Modified mesenchymal stem cells in cancer therapy: A smart weapon requiring upgrades for wider clinical applications

Mesenchymal stem stromal cells(MSC)are characterized by the intriguing capacity to home toward cancer cells after systemic ***,MSC can be harnessed as targeted delivery vehicles of cytotoxic agents against *** cancer patients,MSC based advanced cellular therapies were shown to be safe but their clinical efficacy was ***,the amount of systemically infused MSC actually homing to human cancer masses is insufficient to reduce tumor ***,induction of an unequivocal anticancer cytotoxic phenotype in expanded MSC is necessary to achieve significant therapeutic *** vivo cell modifications are,thus,required to improve anti-cancer properties of *** based cellular therapy products must be handled in compliance with good manufacturing practice(GMP)*** the present review we include MSCimproving manipulation approaches that,even though actually tested at preclinical level,could be compatible with GMP *** particular,we describe possible approaches to improve MSC homing on cancer,including genetic engineering,membrane modification and cytokine ***,we discuss appropriate modalities aimed at inducing a marked cytotoxic phenotype in expanded MSC by direct chemotherapeutic drug loading or by genetic *** conclusion,we suggest that,to configure MSC as a powerful weapon against cancer,combinations of clinical grade compatible modification protocols that are currently selected,should be introduced in the final *** standardized cancer clinical trials are required to test the efficacy of ameliorated MSC based cell therapies.

Imperfect mimicry of host begging calls by a brood parasitic cuckoo:a cue for nestling rejection by hosts?

Coevolutionary interactions between avian brood parasites and their hosts often lead to the evolution of discrimination and rejection of parasite eggs or chicks by hosts based on visual cues,and the evolution of visual mimicry of host eggs or chicks by brood *** may also base rejection of brood parasite nestlings on vocal cues,which would in turn select for mimicry of host begging calls in brood parasite *** cuckoos that exploit multiple hosts with different begging calls,call structure may be plastic,allowing nestlings to modify their calls to match those of their various hosts,or fixed,in which case we would predict either imperfect mimicry or divergence of the species into host-specific *** our study of the little bronze-cuckoo(LBC)Chalcites minutillus and its primary host,the large-billed gerygone Gerygone magnirostris,we tested whether:(1)hosts use nestling vocalizations as a cue to discriminate cuckoo chicks;(2)cuckoo nestlings mimic the host begging calls throughout the nestling period;and(3)the cuckoo begging calls are plastic,thereby facilitating mimicry of the calls of different *** found that the begging calls of LBCs are most similar to their gerygone hosts shortly after hatching(when rejection by hosts typically occurs)but become less similar as cuckoo chicks get *** call structure may be used as a cue for rejection by hosts,and these results are consistent with gerygone defenses selecting for age-specific vocal mimicry in cuckoo *** found no evidence that LBC begging calls were plastic.

The Effect of Overseas Educational Experience on Pro-Environmental Practices: Evidence from Cambodian Academic Scholars

This paper revealed that after spending years overseas for learning, Cambodian fellows realized that a clean society more likely results from the adopting of a waste management system. The lack of such is the main barrier for waste separation willingness and practices in Cambodia. Self-transcendence values and volunteerism are empirically found as the fundamental factors for any interventions designed to promote pro-environmental intentions and practices respectively. The fellows engaging in volunteerisms appear to do waste separation, ignoring how inconvenient the waste management system or facilities are. Similarly, the fellows concerning the environmental and social issues appear to spare their valuable time to do waste-separation which they find not difficult. This is not the case in this COVID-19 epidemic though. The study results imply that in a society where an inconvenient waste management system is in place the fellows without past volunteering, no matter how knowledgeable they are, will less likely engage in the waste-sorting practice.

Generation and expansion of laser-induced plasma as a spectroscopic emission source

Laser-induced plasma represents today a widespread spectroscopic emission source. It can be easily generated using compact and reliable nanosecond pulsed laser on a large variety of materials. Its application for spectrochemical analysis for example with laser-induced breakdown spectroscopy （LIBS） has become so popular that one tends to forget the complex physical and chemical processes leading to its generation and governing its evolution. The purpose of this review article is to summarize the backgrounds necessary to understand and describe the laser-induced plasma from its generation to its expansion into the ambient gas. The objective is not to go into the details of each process; there are numerous specialized papers and books for that in the literature. The goal here is to gather in a same paper the essential understanding elements needed to describe laser-induced plasma as results from a complex process. These elements can be dispersed in several related but independent fields such as laser-matter interaction, laser ablation of material, optical and thermo-dynamic properties of hot and ionized gas, or plasma propagation in a background gas. We believe that presenting the ensemble of understanding elements of laser-induced plasma in a comprehensive way and in limited pages of this paper will be helpful for further development and optimized use of the LIBS technique. Experimental results obtained in our laboratory are used to illustrate the studied physical processes each time such illustration becomes possible and helpful.

Iron homeostasis and H63D mutations in alcoholics with and without liver disease

MM： To evaluate the prevalence of HFE gene mutation and indices of disturbed iron homeostasis in alcoholics with and without liver disease. METHODS： One hundred and fifty-three heavy drinkers （defined as alcohol consumption 〉 80 g/d for at least 5 years） were included in the study. These comprised 78 patients with liver disease [liver disease alcoholics （LDA）] in whom the presence of liver disease was confirmed by liver biopsy or clinical evidence of hepatic decompensation, and 75 subjects with no evidence of liver disease, determined by normal liver tests on two occasions [non-liver disease alcoholics （NLDA）], were consecutively enrolled. Serum markers of iron status and HFE C282Y and H63D mutations were determined. HFE genotyping was compared with data obtained in healthy blood donors from the same geographical area. RESULTS： Gender ratio was similar in both study groups. LDA patients were older than NLDA patients （52 ± 10 years vs 48 ± 11 years, P = 0.03）. One third and one fifth of the study population had serum transferrin saturation （TS） greater than 45% and 60% respectively. Serum iron levels were similar in both groups. However, LDA patients had higher TS （51 ± 27 vs 36 ± 13, P 〈 0.001） and ferritin levels （559 ± 607 ng/mL vs 159 ± 122 ng/mL, P 〈 0.001）, and lower total iron binding capacity （TIBC）（241 ± 88 μg/dL vs 279 ± 40 μg/dL, P = 0.001）. The odds ratio for having liver disease with TS greater than 45% was 2.20 （95% confidence interval （CI）： 1.37-3.54）. There was no difference in C282Y allelic frequency between the two groups. However, H63D was more frequent in LDA patients （0.25 vs 0.16, P = 0.03）. LDA patients had a greater probability of carrying at least one HFE mutation than NLDA patients （49.5% vs 31.6%, P = 0.02）. The odds ratio for LDA in patients with H63D mutation was 1.57 （95% CI： 1.02-2.40）. CONCLUSION： The present study confirms the presence of iron overload in alcoholics, which was more severe in the subset of subjects with liver disease,

Assessment of ghrelin and leptin receptor levels in postmenopausal women who received oral or transdermal menopausal hormonal therapy

Objective： In postmenopausal women, an increased leptin concentration and reduced levels of ghrelin and adiponectin were observed. The aim of this study was to evaluate the concentrations of the active form of ghrelin, total ghrelin, leptin receptor, lipoprotein（a）（Lp（a））, and plasminogen activator inhibitor type 1 （PAl-l） in postmenopausal women who received oral or transdermal menopausal hormonal therapy （MHT）. Methods： The study involved 76 healthy women： 46 women aged from 44 to 58 years who received oral （26） or transdermal （20） MHT; the control group consisted of 30 women aged from 44 to 54 years who did not receive MHT. The plasma concentrations of total ghrelin, the active form of ghrelin, Lp（a）, and PAI-I：Ag were measured by enzyme-linked immunosorbent assay （ELISA）. The concentration of the leptin receptor was measured by enzyme immunometric assay （EIA）. Results： We observed a significantly higher concentration o~ total ghrelin and the active form of ghrelin in women who received transdermal MHT in comparison with those who took oral MHT. We also found a significantly lower concentration of total ghrelin in women who received oral MHT compared with the control group. A higher concentration of PAl-1 ：Ag was found in the group of women who took transdermal MHT in comparison with those who took oral MHT and with the control group. The differences were statistically significant. Additionally, we found a significant negative correlation between the concentrations of total ghrelin and PAl-1 ：Ag and a positive correlation between the concentrations of total ghrelin and leptin receptor in women who received transdermal MHT. Conclusions： The study showed that women who used transdermal MHT had higher levels of total ghrelin than women who took oral MHT. This indicates a beneficial effect of the transdermal route of MHT. Guided by the theoretical prediction,a new MAX phase V2SnC was synthesized experimentally for the first time by reaction of V,Sn,and C mixtures at 1000°*** chemical composition and crystal structure of this new compound were identified by the cross-check combination of first-principles calculations,X-ray diffraction(XRD),energy dispersive X-ray spectroscopy(EDS),and high resolution scanning transmission electron microscopy(HR-STEM).The stacking sequence of V2C and Sn layers results in a crystal structure of space group P63/*** a-and c-lattice parameters,which were determined by the Rietveld analysis of powder XRD pattern,are 0.2981(0)nm and 1.3470(6)nm,*** atomic positions are V at 4f(1/3,2/3,0.0776(5)),Sn at 2d(2/3,1/3,1/4),and C at 2a(0,0,0).A new set of XRD data of V2SnC was also *** calculations suggest that this new compound is stable with negative formation energy and formation enthalpy,satisfied Born-Huang criteria of mechanical stability,and positive phonon branches over the Brillouin *** also has low shear deformation resistance c44(second-order elastic constant,cij)and shear modulus(G),positive Cauchy pressure,and low Pugh’s ratio(G/B=0.500<0.571),which is regarded as a quasi-ductile MAX *** mechanism underpinning the quasi-ductility is associated with the presence of a metallic bond.

Anabolic steroid-induced cardiomyopathy underlying acute liver failure in a young bodybuilder

Heart failure may lead to subclinical circulatory disturbances and remain an unrecognized cause of ischemic liver injury. We present the case of a previously healthy 40-year-old bodybuilder, referred to our Intensive-Care Unit of Hepatology for treatment of severe acute liver failure, with the suspicion of toxic hepatitis associated with anabolic steroid abuse. Despite the absence of symptoms and signs of congestive heart failure at admission, an anabolic steroid-induced dilated cardiomyopathy with a large thrombus in both ventricles was found to be the underlying cause of the liver injury. Treatment for the initially unrecognized heart failure rapidly restored liver function to normal. To our knowledge, this is the f irst reported case of severe acute liver failure due to an unrecognized anabolic steroid-induced cardiomyopathy. Awareness of this unique presentation will allow for prompt treatment of this potentially fatal cause of liver failure.
