NeCAB family of neuronal calcium-binding proteins in health and disease

The N-terminal eF-hand calcium-binding proteins 1–3(NeCAB1–3) constitute a family of predominantly neuronal proteins characterized by the presence of at least one eF-hand calcium-binding domain and a functionally less well characterized C-terminal antibiotic biosynthesis monooxygenase domain. All three family members were initially discovered due to their interactions with other proteins. NeCAB1 associates with synaptotagmin-1, a critical neuronal protein involved in membrane trafficking and synaptic vesicle exocytosis. NeCAB2 interacts with predominantly striatal G-protein-coupled receptors, while NeCAB3 partners with amyloid-β A4 precursor protein-binding family A members 2 and 3, key regulators of amyloid-β production. This demonstrates the capacity of the family for interactions with various classes of proteins. NeCAB proteins exhibit distinct subcellular localizations: NeCAB1 is found in the nucleus and cytosol, NeCAB2 resides in endosomes and the plasma membrane, and NeCAB3 is present in the endoplasmic reticulum and Golgi apparatus. The antibiotic biosynthesis monooxygenase domain, an evolutionarily ancient component, is akin to atypical heme oxygenases in prokaryotes but is not wellcharacterized in vertebrates. Prokaryotic antibiotic biosynthesis monooxygenase domains typically form dimers, suggesting that calcium-mediated conformational changes in NeCAB proteins may induce antibiotic biosynthesis monooxygenase domain dimerization, potentially activating some enzymatic properties. However, the substrate for this enzymatic activity remains uncertain. Alternatively, calcium-mediated conformational changes might influence protein interactions or the subcellular localization of NeCAB proteins by controlling the availability of protein–protein interaction domains situated between the eF hands and the antibiotic biosynthesis monooxygenase domain. The 3d Inverse Problem of the Wave equation for a General multi-connected Vibrating membrane with a Finite Number of Piecewise Smooth Boundary Conditions

The trace of the wave kernel μ（t） =∑ω=1^∞ exp（-iteω^1/2）, where {eω}ω^∞=1 are the eigenvalues of the negative Laplacian -△↓2 = -∑k^3=1 （δ/δxk）^2 in the （x^1, x^2, x^3）-space, is studied for a variety of bounded domains, where -∞ 〈 t 〈 ∞ and i= √-1. The dependence of μ （t） on the connectivity of bounded domains and the dirichlet, Neumann and Robin boundary conditions are analyzed. Particular attention is given for a multi-connected vibrating membrane Ω in Ra surrounded by simply connected bounded domains Ω j with smooth bounding surfaces S j （j = 1,……, n）, where a finite number of piecewise smooth dirichlet, Neumann and Robin boundary conditions on the piecewise smooth components Si^＊（i = 1 ＋ kj-1,……, kj） of the bounding surfaces S j are considered, such that S j = Ui-1＋kj-1^kj Si^＊, where k0=0. The basic problem is to extract information on the geometry Ω by using the wave equation approach from a complete knowledge of its eigenvalues. Some geometrical quantities of Ω （e.g. the volume, the surface area, the mean curvuture and the Gaussian curvature） are determined from the asymptotic expansion ofexpansion of μ（t） for small │t│.

Influence of the Choice of NN Potential model on γd→τ~0d Observables near η-Threshold

Influence of the choice of the NN potential model governing the deuteron wave function on the observables for coherent π0-photoproduction on the deuteron near η-threshom is investigated by using a three-body model for the intermediate ηN N system with separable two-body interactions. Results for unpolarized differential cross section and polarization observables are predicted. It is revealed that the choice of the NN potential model has a visible effect on the differential cross section and most of the polarization observables, especially in the photon energy range of 600-800 meV and extreme backward pion angles. We find that the deviation among results obtained by using different deuteron wave functions is quite large. The use of the Cd-Bonn NN potential for deuteron wave function doubles the differential cross section in this kinematic region. Compared with the experimental data from CLAS collaboration for differential cross section, sizeable discrepancies are found.

Bound states in the continuum （BICs） are perfectly localized resonances despite embedding in the continuum ***,an isolated BIC is very sensitive to the structure ***,we report merging acoustic BICs in a single open resonator,robust against the structure *** find that both symmetry-protected BIC and Friedrich-Wintgen BIC are sustained in a single coupled waveguide-resonator *** varying the height and length of the resonator,these two BICs move toward each other and merge into a single one at a critical *** to an individual BIC,the merged BIC is robust against fabrication error because its Q-factor is proportional to ΔL-4,whereΔL embodies the structure *** essence of this extraordinary phenomenon is perfectly explained by the two-and three-level approximations of the effective nonHermitian ***,we present direct experimental demonstrations of the moving and merging of BICs in a coupled 3d waveguide-resonator,which are evidenced by the vanishing of the linewidth of Fano resonance in the transmission *** results may find exciting applications in designing high-quality acoustic sources,sensors and filters.

This paper shows how the so called von Karman model can be obtained as a singular limit of a modified mindlin-Timoshenko system when the modulus of elasticity in shear κ tends to infinity, provided a regularizing term through a fourth order dispersive operator is added. Introducing damping mechanisms, the authors also show that the energy of solutions for this modified mindlin-Timoshenko system decays exponentially, uniformly with respect to the parameter k. As κ→∞, the authors obtain the damped von Karman model with associated energy exponentially decaying to zero as well.

Therapeutic potential of exercise-hormone irisin in Alzheimer's disease

Irisin is a myokine that is generated by cleavage of the membrane protein fibronectin type Ⅲ domain-containing protein 5(FNdC5) in response to physical exercise. Studies reveal that irisin/FNdC5 has neuroprotective functions against Alzheimer's disease, the most common form of dementia in the elderly, by improving cognitive function and reducing amyloid-β and tau pathologies as well as neuroinflammation in cell culture or animal models of Alzheimer's disease. Although current and ongoing studies on irisin/FNdC5 show promising results, further mechanistic studies are required to clarify its potential as a meaningful therapeutic target for alleviating Alzheimer's disease. We recently found that irisin treatment reduces amyloid-β pathology by increasing the activity/levels of amyloid-β-degrading enzyme neprilysin secreted from astrocytes. Herein, we present an overview of irisin/FNdC5's protective roles and mechanisms against Alzheimer's disease.

Although many causes of Alzheimer’s disease(Ad)may exist,both the original amyloid cascade and tau hypotheses posit that abnormal misfolding and accumulation of amyloid-β(Aβ)and tau protein is the central event causing the ***,that conclusion could be only partly true,and there is conflicting evidence about the role of both proteins in Ad,being able to precede and influence one *** researchers argue that these proteins are mere executors rather than primary causes of ***,there have been continuing refinements of both hypotheses,with alternative explanations proposed.Aβand tau proteins may be independently involved in specific neurotoxic pathways;yet there may be other crucial processes going on in early ***,accumulating evidence suggests that Aβand tau act synergistically,rather than additively in disease onset(Jeremic et al.,2021,2023a).

effect of synthesis conditions and surrounding medium on luminescence properties of YVO_4:eu^(3+) nanopowders

Nanocrystalline yttrium vanadate doped with europium ions powders were synthesized via sol-gel method based on decomposition of metal-polymer complex. X-ray diffraction analysis showed that samples had pure tetragonal phase without any impurities. Scanning electron microscopy and static light scattering technique were used to study morphology and size of prepared nanoparticles. Average diameter of the nanoparticles was about 40 nm. The changes in structural and luminescence properties were observed as a function of the first and second calcination temperature. The optimal conditions for synthesis of nanoparticles were determined as Т1=500℃, t1=1 h; Т2=950 ℃, t2=1.5 h. The effect of different media surrounding the nanoparticles on their luminescence properties and lifetime was investigated and discussed in terms of effective refractive index. It was found that the observed lifetime of YVO4：eu^3＋ 5 at.% nanophosphor was decreased from 0.64 ms in air（nmed=1） to 0.45 ms in chalcogenide glass As39S61（nmed=2.39）.
