Endomembrane-biased dimerization of ABCG16 and ABCG25 transporters determines their substrate selectivity in ABA-regulated plant growth and stress responses

ATP-binding cassette(ABC)transporters are integral membrane proteins that have evolved diverse func-tions fulfilled via the transport of various substrates.In Arabidopsis,the G subfamily of ABC proteins is particularly abundant and participates in multiple signaling pathways during plant development and stress responses.In this study,we revealed that two Arabidopsis ABCG transporters,ABCG16 and ABCG25,engage in ABA-mediated stress responses and early plant growth through endomembrane-specific dimerization-coupled transport of ABA and ABA-glucosyl ester(ABA-GE),respectively.We first revealed that ABCG16 contributes to osmotic stress tolerance via ABA signaling.More specifically,ABCG16 induces cellular ABA efflux in both yeast and plant cells.Using FRET analysis,we showed that ABCG16 forms oblig-atory homodimers for ABA export activity and that the plasma membrane-resident ABCG16 homodimers specifically respond to ABA,undergoing notable conformational changes.Furthermore,we demonstrated that ABCG16 heterodimerizes with ABCG25 at the endoplasmic reticulum(ER)membrane and facilitates the ER entry of ABA-GE in both Arabidopsis and tobacco cells.The specific responsiveness of the ABCG16-ABCG25 heterodimer to ABA-GE and the superior growth of their double mutant support an inhib-itory role of these twoABCGs in early seedling establishment via regulation of ABA-GE translocation across the ER membrane.Our endomembrane-specific analysis of the FRET signals derived from the homo-or heterodimerized ABcG complexes allowed us to link endomembrane-biased dimerization to the transloca-tion of distinct substrates by ABcG transporters,providing a prototypic framework for understanding the omnipotence of ABcG transporters in plant development and stress responses.

Multiple Nanoparticles Coupling Strategy for Enhancing Optical Filter Performance of Spectral Splitter Used in Photovoltaic/Thermal System

Selective absorptive nanofluid can pre-absorb certain sunlight wavelength that cannot be used by PV and transmits remaining sunlight to the surface of PV,which can decouple PV from the thermal receiver spatially.In order to improve the harvesting of electricity and high-temperature thermal nanofluid,it is important to design an optimal optical filter window(transmit sunlight with wavelengths of 732-1067 nm to the surface of the photovoltaic cell and absorb the remaining sunlight).However,designing optimal optical filter is facing following challenges:(1) inherently narrow selective absorptivity property of single nanoparticle;(2) simplified numerical calculation method calculating transmittance;(3) ignoring the shape of the nanoparticle.In this study,the idea of using multiple nanoparticles coupling effect to design an optical filter is proposed,which can superimpose the narrow absorption bandwidth of different nanoparticles to obtain a wide absorption bandwidth of the whole system.In addition,an improved transmission method considering light-matter interaction at air/vessel and liquid/vessel interfaces is adopted to compute the transmittance.The results calculated by improved transmission method are more accurate than widely used traditional Lambert-Beer law,which is verified by experimental test.Furthermore,the effect of nanoparticle shape on spectral transmittance is also investigated,which shows that spiny Ag can approximately extend absorbance from 400 nm to 600 nm compared to nanosphere silver.Finally,the results show that optical filter efficiency of nanofluids with multiple nanoparticles coupling(Ag,spiny Ag,ZnO,ITO) can reached up to 35%.

Through exploring the stress corrosion cracking(SCC)behaviors of the as-cast Mg-8%Li and Mg-8%Li-6%Zn-1.2%Y alloys in a 0.1 M NaCl solution,it revealed that the SCC susceptibility index(I_(SCC))of the Mg-8%Li alloy was 47%,whilst the I_(SCC)of the Mg-8%Li-6%Zn-1.2%Y alloy was 68%.Surface,cross-sectional and fractography observations indicated that for the Mg-8%Li alloy,theα-Mg/β-Li interfaces acted as the preferential crack initiation sites and propagation paths during the SCC process.With regard to the Mg-8%Li-6%Zn-1.2%Y alloy,the crack initiation sites included the I-phase and the interfaces of I-phase/β-Li andα-Mg/β-Li,and the preferential propagation paths were the I-phase/β-Li andα-Mg/β-Li interfaces.Moreover,the SCC of the two alloys was concerned with hydrogen embrittlement(HE)mechanism.

Near-infrared(NIR)light has shown great potential for military and civilian applications owing to its advantages in the composition of sunlight,invisibility to human eyes,deeper penetration into biological tissues,and low optical loss in optical fibers.Therefore,organic optoelectronic materials that can absorb or emit NIR light have aroused great scientific interest in basic science and practical applications.Based on these NIR organic optoelectronic materials,NIR optoelectronic devices have been greatly improved in performance and application.In this review,the representative NIR organic optoelectronic materials used in organic solar cells,organic photodetectors,organic light-emitting diodes,organic lasers,and organic optical waveguide devices are briefly introduced,and the potential applications of each kind of device are briefly summarized.Finally,we summarize and take up the development of NIR organic optoelectronic materials and devices.

Three-dimensional stability calculation method for high and large composite slopes formed by mining stope and inner dump in adjacent open pits

The 2D limit equilibrium method is widely used for slope stability analysis.However,with the advancement of dump engineering,composite slopes often exhibit significant 3D mechanical effects.Consequently,it is of significant importance to develop an effective 3D stability calculation method for composite slopes to enhance the design and stability control of open-pit slope engineering.Using the composite slope formed by the mining stope and inner dump in Baiyinhua No.1 and No.2 open-pit coal mine as a case study,this research investigates the failure mode of composite slopes and establishes spatial shape equations for the sliding mass.By integrating the shear resistance and sliding force of each row of microstrip columns onto the bottom surface of the strip corresponding to the main sliding surface,a novel 2D equivalent physical and mechanical parameters analysis method for the strips on the main sliding surface of 3D sliding masses is proposed.Subsequently,a comprehensive 3D stability calculation method for composite slopes is developed,and the quantitative relationship between the coordinated development distance and its 3D stability coefficients is examined.The analysis reveals that the failure mode of the composite slope is characterized by cutting-bedding sliding,with the arc serving as the side interface and the weak layer as the bottom interface,while the destabilization mechanism primarily involves shear failure.The spatial form equation of the sliding mass comprises an ellipsoid and weak plane equation.The analysis revealed that when the coordinated development distance is 1500 m,the error rate between the 3D stability calculation result and the 2D stability calculation result of the composite slope is less than 8%,thereby verifying the proposed analytical method of equivalent physical and mechanical parameters and the 3D stability calculation method for composite slopes.Furthermore,the3D stability coefficient of the composite slope exhibits an exponential correlation with the coordinated development dista

A semi-analytical model for coupled flow in stress-sensitive multi-scale shale reservoirs with fractal characteristics

A large number of nanopores and complex fracture structures in shale reservoirs results in multi-scale flow of oil. With the development of shale oil reservoirs, the permeability of multi-scale media undergoes changes due to stress sensitivity, which plays a crucial role in controlling pressure propagation and oil flow. This paper proposes a multi-scale coupled flow mathematical model of matrix nanopores, induced fractures, and hydraulic fractures. In this model, the micro-scale effects of shale oil flow in fractal nanopores, fractal induced fracture network, and stress sensitivity of multi-scale media are considered. We solved the model iteratively using Pedrosa transform, semi-analytic Segmented Bessel function, Laplace transform. The results of this model exhibit good agreement with the numerical solution and field production data, confirming the high accuracy of the model. As well, the influence of stress sensitivity on permeability, pressure and production is analyzed. It is shown that the permeability and production decrease significantly when induced fractures are weakly supported. Closed induced fractures can inhibit interporosity flow in the stimulated reservoir volume (SRV). It has been shown in sensitivity analysis that hydraulic fractures are beneficial to early production, and induced fractures in SRV are beneficial to middle production. The model can characterize multi-scale flow characteristics of shale oil, providing theoretical guidance for rapid productivity evaluation.

Semiannual oscillation,annual oscillation,quasibiennial oscillation,and solar cycle variation of the OH airglow emission in the mesopause region

The vertically integrated emission rate,centroid altitude,peak emission rate,and peak height of the hydroxyl(OH)airglow were calculated from Thermosphere Ionosphere Mesosphere Energetics and Dynamics(TIMED)/Sounding of the Atmosphere using Broadband Emission Radiometry(SABER)observations to study the seasonal and interannual variations in the intensity and location of the OH emission.The emission rate is inversely proportional to the height of the emission,with the semiannual oscillation dominating at low latitudes and the annual oscillation dominating at higher latitudes.The OH emission is modulated by the quasibiennial oscillation at the equator,and the quasibiennial oscillation signal is weak at other latitudes.We represented the vertical transport of atomic oxygen by using atomic oxygen concentrations obtained from a global atmospheric model,the Specified Dynamics Whole Atmosphere Community Climate Model with thermosphere and ionosphere eXtension simulations.Compared with the amplitudes of the migrating diurnal tide(DW1)calculated from temperature data observed by TIMED/SABER,we found that both the vertical transport of atomic oxygen and DW1 amplitudes in the equatorial region exhibit semiannual oscillation and quasibiennial oscillation,which have a strong correlation with the variations in the amplitude and phase of semiannual oscillation and quasibiennial oscillation in OH emission.It is likely that the DW1 affects the vertical transport of atomic oxygen that is involved in the reaction to produce O3,thus affecting the OH emission.We analyzed the relationship between OH emission and solar activity by using the solar radio flux at 10.7 cm as a proxy for solar activity.The results showed that the OH emission is well correlated with solar activity,and the modulation of OH emission by solar activity has a significant latitudinal variation.The small correlation between emission height and solar activity indicates that solar activity modulates OH emission mainly through chemical rather than dynamic processes.

Percutaneous transhepatic stenting for acute superior mesenteric vein stenosis after pancreaticoduodenectomy with portal vein reconstruction:A case report

BACKGROUND Percutaneous transhepatic stent placement has become a common strategy for the postoperative treatment of portal vein(PV)/superior mesenteric veins(SMV)stenosis/occlusion.It has been widely used after liver transplantation surgery;however,reports on stent placement for acute PV/SMV stenosis after pancreatic surgery within postoperative 3 d are rare.CASE SUMMARY Herein,we reported a case of intestinal edema and SMV stenosis 2 d after pancreatic surgery.The patient was successfully treated using stent grafts.Although the stenosis resolved after stent placement,complications,including bleeding,pancreatic fistula,bile leakage,and infection,made the treatment highly challenging.The use of anticoagulants was adjusted multiple times to prevent venous thromboembolism and the risk of bleeding.After careful treatment,the patient stabilized,and stent placement effectively managed postoperative PV/SMV stenosis.CONCLUSION Stent placement is effective and feasible for treating acute PV/SMV stenosis after pancreatic surgery even within postoperative 3 d.

Enzymatic self-assembly/disassembly turns “ON”/“OFF” the mimetic hydrolytic activity of histidine nanofibers

Mimetic enzymes are devised as alternates or supplements of natural enzymes in broad fields but regulating their activities in a switchable manner remains challenging.Herein,we proposed an enzymatic self-assembly/disassembly strategy to address this issue.A peptide molecule Nap FFEYIH(YH) was rationally designed which,after self-assembling into nanofibers,lined up the histidine moieties to form active hydrolysis centers for mimicking hydrolase activity.Enzymatic dephosphorylation of Nap FFEYp IH(Yp H) by alkaline phosphatase to yield YH also turned “ON” the hydrolase activity.In turn,phosphorylation of YH by phosphokinase epidermal growth factor receptor to yield Yp H disassembled the nanofibers and thus turned the activity “OFF”.As such,the “ON”/“OFF” of the mimetic hydrolase activities could be regulated under physiological conditions through ALP/EGFR-mediated self-assembly/disassembly of histidine nanofibers.This work provides a feasible strategy for the on-demand fabrication of artificial enzymes with controllable and superior activities.

The practical application of lithium-sulfur(Li-S)batteries,as promising next-generation batteries,is hindered by their shuttle effect and the slow redox kinetics.Herein,a tungsten and molybdenum nitride heterostructure functionalized with hollow metal-organic framework-derived carbon(W_(2)N/Mo_(2)N)was proposed as the sulfur host.The hollow spherical structure provides storage space for sulfur,enhances electrical conductivity,and inhibits volume expansion.The metal atoms in the nitrides bonded with lithium polysulfides(Li PSs)through Lewis covalent bonds,enhancing the high catalytic activity of the nitrides and effectively reducing the energy barrier of Li PSs redox conversion.Moreover,the high intrinsic conductivity of nitrides and the ability of the heterostructure interface to accelerate electron/ion transport improved the Li+transmission.By leveraging the combined properties of strong adsorption and high catalytic activity,the sulfur host effectively inhibited the shuttle effect and accelerated the redox kinetics of Li PSs.High-efficiency Li+transmission,strong adsorption,and the efficient catalytic conversion activities of Li PSs in the heterostructure were experimentally and theoretically verified.The results indicate that the W_(2)N/Mo_(2)N cathode provides stable,and long-term cycling(over 2000 cycles)at 3 C with a low attenuation rate of 0.0196%per cycle.The design strategy of a twinborn nitride heterostructure thus provides a functionalized solution for advanced Li-S batteries.
