Human Seroreactivity to Secreted Molecules of Corynebacterium pseudotuberculosis

Corynebacterium pseudotuberculosis is an infectious agent that occurs in small ruminants causing caseous lymphadenitis, and more rarely in humans causing lymphadenitis and pneumonia. The breeding small ruminants have great economic importance in Brazil. Rural farm workers and veterinary students who acquired this disease suffered from weakening symptoms for weeks, and the identification of the etiological agent was time-consuming and complex. Due to the low prevalence of case records, there is probably no available commercial diagnostic kit for C. pseudotuberculosis infection in humans. This study aimed to describe human seroreactivity to secreted antigens from C. pseudotuberculosis. Reactivity of serum from farm workers (n = 14), individuals who work with the bacillus at laboratory (n = 8) or individuals without contact (n = 25) was tested with secreted proteins from PAT10 strain of C. pseudotuberculosis by Western blotting. Samples of all (100%) farm workers showed reactivity to 31 kDa, 71 kDa and 164 kDa proteins, while laboratory workers showed 87.5%, 62.5 % and 37.5%, and no-contact 20%, 0% and 16%, respectively. All sera recognized the 275 kDa protein. Our data suggest that C. pseudotuberculosis secreted proteins are antigenic in humans and the recognition profiles allowed the identification of individuals with and without prior contact with this bacillus. This is the first paper which describes human reactivity to C. pseudotuberculosis in serum samples of workers in Brazil.

Atlantic Forest Hotspot: Why We Should Consider Mature Forests a Conservation Priority in the São Paulo Metropolitan Region, Southeastern Brazil

The Atlantic Forest is among the 35 hotspots on the planet, and yet few floristic or structural studies have been conducted on mature forest fragments of that biome in Brazil. In view of this knowledge gap and of the need to further comprehend the importance of conserving mature forests, we surveyed the arboreal component of three little-altered forest remnants from the S?o Paulo metropolitan region (SPMR), southeastern Brazil, and compared our data with the one from other phytosociological studies performed in the region. Mature forests showed a different floristic composition from one of young forests. Overall, the former has a higher number and percentage of threatened and endemic species than the latter, and also a higher exclusivity of occurrence of threatened species. In areas up to twice as smaller, mature forests have 1.5 to 4.3 times more endemic species to the Atlantic Forest and up to 9 times more species threatened with extinction than young forests. These facts, along with the scarcity of remnants of mature forests, led us to consider such forests as hotspots within the Atlantic Forest hotspot in the SPMR, as well as to categorize them as of high relevance for conservation.

ProTSA: A Testing Process for Automotive Software Domain

This study evaluates the development of a testing process for the automotive software domain, highlighting challenges stemming from the absence of adequate processes. The research demonstrates the application of Design Science Research methodology in developing, an automotive software testing process—ProTSA, using six functional testing modules. Additionally, the study evaluates the benefits of implementing ProTSA in a specific Original Equipment Manufacturer (OEM) using an experimental single-case approach with industry professionals’ participation through a survey. The study concludes that combining testing techniques with effective communication and alignment is crucial for enhancing software quality. Furthermore, survey data indicates that implementing ProTSA leads to productivity gains by initiating tests early, resulting in time savings in the testing program and increased productivity for the testing team. Future work will explore implementing ProTSA in cybersecurity, over-the-air software updates, and autonomous vehicle testing processes. .

Numerical Analysis of Physical and Geometrical Imperfections in Cellular Beams

Cellular beams are appropriate for large spans with relatively small loads or for conditions in which strain dictates dimensioning. Another important advantage of cellular beams is the possibility of passing utility ducts through the openings, which avoids cutting through the web of the beam or increasing the construction height, which occurs when the ducts pass under the beams. Geometrical imperfections result from a lack of straightness during fabrication of rolled profiles. Geometric imperfections are represented numerically by an initial curvature. Additionally, the physical imperfections of rolled profiles result from the appearance of residual stresses. The condition that creates residual stresses in steel is the result of thermal and mechanical industrial processes. In this study, numerical analyses are performed with cellular beams using Finite Element Method software. During the simulations, through non-linear geometric and physical analyses, geometric imperfections were varied, where lateral torsional buckling in cellular beams was considered as a function of the unrestrained length. In the boundary, conditions were restrained displacements in the axis X, Y, Z and rotation about X-axis, thus simulating the fork support. The beams are submitted to uniform bending and concentrated load. The results from the numerical analyses were compared with the calculation procedures, which verified that the results were significant.

High-throughput phenotyping of two plant-size traits of Eucalyptus species using neural networks

In forest modeling to estimate the volume of wood,artificial intelligence has been shown to be quite effi-cient,especially using artificial neural networks(ANNs).Here we tested whether diameter at breast height(DBH)and the total plant height(Ht)of eucalyptus can be pre-dicted at the stand level using spectral bands measured by an unmanned aerial vehicle(UAV)multispectral sensor and vegetation *** do so,using the data obtained by the UAV as input variables,we tested different configurations(number of hidden layers and number of neurons in each layer)of ANNs for predicting DBH and Ht at stand level for different Eucalyptus *** experimental design was randomized blocks with four replicates,with 20 trees in each experimental *** treatments comprised five Eucalyptus species(***,***,***,***-dis,and ***)and Corymbria *** and Ht for each plot at the stand level were measured seven times in separate overflights by the UAV,so that the multispectral sensor could obtain spectral bands to calculate vegetation indices(VIs).ANNs were then constructed using spectral bands and VIs as input layers,in addition to the categorical variable (species), to predict DBH and Ht at the stand level simultaneously. This report represents one of the first appli-cations of high-throughput phenotyping for plant size traits in Eucalyptus species. In general, ANNs containing three hidden layers gave better statistical performance (higher esti-mated r, lower estimated root mean squared error-RMSE) due to their greater capacity for self-learning. Among these ANNs, the best contained eight neurons in the first layer, seven in the second, and five in the third (8 − 7 − 5). The results reported here reveal the potential of using the gener-ated models to perform accurate forest inventories based on spectral bands and VIs obtained with a UAV multispectral sensor and ANNs, reducing labor and time.

Perceived Efficacy of Vaginal Dryness Relief: A Comparative Clinical Study between Sodium hyaluronate vaginal gel1vs. Promestriene cream2

In the study, we report the comparative efficacy of the sodium hyaluronate gel vaginal application and promestriene cream which were tested to treat the gynecological symptoms of people who suffered from vaginal dryness. Method: 35 patients applied sodium hyaluronate vaginal gel one time a day for a period of two times a week during three weeks and other 33 patients using promestriene cream applied it every day for a period of three weeks also. Results: No significant difference was observed between vaginal application of sodium hyaluronate gel and promestriene cream, regarding both, as regarding the dryness of the intimate mucosa (p = 0.786), the attribute of moisturizing properties to the intimate mucosa (p = 0.142), the comfort sensation (p = 0.528), and no significant difference was observed regarding the fragrance of the product (p = 0.088). Conclusion: The similar results between vaginal application of both products support the use of sodium hyaluronate vaginal gel (Lubrinat®) in the initial approach of symptoms of vaginal dryness.

Oceanic Indices for Forecasting Seasonal Rainfall over the Northern Part of Brazilian Northeast

A relationship between oceanic conditions in the northwestern equatorial Atlantic (NWEA) and the seasonal rainfall over the northern part of Brazilian Northeast (NNEB) allows large climate events to be forecasted with a delay of a few months. Observed sea surface variables (sea surface temperature, wind stress and latent heat flux) and reanalyzed temperature and salinity profiles at depths of 0 - 150 m are used during 1974-2008. Perturbations in the Wind-Evaporation-SST mechanism over the NWEA during the last months of the year and the first months of the following year are of primary importance in evaluating the risk that strong climate events will affect the subsequent seasonal rainfall (in March-April) over the NNEB. Especially interesting are the Barrier Layer Thickness (BLT) and Ocean Heat Content (OHC) in the NWEA region from August-September through the subsequent months, during which a slow and steady evolution is apparent, with the highest signal occurring in October-November. Through their relationship with the local surface dynamic conditions, such BLT and OHC perturbations during the last months of the year can be used as a valuable indicator for forecasting wet or dry events over the NNEB during the subsequent rainfall season. A proposal is discussed to deploy additional temperature/conductivity sensors down to a depth of 140 m at three PIRATA moorings located in the NWEA region. That will be necessary if the BLT and other parameters of energy exchange between the ocean and atmosphere are to be estimated in real time and with a sufficiently high vertical resolution.
