Electroacupuncture with different waveforms for primary dysmenorrhea: A randomized controlled trial

Obj.ctive: To observe and compare the clinical effects of different electroacupuncture waveforms on primary dysmenorrhea. Methods: This was a prospective, randomized, three-group, parallel-controlled trial. Participants with primary dysmenorrhea were randomly divided into dense-sparse wave, continuous wave, and discontinuous wave groups in a 1:1:1 ratio. Two lateral Ciliao (BL 32) points were used. All three groups started treatment 3–5 days before menstruation, once a day for six.sessions per course of treatment, one course of treatment per menstrual cycle, and three menstrual cycles. The primary outcome measure was the proportion with an average visual analog scale (VAS) score reduction of ≥50% from baseline for dysmenorrhea in the third menstrual cycle during treatment. The secondary outcome measures included changes in dysmenorrhea VAS scores, Cox.Menstrual Symptom Scale scores and the proportion of patients taking analgesic drugs. Results: The proportion of cases where the average VAS score for dysmenorrhea decreased by ≥50% from baseline in the third menstrual cycle was not statistically significant (P > .05). Lassa virus (LASV) is an enveloped, negative-sense RNA virus that causes Lassa hemorrhagic fever. Successful entry of LASV requires the viral glycoprotein 1 (GP1) to undergo a receptor switch from its primary receptor alpha-dystroglycan (α-DG) to its endosomal receptor lysosome-associated membrane protein 1 (LAMP1). A conserved histidine triad in LASV GP1 has been reported to be responsible for receptor switch. To test the hypothesis that other non-conserved residues also contribute to receptor switch, we constructed a series of mutant LASV GP1 proteins and tested them for binding to LAMP1. Four residues, L84, K88, L107, and H170, were identified as critical for receptor switch. Substituting any of the four residues with the corresponding lymphocytic choriomeningitis virus (LCMV) residue (L84 N, K88E, L10F, and H170S) reduced the binding affinity of LASV GP1 for LAMP1. Moreover, all mutations caused decreases in glycoprotein precursor (GPC)-mediated membrane fusion at both pH 4.5 and 5.2. The infectivity of pseudotyped viruses bearing either GPCL84N or GPCK88E decreased sharply in multiple cell types, while L107F and H170S had only mild effects on infectivity. Using biolayer light interferometry assay, we found that all four mutants had decreased binding affinity to LAMP1, in the order of binding affinity being L84 N > L107F > K88E > H170S. The four amino acid loci identified for the first time in this study have important reference significance for the in-depth investigation of the mechanism of receptor switching and immune escape of LASV occurrence and the development of reserve anti-LASV infection drugs. © 2024 The Authors

Obj.ctive: To evaluate the anti-fatigue effects of different ex.racts from Cistanche tubulosa (Schenk) Wight (C. tubulosa, Rou Cong Rong), focusing on central and ex.rcise-induced fatigue in mice. This study investigated the pharmacological effects of the total oligosaccharides, polysaccharides, and phenylethanoid glycosides (CPhGs) ex.racted from C. tubulosa. Methods: Models of sleep deprivation and forced swimming fatigue were established to simulate central and ex.rcise-induced fatigue. The mice were treated with different ex.racts of C. tubulosa, and their effects were assessed using behavioral tests to measure ex.rcise capacity, learning, and memory function. Biochemical analyses were performed to evaluate the changes in serum and brain neurotransmitter levels, liver and muscle glycogen storage, and various fatigue-related biomarkers. Results: This study found that treatment with C. tubulosa ex.ract improved ex.rcise capacity, learning, and memory in mice. Total oligosaccharides from C. tubulosa enhanced adrenocorticotropic hormone, cholinesterase, and thyroid-stimulating hormone levels, reduced cortisol levels in central fatigue models, and ameliorated biochemical markers of ex.rcise-induced fatigue, including lowering lactic acid, blood urea nitrogen, and malondialdehyde levels. Among the tested ex.racts, the total oligosaccharides showed the most comprehensive anti-fatigue effects. Conclusion: The anti-fatigue effects of C. tubulosa, particularly those of its total oligosaccharides, are pronounced in both central and ex.rcise-induced fatigue. These effects are mediated by the regulation of neurotransmitter levels, enhancement of glycogen storage, and improvement of antiox.dant enzyme activity, suggesting potential therapeutic benefits in fatigue-related conditions. © 2024 Beij.ng University of Chinese Medicine

Background: Recently, microbotulinum, a new technique that involves inj.cting botulinum tox.n type A (BoNTA) microdroplets into superficial cutaneous tissue, has gained popularity. The precise distribution of BoNTA in the targeted area profoundly affects outcomes. Many factors may influence the effective area of BoNTA in the dermis. This study aimed to determine the dermal distribution properties of BoNTA to guide microbotulinum inj.ction. Methods: Ten healthy males aged 18–65 years without BoNTA treatment in the previous year were recruited to receive intradermal inj.ctions in the chest and back. Ultrasound was used to ensure the intradermal delivery of inj.ctions and measure the dermal thickness. The minor iodine starch test was performed at baseline and 3 days, 7 days, 21 days, 1 month, and 2 months after treatment. Results: All participants received intradermal inj.ctions. The dermis was thinner on the chest (thickness, 0.20 ± 0.03 cm) than on the back (thickness, 0.39 ± 0.07 cm) (Pj.ction in the thicker dermis had a significantly smaller effective area at every follow-up visit. The drug concentration did not affect the effective area ex.ept at 3 days after treatment. Inj.ction speed did not influence the effective area at any follow-up visits. Conclusion: An inj.ction in a thicker dermis leads to a smaller effective area for intradermal inj.ctions. When the BoNTA dose is the same, the drug concentration and inj.ction speed do not matter. Obj.ctive: To investigate the mechanism underlying the effects ex.rted by the Qizhu prescription (QZP) in breast cancer (BC), and the respective targets. Methods: Ex.ression data from the ArrayEx.ress and The Cancer Genome Atlas (TCGA) were used to identify differentially ex.ressed genes (DEGs) in BC. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses were performed on the DEGs to identify genes involved in protein–protein interactions. Molecular docking was used to ex.lore the dynamic relationship between active molecules and targets. Cell function ex.eriments and animal studies were conducted to evaluate the effects of hub genes and active QZP compounds on BC cell behavior. Results: Among the 25 evaluated BC-related targets of QZP, matrix.metalloproteinase-1 (MMP1) and epidermal growth factor receptor (EGFR) ex.ibited the highest degrees of dysregulation. GO and KEGG enrichment analyses revealed that the anti-BC targets of QZP primarily affected drug responses and pathways in cancer cells. Molecular docking analysis suggested potential interactions between EGFR and quercetin/luteolin, as well as between MMP1 and luteolin/kaempferol/quercetin. Quercetin significantly reduced BC cell proliferation, migration, invasion, and tumor development in vivo. Treatment of BC cells with quercetin decreased the ex.ression or activation of several associated proteins. Conclusion: The findings of our study provide new insights into the therapeutic potential of traditional Chinese medicine against BC, with particular reference to QZP. © 2024 Beij.ng University of Chinese Medicine

In this paper,numerical ex.eriments are carried out to investigate the impact of penalty parameters in the numerical traces on the resonance errors of high-order multiscale discontinuous Galerkin(DG)methods(dong et *** j.Sci Comput 66:321–345,2016;dong and Wang in j.Comput Appl Math 380:1–11,2020)for a one-dimensional stationary Schrödinger *** work showed that penalty parameters were required to be positive in error analysis,but the methods with zero penalty parameters worked fine in numerical simulations on coarse *** this work,by performing ex.ensive numerical ex.eriments,we discover that zero penalty parameters lead to resonance errors in the multiscale DG methods,and taking positive penalty parameters can effectively reduce resonance errors and make the matrix.in the global linear system have better condition numbers.
