Modern drugs have changed epilepsy,which affects people of all ***,for young people with epilepsy,the framework of drug development has *** the wake of the thalidomide catastrophe,the misconception emerged that for people<18 years of age drugs,including antiseizure medications(ASMs),need separate proof of efficacy and safety,overall called"pediatric drug development".For ASMs,this has changed to some *** now accept that ASMs are effective in<18 years as well,but they still require"extrapolation of efficacy,"as if minors were another *** a result,some of the pediatric clinical epilepsy research over the past decades was *** more importantly,this has hampered research on meaningful research *** do not need to confirm that ASMs work before as they do after the 18th ***,we need to learn how to prevent brain damage in young patients by preventing seizures and optimize ASMs’*** we discuss how to proceed in this endeavor.

Prediction of the response of cancer patients to different treatments and identification of biomarkers of drug response are two major goals of individualized ***,we developed a deep learning framework called TINDL,completely trained on preclinical cancer cell lines(CCLs),to predict the response of cancer patients to different *** utilizes a tissue-informed normalization to account for the tissue type and cancer type of the tumors and to reduce the statistical discrepancies between CCLs and patient ***,by making the deep learning black box interpretable,this model identifies a small set of genes whose expression levels are predictive of drug response in the trained model,enabling identification of biomarkers of drug *** data from two large databases of CCLs and cancer tumors,we showed that this model can distinguish between sensitive and resistant tumors for 10(out of 14)drugs,outperforming various other machine learning *** addition,our small interfering RNA(siRNA)knockdown experiments on 10 genes identified by this model for one of the drugs(tamoxifen)confirmed that tamoxifen sensitivity is substantially influenced by all of these genes in MCF7 cells,and seven of these genes in T47D ***,genes implicated for multiple drugs pointed to shared mechanism of action among drugs and suggested several important signaling *** summary,this study provides a powerful deep learning framework for prediction of drug response and identification of biomarkers of drug response in *** code can be accessed at https://***/ddhostallero/tindl.

Disaster Risk Reduction and Management: Recalling the Need for Paradigm Shift in Definition

The paper articulates the need for a paradigm shift in defining the basis of what constitutes a disaster. This new framework must be sensitive to the need for a more theoretical approach to inform disaster and hazard management. Central to this is that the resulting approach cannot be muddled in linear rational and procedural doctrines but appreciate the dynamics of complex nonlinearity of disaster events. By engaging in thought experiment and critical analysis of existing literature, the paper deconstructs the normative paradigm of defining disaster. The end is to inform disaster management and risk reduction intervention and mitigation programmes. The presented alternative approach is sensitive of the need to equally include;spatial, political, social, parameters. These are appreciated as being equally important as those dealing with ecological and economic. The resulting not only include other elements, but also expands the definition to the complex conditions inherent in the origin of phenomena to the interaction of the phenomena with multiple and complex socio-spatial and demographic dynamics, and then appreciating the complex results of this nonlinear interaction. The need for a more substantive definition of disaster underlines the pre-analysis that is necessary for implementation of mitigation and prevention strategies. That disasters are becoming more complex is synonymous with the complexities inherent in post-war development dynamics.

Feature identification in complex fluid flows by convolutional neural networks

Recent advancements have established machine learning's utility in predicting nonlinear fluid dynamics,with predictive accuracy being a central motivation for employing neural ***,the pattern recognition central to the networks function is equally valuable for enhancing our dynamical insight into the complex fluid *** this paper,a single-layer convolutional neural network(CNN)was trained to recognize three qualitatively different subsonic buffet flows(periodic,quasi-periodic and chaotic)over a high-incidence airfoil,and a near-perfect accuracy was obtained with only a small training *** convolutional kernels and corresponding feature maps,developed by the model with no temporal information provided,identified large-scale coherent structures in agreement with those known to be associated with buffet *** to hyperparameters including network architecture and convolutional kernel size was also *** coherent structures identified by these models enhance our dynamical understanding of subsonic buffet over high-incidence airfoils over a wide range of Reynolds numbers.

Stress distribution and fatigue life of nonlinear vibration of an axially moving beam

In this paper, the effects of the nonlinear vibration on stress distribution and fatigue life of the axially moving beam are *** parametric excitation of the flexible material is created by the pulsating moving speed. Three-to-one internal resonance condition is satisfied. The three-parameter model is adopted in the viscoelastic constitutive relation. The nonlinear vibration of the axially moving beam with parametric and internal resonance are studied by using the direct multiple scales method(MSM)with numerical simulation confirmation. Based on the approximate analytical solution, the distribution of tensile stress and bending stress on the axially moving beam is presented by adopting a V-belt as the prototype. Based on the maximum stable cyclic stress, the limit cycle response of the V-belt is utilized to evaluate the effect of the resonance on the fatigue life. Also, the influences of the internal resonance on the steady-state responses and the fatigue life of the V-belt are revealed. Numerical examples illustrate that large unwanted resonances occur and the second-order mode receives vibration energy from to the firstorder mode. The numerical results demonstrate that the nonlinear vibration significantly reduces the fatigue life of the *** fatigue life analysis method in this paper can be applied to the excited vibration of other axially moving systems and even static continuum.
