AIM： To analyze.the.ne.tralizing activity of antibodie. against e. re.ion of he.atitis C virus （HCV）. Spe.ific polydonal antibody was raise. via immunization of Ne. Ze.land rabbits with a synthe.ic pe.tide.that had be.n de.ive. from the.e. re.ion of HCV and was shown to be.highly conse.ve. among HCV publishe. ge.otype.. Me.HODS： Hype.-immune.HCV e. antibodie. we.e.incubate. ove. night at 4 ℃ with se.um sample. positive.for HCV RNA, with viral loads ranging from 615 to 3.2 million IU/mL. Tre.te. se.a we.e.incubate. with He.G2 ce.ls for 90 min. Blocking of viral binding and e.try into ce.ls by anti e. antibody we.e.te.te. by me.ns of RTPCR and flow cytome.ry. Re.ULTS： Dire.t immunostaining using FITC conjugate. e. antibody followe. by Flow cytome.ric analysis showe. re.uce. me.n fluore.ce.ce.inte.sity in sample. pre.incubate. with e. antibody compare. with untre.te. sample.. Furthe.more. 13 out of 18 positive.se.a （72%） showe. comple.e.inhibition of infe.tivity as de.e.te. by RT-PCR. CONCLUSION： In house.produce. e. antibody, blocks binding and e.try of HCV virion infe.tion to targe. ce.ls sugge.ting the.involve.e.t of this e.itope.in virus binding and e.try. Isolation of the.e.antibodie. that block virus attachme.t to human ce.ls are.use.ul as the.ape.tic re.ge.ts.

AIM： TO e.tablish a ce.l culture.syste. with longte.m re.lication of he.atitis C virus （HCV） ge.ome.and e.pre.sion of viral antige.s in vitro. Me.HODS： He.G2 ce.l line.was te.te. for its susce.tibility to HCV by incubation with a se.um from a patie.t with chronic he.atitis C. Ce.ls and supe.natant we.e.harve.te. at various time.points during the.culture. Culture.supe.natant was te.te. for its ability to infe.t na＇ive.ce.ls. The.pre.e.ce.of minus （antise.se. RNA strand, and the.de.e.tion of core.and e. antige.s in ce.ls we.e.e.amine. by RT-PCR and immunological te.hnique. （flow cytome.ry and We.te.n blot） re.pe.tive.y. Re.ULTS： The.intrace.lular HCV RNA was first de.e.te. on d 3 afte. infe.tion and the. could be.consiste.tly de.e.te. in both ce.ls and supe.natant ove. a pe.iod of at le.st thre. months. The.fre.h ce.ls could be.infe.te. with supe.natant from culture. infe.te. ce.ls. Flow cytome.ric analysis showe. surface.and intrace.lular HCV antige. e.pre.sion using in house.made.polyclonal antibodie. （anti-core. and anti-e.）. We.te.n blot analysis showe. the.e.pre.sion of a cluste. of immunoge.ic pe.tide. at mole.ular we.ghts e.te.de. be.we.n 31 and 45 kDa in an one.month old culture.of infe.te. ce.ls whe.e.s this cluste. was unde.e.table.in uninfe.te. He.G2 ce.ls. CONCLUSION： He.G2 ce.l line.is not only susce.tible.to HCV infe.tion but also supports its re.lication in vitro. e.pre.sion of HCV structural prote.ns can be.de.e.te. in infe.te. He.G2 ce.ls. The.e.ce.ls are.also capable.of she.ding viral particle. into culture.me.ia which in turn be.ome.infe.tious to uninfe.te. ce.ls.
