Preparation of PrP-specific Polyclonal Antibody via Immunization of PRNP-knockout Mice with Recombinant Human PrP Protein

Objective The definite diagnosis of human and animal prion diseases depends on the examination of special pathological changes and/or detection of PrPSc in the brain tissues of suspected ***,developing methods to obtain PrP antibody with good specificity and sensitivity is fundamental for prion *** We prepared a PrP-specific polyclonal antibody(pAb P54)in a PRNP-knockout mouse model via immunization with recombinant full-length human PrP protein residues 23–***,we verified that pAb in Western blot,immunohistochemistry(IHC),and immunofluorescent(IFA)*** Western blot illustrated that the newly prepared pAb P54 could react with recombinant PrP protein,normal brain PrPC from healthy rodents and humans,and pathological PrPSc in the brains of experimental rodents infected with scrapie and humans infected with different types of prion *** electrophoretic patterns of brain PrPC and PrPSc observed after their reaction with pAb P54 were nearly identical to those produced by commercial PrP monoclonal *** glycosylated PrP molecules in the brain homogenates were clearly demonstrated in the reactions of these molecules with pAb *** assay revealed apparent PrP deposits in the GdnCl-treated brain slices of 139 A-infected mice and 263 K-infected *** tests with pAb P54 also showed clear green signals surrounding blue-stained cell *** The newly prepared pAb P54 demonstrated reliable specificity and sensitivity and,thus,may have potential applications not only in studies of prion biology but also in the diagnosis of human and experimental rodent prion diseases.

Objective To describe the global profiles of acetylated proteins in the brains of scrapie agents 139Aand ME7-infected mice collected at mid-early,mid-late,and terminal *** The acetylated proteins from the cortex regions of scrapie agent(139A-and ME7)-infected mice collected at mid-early(80 days postinfection,dpi),mid-late(120 dpi),and terminal(180 dpi) stages were extracted,and the global profiles of brain acetylated proteins were assayed with proteomic mass *** proteins in the infected mice showing 1.5-fold higher or lower levels than that of agematched normal controls were considered as differentially expressed acetylated peptides(DEAPs).Results A total of 118,42,and 51 DEAPs were found in the brains of 139A-80,139A-120,and 139A-180dpi mice,***,390,227,and 75 DEAPs were detected in the brains of ME7-80,ME7-120,and ME7-180 dpi mice,*** overwhelming majority of DEAPs in the mid-early stage were down-regulated,and more portions of DEAPs in the mid-late and late stages were *** 22.1%(328/1,485) of acetylated peptides mapped to 74 different proteins were mitochondrial *** Encyclopedia of Genes and Genomes pathway analysis identified 39(80dpi),13(120 dpi),and 10(180 dpi) significantly changed pathways in 139A-infected ***,55,25,and 18 significantly changed pathways were observed in the 80,120,and 180 dpi samples of139A-and ME7-infected mice(P < 0.05),*** pathways were commonly involved in all tested ***,many steps in the citrate cycle(tricarboxylic acid cycle) were affected,represented by down-regulated acetylation for relevant enzymes in the mid-early stage and upregulated acetylation in the mid-late and late *** Our data here illustrated the changes in the global profiles for brain acetylated proteins during prion infection,showing remarkably inhibited acetylation in the early stage and relatively enhanced acetylation in the la

In this article, a new approach for modeling multiinput multi-output （MIMO） systems with unknown nonlinear interference is introduced. The semiparametric theory based MIMO model is established, and Kernel estimation is applied to combat the nonlinear interference. Furthermore, we derive MIMO capacity for these systems and explore the asymptotic properties of the new channel matrix via theoretical analysis. The simulation results show that the semiparametric theory based modeling and kernel estimation are valid to combat this kind of interference.

Effect of Ni/(La+Mg) Ratio on Structure and Electrochemical Performance of La-Mg-Ni Alloy System

As the alloy with the most suitable Ni/（La＋Mg） ratio has higher capacity and good cycle stability,the effects of Ni/（La＋Mg） ratios on the electrochemical performances of the La0.80Mg0.20Nix （x=3.5 to 5.0） alloys have been investigated to find the most suitable Ni/（La＋Mg） ratio. The results of XRD and SEM observations show that the phase composition of the alloys varies with different Ni/（La＋Mg） ratios. When Ni/（La＋Mg） is not more than 4.25,all the alloys contain LaNi5 and （La,Mg）2Ni7 phases,in addition,the LaMg and （La,Mg）Ni3 phases exist in the x=3.5 and 3.75 alloys,respectively. The LaMg2Ni9 phase exists in the x=4.25 alloy. There are the LaNi5 and LaMg2Ni9 phases in the x=4.5,4.75,and 5.0 alloys. The phase abundance and cell volume change with different Ni content. When the Ni/（La＋Mg） ratio is not more than 4.25,the alloys possess excellent activation capability,however,the activation capabilities of the alloys decrease with a further increase in the Ni/（La＋Mg） ratio. With increasing the Ni/（La＋Mg） ratio,the maximum discharge capacities,the medium voltages,and the cycle stabilities of the alloys first increase and then decrease. When the Ni/（La＋Mg） ratio is 3.75,the corresponding alloy has the maximum discharge capacity among all the alloys. However,the cycle stability of the Ni/（La＋Mg）=4.0 alloy is better than that of the others.

Effects of Two Curcuminoids on Candida albicans

Objective To investigate and compare the action of curcuminoids on the causal pathogens of Candida albicans growth. Methods The effects of curcumin (CUR) and demethoxycurcumin (DMC) on C. albicans growth were first investigated and compared by microcalorimetry coupled with multiple analytical methods. The quantitative thermo-kinetic parameters obtained from these curves were analyzed to show difference of the actions. Results By analyzing the main parameters screened from principal component analysis together with 50% inhibiting concentration values, it was demonstrated that both CUR and DMC showed good antifungal activities and CUR was stronger. It was further concluded from structure-activity relationship that the existence of methoxy group might enhance lipophilicity of the mother nucleus, which made it easier for the molecular to enter into the cell membrane of fungi to inhibit its growth. Conclusion This study provides a new method for screening new antifungal agents with high efficacy and low toxicity. Meanwhile, it contributes to the application of curcuminoids as food additive, colorant, and drug. Microcalorimetry is real-time, online, and dynamic, and it could be used to characterize the subtle difference among the effects of synthetic and natural products on the vital process of fungi.
