result in significant increases in nutrient uptake and yields for both corn and *** N application rate positively affected the uptake and removal of all macronutrients by corn and *** N as a topdressing increased yields of corn and wheat by up to 83%and 22%,*** results suggest that in the soil with a high CECpH7.0(162.1 mmolc dm-3),the recommendation for K application made by considering the percentage of K+in the CECpH7.0 may result in excessive application of K fertilizer to crops with high K-recycling potential grown under a continuous no-till system.

The molecular mechanisms controlling mouse embryonic stem cell(ESC)metastability,*** capacity to fluctuate between different states of pluripotency,are not fully *** developed and used a novel automation platform,the Cellmaker,to screen a library of metabolites on two ESC-based phenotypic assays(*** and colony phenotype)and identified two metabolically related amino acids,namely l-proline(l-Pro)and l-ornithine(l-Orn),as key regulators of ESC *** compounds,but mainly l-Pro,force ESCs toward a novel epiblast stem cell(EpiSC)-like state,in a dose-and time-dependent *** EpiSCs,l-Pro-induced cells(PiCs)contribute to chimeric embryos and rely on leukemia inhibitor factor(lIF)to ***,PiCs revert to ESCs or differentiate randomly upon removal of either l-Pro or lIF,***,PiC generation depends on both l-Pro metabolism(uptake and oxidation)and Fgf5 induction,and is strongly counteracted by antioxidants,mainly l-ascorbic acid(vitamin C,Vc).ESCs↔PiCs phenotypic transition thus represents a previously undefined dynamic equilibrium between pluripotent states,which can be unbalanced either toward an EpiSC-like or an ESC phenotype by l-Pro/l-Orn or Vc treatments,*** together,our data provide evidence that ESC metastability can be regulated at a metabolic level.

AIM To examine the association between weekend alcohol consumption and the biochemical and histological alterations at two different concentrations of alcohol in both genders in *** Wistar rats weighing 170-200 g were divided into groups as follows:(1) Control groups; and(2) weekend alcohol-consumption group: 2 d/weekly per 12 wk, at two different concentrations:(1) Group of males or females with a consumption of a solution of alcohol at 40%; and(2) group of males or females with a consumption of a solution of alcohol at 5%. At the end of the experiment, serum and liver samples were obtained. The following enzymes and metabolites were determined in serum: Alanine Aminotransferase(AlT), Aspartate Aminotransferase(AST), lactate Dehydrogenase, and Gamma-Glutamyltransferase, and glucose, triglycerides, cholesterol, bilirubin, and albumin. liver samples from each group were employed to analyze morphological abnormalities by light *** In all of the weekend alcohol-consumption groups, AST activity presented a significant, 10-fold rise. Regarding AlT activity, the groups with weekend alcohol consumption presented a significant increase that was six times greater. Bilirubin levels increased significantly in both groups of females. We observed a significant increase in the parameters of fatty change and inflammation due to weekend alcohol consumption. Only the group of females that consumed alcohol at 40% presented slight hepatocel ular disorganization CONClUSION The results obtained herein provide solid evidence that weekend alcohol consumption gives rise to liver damage, demonstrated by biochemical and histological alterations, first manifested acutely, and prolonged weekend alcohol consumption can cause greater, irreversible damage.

Background In beef cattle, more than 50% of the energy input to produce a unit of beef is consumed by the female that produced the calf. Development of genomic tools to identify females with high genetic merit for reproductive function could increase the profitability and sustainability of beef *** Genome-wide association studies(GWAS) were performed using a single-step genomic best linear unbiased prediction approach on pregnancy outcome traits from a population of Angus–Brahman crossbred heifers. Furthermore, a validation GWAS was performed using data from another farm. Heifers were genotyped with the Bovine GGP F250 array that contains 221,077 SNPs. In the discovery population, heifers were bred in winter breeding seasons involving a single round of timed artificial insemination(AI) followed by natural mating for 3 months. Two phenotypes were analyzed: pregnancy outcome to first-service AI(PAI;n = 1,481) and pregnancy status at the end of the breeding season(PEBS;n = 1,725). The heritability was estimated as 0.149 and 0.122 for PAI and PEBS, respectively. In the PAI model, one quantitative trait locus(QTl), located between 52.3 and 52.5 Mb on BTA7, explained about 3% of the genetic variation, in a region containing a cluster of γ-protocadherin genes and SlC25A2. Other QTls explaining between 0.5% and 1% of the genetic variation were found on BTA12 and 25. In the PEBS model, a large QTl on BTA7 was synonymous with the QTl for PAI, with minor QTls located on BTA5, 9, 10, 11, 19, and 20. The validation population for pregnancy status at the end of the breeding season were Angus–Brahman crossbred heifers bred by natural mating.
