Adoptive cell therapy(ACT)is an immunotherapy strategy for cancer that has seen widespread clinical *** ACT,patient-derived lymphocytes are stimulated with the antigen of interest ex vivo,proliferated,then returned to the patient to initiate an antigen-specific antitumor *** effective,this process is resource-intensive and logistically impossible for many *** artificial antigen presenting cells(aAPCs)offer a potential“off-the-shelf”alternative to ex vivo *** particulate aAPCs perform well in vitro,they have had limited success in vivo due to poor bioavailability after *** to bioavailability include rapid clearance,unfavorable biodistribution,and inadequate interactions with CD8+T cells at sites of *** properties such as elasticity have been shown to vastly impact the bioavailability and particle-cell interactions,but this has yet to be investigated in the context of aAPCs for in vivo T-cell *** literature likewise indicates that biomaterial properties,especially elasticity,can modulate T-cell activation in *** the goal of creating a more biomimetic,next-generation particulate aAPC,we developed a poly(ethylene)glycol hydrogel particle platform with tunable elasticity to investigate the impact of elasticity on antigen-specific T cell activation for in vivo adoptive *** this knowledge,we were able to gain more precise control over in vivo T cell activation and investigate possible mechanisms including the effects of aAPC elasticity on T cell binding,macrophage uptake,and the protein corona.

Re nanoclusters anchored on nanosheet supports: Formation of Re-O-matrix bonding and evaluation as all-pH-range hydrogen evolution reaction (HeR) electrocatalysts

Although the water splitting-based generation of hydrogen as an energy carrier can help to mitigate the global problems of energy shortage and climate change,the practical implementation of this strategy is hindered by the absence of inexpensive high-performance electrocatalysts for the hydrogen evolution reaction (HeR).Re-based HeR electrocatalysts exhibit predictable high performance within the entire pH range but suffer from arduous formation (i.e.,vulnerability to oxidation) and uncontrollable aggregation,which strongly discourages the maximisation of active site exposure required for activity *** overcome these limitations,we herein hydrothermally synthesise Re nanoclusters uniformly distributed on nanosheet supports,such as reduced graphene oxide nanosheets (Re NCs@rGO),revealing that this hybrid features abundant exposed active sites and high oxidation *** obtained electrocatalysts were elaborately characterized by microscopic and spectroscopic ***,density functional theory calculations confirm the optimised synthesis of Re NCs@rGO and indicate the crucial role of Re–O–C junction formation in securing *** effective suppression of Re nanocluster detachment/dissolution under HeR conditions endows Re NCs@rGO with high electron conductivity and electrochemical stability,resulting in a durability superior to that of commercial Pt/C and an activity similar to that of this *** a result,Re NCs@rGO exhibited remarkably small HeR overpotentials of 110,130,and 93 m V to deliver a current density of 10 mA cm^(-2) in 0.5 M H_(2)SO_(4),1 M PBS,and 1 M KOH,***,Re NCs@rGO is a promising alternative to conventional Pt-group-metal catalysts and should find applications in next-generation high-performance water splitting systems.

Reduction of Pt Usage in Fuel Cell electrocatalysts Using Carbon Nanotubes and Non-Pt Metals

The high-priced and limited Pt constitutes a high barrier to commercialization of fuel cells. Pt is essential for the electrode catalyst of polymer electrolyte fuel cells (PeFCs). A reduction in Pt usage is one of the key requirements for the commercialization of fuel cells for use in everyday life, because of its high price and limited availability, and the difficulty of finding suitable substitutes. Non-Pt fuel cell catalysts will decrease the demand for Pt by PeFCs, enabling more Pt to be available for use in other essential products, and make fuel cells more popular[1]. The cheaper Mo2C is known to possess similar catalytic activities and electronic structures to Pt[2]. Carbon black (CB) is widely used as the support for Pt nanoparticles. However, we found that when carbon nanotubes (CNTs) rather than CB are used as the support, the performance is improved, especially below 600 mA/cm2[3,4]. Here, we show that a combination of Mo2C catalyst and carbon nanotubes in the anode provides performance as high as half that of the current PeFCs with Pt catalysts below 600mA/cm2.

Single-center study comparing computed tomography colonography with conventional colonoscopy

AIM:To compare the results from computed tomography (CT) colonography with conventional colonoscopy in symptomatic patients referred for colonoscopy. MeTHODS: The study included 227 adult outpatients, mean age 60 years, with appropriate indications for colonoscopy. CT colonography and colonoscopy were performed on the same day in a metropolitan teaching hospital. Colonoscopists were initially blinded to the results of CT colonography but there was segmental unblinding during the procedure. The primary outcome measures were the sensitivity and specificity of CT colonography for the identification of polyps seen at colonoscopy (i.e. analysis by polyp). Secondary outcome measures included an analysis by patient, extracolonic findings at CT colonography, adverse events with both procedures and patient acceptance and preference. ReSULTS: Twenty-five patients (11%) were excluded from the analysis because of incomplete colonoscopy or poor bowel preparation that affected either CT colonography, colonoscopy or both procedures. Polyps and masses (usually cancers) were detected at colonoscopy and CT colonography in 35% and 42% of patients, respectively. Of nine patients with a finaldiagnosis of cancer, eight (89%) were identified by CT colonography as masses (5) or polyps (3). For polyps analyzed according to polyp, the overall sensitivity of CT colonography was 50% (95% CI, 39%-61%) but this increased to 71% (95% CI, 52%-85%) for polyps ≥ 6 mm in size. Similarly, specificity for all polyps was 48% (95% CI, 39%-58%) increasing to 67% (95% CI, 56%-76%) for polyps ≥ 6 mm. Adverse events were uncommon but included one colonic perforation at colonoscopy. Patient acceptance was high for both procedures but preference favoured CT colonography. CONCLUSION: Although CT colonography was more sensitive in this study than in some previous studies, the procedure is not yet sensitive enough for widespread application in symptomatic patients.

Role of microbial dysbiosis in the pathogenesis of esophageal mucosal disease:A paradigm shift from acid to bacteria?

Genomic sequencing,bioinformatics,and initial speciation(e.g.,relative abundance)of the commensal microbiome have revolutionized the way we think about the“human”body in health and *** interactions between the gut bacteria and the immune system of the host play a key role in the pathogenesis of gastrointestinal diseases,including those impacting the *** relatively stable,there are a number of factors that may disrupt the delicate balance between the luminal esophageal microbiome(eM)and the *** changes are thought to be a product of age,diet,antibiotic and other medication use,oral hygiene,smoking,and/or expression of antibiotic products(bacteriocins)by other *** effects may lead to persistent dysbiosis which in turn increases the risk of local inflammation,systemic inflammation,and ultimately disease *** has suggested that the etiology of gastroesophageal reflux disease-related esophagitis includes a cytokine-mediated inflammatory component and is,therefore,not merely the result of esophageal mucosal exposure to corrosives(i.e.,acid).emerging evidence also suggests that the eM plays a major role in the pathogenesis of disease by inciting an immunogenic response which ultimately propagates the inflammatory ***,we discuss the potential role for manipulating the eM as a therapeutic option for treating the root cause of various esophageal disease rather than just providing symptomatic relief(i.e.,acid suppression).

Optimization of Casting Process for Heat and Abrasion Resistant Large Gray Iron Castings

Numerical simulations were used to optimize the casting design and conditions for large cast iron castings for marine engines, Simulations of the mold filling and solidification sequences were used to analyze the problems of previous casting conditions with marked improvements for large cylinder liner parts, The amount and positions of chills were optimized to improve the mechanical properties and to minimize the shrinkage and micro porosity in the castings. Ultra sonic testing, penetration testing, and mechanical property testing show no defects in the castings with the productivity significantly increased.

Dear editor,Flowering is a major developmental switch in the life cycle of flowering plants. FRIGIDA （FRI） activates FLOWeRING LOCUS C （FLC） transcription, a major repressor of this switch regulating flow- ering time （johanson et al., 2000）. FRI is highly conserved and many natural variations in FRI are found to affect flowering time. However, little is known about the molecular characteristics of FRI.

Optimization of casting conditions for heat and abrasion resistant large grey iron castings

Numerical simulation technology was applied for optimizing the casting design and conditions in large cast iron castings for marine engine. By the simulation of mold filling and solidification sequences the problems of the previous casting conditions were analyzed and marked improvements for large cylinder liner parts were derived from these results. especially the amount and positions of chills were optimized to increase the mechanical properties and to minimize the shrinkage and microporosity in the castings. Ultrasonic testing, penetration testing and mechanical property testing were carried out for the parts with the modified casting conditions. It showed that no defects in the castings were found and the productivity could be distinctly increased. The mechanical properties satisfied also the specification demanded.
