Anti-viral activity of Staphylococcus aureus lysates against herpes simplex virus type-Ⅰinfection:an in vitro and in vivo study

AIM:To investigate the effect of Staphylococcus aureus(***)lysates(SALs)on herpes simplex virus type-Ⅰ(HSV1)infection in human corneal epithelial(HCE)cells and in a mouse model of HSV1 ***:HCE,Vero,HeLa,and BV2 cells were infected with HSV1[HSV1f strain,HSV1f;HSV-1-H129 with green fluorescent protein(GFP)knock-in,HSV1g].Pre-or post-infection,SAL at various concentrations was added to the culture medium for 24 *** fluorescence in HSV1g or plaque formation by HSV1f were *** effects of heat-treated SAL,precooled acetone-precipitated SAL,and SAL subjected to ultrafiltration(100 kDa)were *** effects of other bacterial components and lysates on HSV1 infection were also tested,including lipoteichoic acid(LTA),peptidoglycan(PGN),staphylococcal protein A(SPA),andα-hemolysin from ***(α-toxin)as well as lysates from a wild-type *** strain,***,and Escherichia coli(W-SAL,SEL,and ECL,respectively).In addition,SAL eye drops were applied topically to BALB/c mice with HSV1 keratitis,followed by in vivo ***:The cytopathic effect,plaque formation(HSV1f),and GFP expression(HSV1g)in infected cells were inhibited by SAL in a dose-dependent *** active component of SAL(≥100 kDa)was heat-sensitive and retained activity after acetone *** HSV1ginfected cells,treatment with LTA-sa,α-toxin,PGN-sa,or SPA did not inhibit GFP ***,W-SAL,and SEL(but not ECL)decreased GFP *** mice with HSV1 keratitis,SAL reduced corneal lesions by 71%.CONCLUSION:The results of this study demonstrate that SAL can be used to inhibit HSV1 infection,particularly *** studies are needed to determine the active components and mechanism underlying the effects of SAL.

转台用液压油的非牛顿特性及随时间推移流变特性的变化,将影响油液在转台内的流动以及承载能力。以双环形静压转台为研究对象,针对油液的流变时变特性,提出了一种虚外力项LBM方法,并利用泊肃叶流的理论解进行了验证,通过仿真分析发现,双环形转台侧边入油口可有效增加中心凹槽的压力,改善承载能力。油液流变时变特性和转台深度特征尺寸是两个重要因素。相对于速度分布,封油边深度及油腔深度对承载能力的影响更加显著。随时间推移,油液黏度增大,使油液的流动减缓,速度减小,承载能力变弱,因此建议定期更换液压油,以保证转台的正常工作。

Effect of Autophagy Over Liver Diseases

In recent years, increasingly evidences show that autophagy plays an important role in the pathogenesis and development of liver diseases, and the relationship between them has increasingly become a focus of concern. Autophagy refers to the process through which the impaired organelles, misfolded protein, and intruding microorganisms is degraded by lysosomes to maintain stability inside cells. This article states the effect of autophagy on liver diseases （hepatic fibrosis, fatty liver, viral hepatitis, and liver cancer）, which aims to provide a new direction for the treatment of liver diseases.

The Effect of Total Cholesterol on Myocardial Infarction in Chinese Male Hypertension Population

Objective Total cholesterol （TC） is an important risk factor for myocardial infarction （M1）, but the effect of TC on MI in Chinese male hypertension population has not been well documented. We conducted a prospective cohort study to determine the incidence and relative risk for M1 across a wide range of TC levels in Chinese male hypertension population. Methods A cohort of 5 298 male employees aged 18-74 years recruited from Capital Steel and Iron Company in Beijing of China in 1974-1980 was followed up for an average of 20.84 years. A total of 122 incident M1 cases were identified during the period of follow-up. Results The incidence of MI among participants with elevated TC and those with desirable TC in male non-hypertension population was 137.20 and 63.81 per 100 000 person-years, respectively; and the corresponding incidence in male hypertension population was 279.80 and 130.96 per 100 000 person-years, respectively. After adjustment for important covariables, 10.38%, 16.71%, and 23.80% of M1 cases were attributable to hypertension, elevated TC, and hypertension plus elevated TC, respectively. In male hypertension population, the multivariate adjusted hazard ratios of MI were 1.21, 2.39, 3.38, and 3.95 for participants with TC level of 5.17-5.68, 5.69-6.20, 6.21-6.71, and ≥6.72 mmol/L, compared with those with TC〈5.17 mmol/L. The corresponding population attributable risks were 2.92%, 9.20%, 8.87%, and 9.84%, respectively. Conclusion Elevated TC is an important independent risk factor of M1 both in male non-hypertension and hypertension populations. There is a linear association between TC level and M1 incidence in Chinese male hypertension population.

BcI-XL Small Interfering RNA Enhances Sensitivity of Hepg2 Hepatocellular Carcinoma Cells to 5-Fluorouracil and Hydroxycamptothecin

Changes in drug sensitivity in Bcl-XL small interfering RNA (siRNA) transfected Hepg2hepatocellular carcinoma cells were investigated in this ***-XL siRNA and negative siRNA expressionvector were constructed and stably transfected into Hepg2 *** transcription (RT)-PCR,westernblot and immunofluorescence were used to detect the target gene expression at mRNA and protein *** sensitivity of the cells to 5-fluorouracil (5-FU) and hydroxycamptothecin (HCPT) were evaluated *** Bcl-XL mRNA and protein expression levels in Bcl-XL siRNA transfectants were reducedcompared with negative siRNA transfectants or mock *** results showed that Bcl-XL siRNAtransfected cells have a higher cell inhibition rate than negative vector transfected cells or untreated cellsafter treatment with 13,130,1300 and 13,000 mg/L of ***-XL siRNA transfected cells also showedincreased drug-sensitivity compared with negative vector transfected cells or untreated cells after treatmentwith 0.18,0.36,0.72 and 1.44 mg/L *** cytometry (FCM) results demonstrated that the sub-G1population increased in the Bcl-XL siRNA group,compared with the negative siRNA group and untreatedcontrol group,after the addition of 5-FU (1300 mg/L) and HCPT (0.72 mg/L),siRNA targeting Bcl-XL genecan specifically down-regulate Bcl-XL expression in Hepg2 cells,and can increase spontaneous cell apoptosisand sensitize cells to 5-FU or HCPT.

甘薯在种植过程中因连年种植出现的病害发生加重、产量降低、品质变劣等因素严重制约了甘薯产业的发展。甘薯脱毒技术是脱除病毒、恢复品种种性、提高甘薯产量和品质的有效途径,但生产中脱毒苗的供应能力不足,为了探索其解决办法,以徐薯18、龙薯9号为试验材料,根据甘薯龙薯9号扦插苗自身吸收矿质元素的量计算所得的基质培配方龙9以及1/2龙9、MS、1/2MS为处理,进行了脱毒苗组培扩繁的比较探究。结果表明,徐薯18和龙薯9号1/2龙9配方的茎节数和茎长均为最好,但试验过程中该配方出现了叶片黄化现象,故对1/2龙9配方进行了完善,并在徐薯18、龙薯9号、烟薯25、绵薯10号、遂宁524及北京1号6个甘薯品种进行了验证,结果表明1/2改良龙9配方处理下的各品种茎长和茎节数均优于MS配方,这对于解决生产中脱毒苗供应不足的问题以及经济效益的增加有极其重要的作用。

Effect of dielectric material on the uniformity of nanosecond pulsed dielectric barrier discharge

Dielectric barrier discharge(DBD)is considered as a promising technique to produce large volume uniform plasma at atmospheric pressure,and the dielectric barrier layer between the electrodes plays a key role in the DBD processes and enhancing discharge *** this work,the uniformity and discharge characteristics of the nanosecond(ns)pulsed DBD with dielectric barrier layers made of alumina,quartz glass,polycarbonate(PC),and polypropylene(PP)are investigated via discharge image observation,voltage-current waveform measurement and optical emission spectral *** analyzing discharge image by gray value standard deviation method,the discharge uniformity is quantitatively *** effects of the space electric field intensity,the electron density(Ne),and the space reactive species on the uniformity are studied with quantifying the gap voltage Ug and the discharge current Ig,analyzing the recorded optical emission spectra,and simulating the temporal distribution of Ne with a one-dimensional fluid *** is found that as the relative permittivity of the dielectric materials increases,the space electric field intensity is enhanced,which results in a higher Ne and electron temperature(Te).Therefore,an appropriate value of space electric field intensity can promote electron avalanches,resulting in uniform and stable plasma by the merging of electron ***,an excessive value of space electric field intensity leads to the aggregation of space charges and the distortion of the space electric field,which reduce the discharge *** surface roughness and the surface charge decay are measured to explain the influences of the surface properties and the second electron emission on the discharge *** results in this work give a comprehensive understanding of the effect of the dielectric materials on the DBD uniformity,and contribute to the selection of dielectric materials for DBD reactor and the realization of atmospheric pre

Perovskite oxides with diverse composition and structure have exhibited grand advances in boosting the oxygen reduction and evolution reaction(ORR/OER),which are essential for the reversible protonic ceramic electrochemical cell(R-PCEC)toward the sustainable hydrogen production and ***,enhancement of their activity and stability remains ***,we develop the Ta-regulated BaCo_(0.7)Fe_(0.3)O_(3-δ)perovskite oxygen electrode(Ba(Co_(0.7)Fe_(0.3))_(1-x)Ta_xO_(3-δ))with abundant oxygen defects and achieve the simultaneous enhancement in the electrocatalytic activity and stability toward ORR and ***-fabricated R-PCEC with(Ba(Co_(0.7)Fe_(0.3))_(0.9)Ta_(0.1)O_(3-δ))(BCFT10)oxygen electrode performs high power density of 1.47 W·cm^(-2)at 650℃in fuel cell mode,and the current density is up to-2.11 A·cm^(-2)at 1.4 V at 650℃in electrolysis mode,as well as the good stability in both the fuel cell and electrolysis ***,the cell also demonstrates a stable cycling operation between fuel cell and electrolysis mode,suggesting a great potential of BCFT10 as oxygen electrode material for R-PCECs.

应用反应力场分子动力学方法,模拟了单层氧化石墨烯在径向压缩作用下的褶皱过程,研究了含氧基团(羟基、环氧基)对氧化石墨烯褶皱行为以及褶皱球结构稳定性的影响.当石墨烯仅含羟基时,该类氧化石墨烯呈现出“推进式”的褶皱行为,而当石墨烯仅含环氧基时,该类氧化石墨烯则呈现出片层与片层“贴合式”的褶皱行为.褶皱行为的不同决定了氧化石墨烯最终褶皱球结构的差异.通过分析原子级势能增量分布与C—C成、断键位置之间的关系,发现氧化石墨烯上C—C成、断键主要发生在变形较为严重的区域,且相较于羟基,环氧基对与其连接的C—C键具有更强的削弱作用.为了研究氧化石墨烯褶皱球的结构稳定性,模拟了其在无约束条件下的释放过程.发现氧化石墨烯褶皱球的结构稳定性取决于其中C—C成、断键数量,即C—C成、断键的数量越多,结构越稳定,且在同一氧化率下,褶皱球的结构稳定性随环氧基比例的增大而提高.本研究表明,通过改变氧化石墨烯片层上含氧基团的相对比例,可实现对其褶皱球稳定性的控制.
