A functional variant in the CD209 promoter is associated with DQ2-negative celiac disease in the Spanish population

AIM： To address the role of CD209 in celiac disease （CD） patients. Non-human leukocyte antigen （HLA） genetic factors in CD predisposition are poorly understood, and environmental factors like infectious pathogens may play a role. CD209 is a dendritic and macrophage surface molecule involved in pathogen recognition and immune activation. Recently, a functional variant in the promoter of the CD209 gene （-336A/G） has been shown to affect the transcriptional CD209 activity in vitro and it has been associated with a higher susceptibility to/or severity of infection. MeTHODS： The study population was composed of two case-control cohorts of 103 and 386 CD patients and 312 y 419 healthy controls as well as a panel of 257 celiac families. Genotyping for the -336A/G CD209 promoter polymorphism was performed using a TaqMan 5＇ allelic discrimination assay. HLA-DQ was determined by hybridization with allele specific probes. ReSULTS： Initially, the case-control and familial studies did not find any association of the -336 A/G CD209 genetic variant with CD susceptibility. However, the stratification by HLA-DQ2 did reveal a significant association of CD209 promoter polymorphism in the HLA-DQ2 （-） group （carrier A vs GG in DQ2 （-） vs DQ2 （＋） patients （P = 0.026, OR = 3.71）. CONCLUSION： The -336G CD209 allele seems to be involved in CD susceptibility in HLA-DQ2 （-） patients. Our results might suggest a possible role of pathogens in the onset of a minor group of CD patients.

Located at the National University in Bogotá,Colombia,this paper presents the development of the integration of kinematics,simplified kinetic and zero-dimensional models of indicator diagram of a double-acting,single-cylinder steam engine;the model integration is done for two configurations of the distribution system to simulate the instantaneous torque and the *** simulations are carried out at different steam entry conditions and an advance angle of 0°for the distribution system,fed with the data obtained from the characterization and metrological survey of the Ve(vapor engine)parts in a CAD(computer-aided design)*** is verified that integrating the models constitutes a simulation tool that allows for the instantaneous torque deliveries and average torque of the accurate operation’s Ve.

Olfactory ensheathing glia promote axonal regeneration in the mammalian central nervous system,including retinal ganglion cell axonal growth through the injured optic ***,it is unknown whether olfactory ensheathing glia also have neuroprotective *** ensheathing glia express brain-derived neurotrophic factor,one of the best neuroprotectants for axotomized retinal ganglion ***,we aimed to investigate the neuroprotective capacity of olfactory ensheating glia after optic nerve *** ensheathing glia cells from an established rat immortalized clonal cell line,TeG3,were intravitreally injected in intact and axotomized retinas in syngeneic and allogeneic mode with or without microglial inhibition or immunosuppressive *** and gene expression analyses were *** bulb-derived primary olfactory ensheathing glia and TeG3 express major histocompatibility complex classⅡ*** and syngenically transplanted TeG3 cells survived in the vitreous for up to 21 days,forming an *** axotomized retinas,only the allogeneic TeG3 transplant rescued retinal ganglion cells at 7 days but not at 21 *** these retinas,microglial anatomical activation was higher than after optic nerve crush *** intact retinas,both transplants activated microglial cells and caused retinal ganglion cell death at 21 days,a loss that was higher after allotransplantation,triggered by pyroptosis and partially rescued by microglial inhibition or ***,neuroprotection of axotomized retinal ganglion cells did not improve with these *** different neuroprotective properties,different toxic effects,and different responses to microglial inhibitory treatments of olfactory ensheathing glia in the retina depending on the type of transplant highlight the importance of thorough preclinical studies to explore these variables.

Acknowledgment of Libraries in the Journal Literature: An exploratory Study

Purpose: This study examines acknowledgments to libraries in the journal literature, as well as the efficacy of using Web of Science(WoS) to locate general acknowledgment ***/methodology/approach: This mixed-methods approach quantifies and characterizes acknowledgments to libraries in the journal literature. Using WoS's Funding Text field, the acknowledgments for six peer universities were identified and then characterized. The efficacy of using WoS to locate library acknowledgments was assessed by comparing the WoS Funding Text search results to the actual acknowledgment text found in the ***: Acknowledgments to libraries were found in articles at all six peer universities, though the absolute and relative numbers were quite low(e library acknowledgments were for resources(collections, funding, etc.), and many were concentrated in natural history(e.g. zoology). examination of Texas A&M University zoology articles found that 91.7% of the funding information came from "acknowledgments" and not specifically a funding acknowledgment section. The WoS Funding Text search found 56% of the library acknowledgments compared to a search of the actual acknowledgment text in the *** limitations: Limiting publications to journals, using a single truncated search term, and including only six research universities in the United *** implications: This study examined library acknowledgments, but the same approach could be applied to searches of other keywords, institutions/organizations, individuals, etc. While not specifically designed to search general acknowledgments, WoS's Funding Text field can be used as an exploratory tool to search acknowledgments beyond funding. H pylori receptor MHC classⅡcontributes to the dynamic gastric epithelial apoptotic response

AIM： To investigate the role of MHC class Ⅱ in the modulation of gastric epithelial cell apoptosis induced by H pylori infection. MeTHODS： After stimulating a human gastric epithelial cell line with bacteria or agonist antibodies specific for MHC class Ⅱ and CD95, the quantitation of apoptotic and anti-apoptotic events, including caspase activation, BCL-2 activation, and FADD recruitment, was performed with a fluorometric assay, a cytometric bead array, and confocal microscopy, respectively. ReSULTS： Pretreatment of N87 cells with the anti-MHC class ⅡIgM antibody RFD1 resulted in a reduction in global caspase activation at 24 h of H pylori infection. When caspase 3 activation was specifically measured, crosslinking of MHC class Ⅱ resulted in markedly reduced caspase activation, while simple ligation of MHC class Ⅱ did not. Crosslinking of MHC class Ⅱ also resulted in an increased activation of the anti-apoptosis molecule BCL-2 compared to simple ligation. Confocal microscope analysis demonstrated that the pretreatment of gastric epithelial cells with a crosslinking anti-MHC class Ⅱ IgM blocked the recruitment of FADD to the cell surface. CONCLUSION： The ability of MHC class Ⅱ to modulate gastric epithelial apoptosis is at least partially dependent on its crosslinking. The crosslinking of this molecule has anti-apoptotic effects during the earlier time points of Hpylori infection. This effect is possibly mediated by the ability of MHC class Ⅱ to modulate the activation of the pro-apoptotic receptor Fas by blocking the recruitment of the accessory molecule FADD, and this delay in apoptosis induction could allow for prolonged cytokine secretion by Hpylori-infected gastric epithelial cells.

efficiency of white lupin in the removal of mercury from contaminated soils: Soil and hydroponic experiments

This study examined the ability of the white lupin to remove mercury （Hg） from a hydroponic system （Hg concentrations 0, 1.25, 2.5, 5 and 10 μmol/L） and from soil in pots and lysimeters （total Hg concentration （19.2 ± 1.9） mg/kg availability 0.07%, and （28.9 ± 0.4） mg/kg availability 0.09%, respectively）, and investigated the accumulation and distribution of Hg in different parts of the plant. White lupin roots efficiently took up Hg, but its translocation to the harvestable parts of the plant was low. The Hg concentration in the seeds posed no risk to human health according to the recommendations of the World Health Organization, but the shoots should not be used as fodder for livestock, at least when unmixed with other fodder crops. The accumulation of Hg in the hydroponically-grown plants was linear over the concentration range tested. The amount of Hg retained in the roots, relative to the shoots, was almost constant irrespective of Hg dose （90%）. In the soil experiments, Hg accumulation increased with exposure time and was the greater in the lysimeter than in the pot experiments. Although Hg removal was the greater in the hydroponic system, revealing the potential of the white lupin to extract Hg, bioaccumulation was the greatest in the lysimeter-grown plants; the latter system more likely reflects the true behaviour of white lupin in the field when Hg availability is a factor that limits Hg removal. The present results suggest that the white lupin could be used in long-term soil reclamation strategies that include the goal of profitable land use in Hg-polluted areas.

Composite wedge failure using photogrammetric measurements and DFN-DeM modelling

Analysis and prediction of structural instabilities in open pit mines are an important design and operational consideration for ensuring safety and productivity of the operation. Unstable wedges and blocks occurring at the surface of the pit walls may be identified through three-dimensional(3D) image analysis combined with the discrete fracture network(DFN) approach. Kinematic analysis based on polyhedral modelling can be used for first pass analysis but cannot capture composite failure mechanisms involving both structurally controlled and rock mass progressive failures. A methodology is proposed in this paper to overcome such limitations by coupling DFN models with geomechanical simulations based on the discrete element method(DeM). Further, high resolution photogrammetric data are used to identify valid model scenarios. An identified wedge failure that occurred in an Australian coal mine is used to validate the methodology. In this particular case, the failure surface was induced as a result of the rock mass progressive failure that developed from the toe of the structure inside the intact rock matrix. Analysis has been undertaken to determine in what scenarios the measured and predicted failure surfaces can be used to calibrate strength parameters in the model.

Carbon stocks in a highly fragmented landscape with seasonally dry tropical forest in the Neotropics

Background:Global modeling of carbon storage and sequestration often mischaracterizes unique ecosystems such as the seasonally dry tropical forest of the central region of the Gulf of Mexico,because species diversity is usually underestimated,as is their carbon *** this study,aboveground and soil carbon stocks were estimated to determine the climate mitigation potential of this highly degraded landscape(est cover).Results:Tree species in the study area had carbon content values that were 30%–40%higher than the standard value proposed by the IPCC(i.e.,50%).Tropical oak forest in the region,despite its restricted distribution and low species richness,accounted for the highest mean carbon stocks per unit *** main factors driving spatial variability in carbon stocks were:maximum precipitation,soil organic matter,clay and silt *** strong relationship was found between aboveground carbon stocks and soil organic carbon in the study ***-fication of carbon stocks is an important consideration in the assessment of the conservation value of remnants of native vegetation in human-modified ***:This study demonstrates the importance of the highly fragmented tropical dry regions of the Neo-tropics in maintaining landscape functionality and providing key ecosystem services such as carbon *** results also highlight how crucial field-based studies are for strengthening the accuracy of global ***,this approach reveals the real contribution of ecosystems that are not commonly taken into account in the mitigation of climate change effects.

Novel electrode structure in a DBD reactor applied to the degradation of phenol in aqueous solution

Phenol degradation experimental results are presented in a similar wastewater aqueous solution using a non-thermal plasma reactor in a coaxial dielectric barrier discharge. The novelty of the work is that one of the electrodes of the reactor has the shape of a hollow screw which shows an enhanced efficiency compared with a traditional smooth structure. The experimentation was carried out with gas mixtures of 90% Ar-10% O2, 80% At-20% 02 and 0% Ar-100% 02. After one hour of treatment the removal efficiency was 76%, 92%, and 97%, respectively, assessed with a gas chromatographic mass spectrometry technique. For both reactors used, the ozone concentration was measured. The screw electrode required less energy, for all gas mixtures, than the smooth electrode, to maintain the same ozone concentration. On the other hand, it was also observed that in both electrodes the electrical conductivity of the solution changed slightly from ,-0.0115 S m^-1 up to -0.0430 S m^-1 after one hour of treatment. The advantages of using the hollow screw electrode structure compared with the smooth electrode were：（1） lower typical power consumption, （2） the generation of a uniform plasma throughout the reactor benefiting the phenol degradation, （3） a relatively lower temperature of the aqueous solution during the process, and （4） the plasma generation length is larger.
