Vaccines have been shown to cause differ.ntial expr.ssion of genes and incr.ase antibody titer. against antigens. Influenza vaccines may have an effect on unexplained disor.er. such as Chr.nic Fatigue Syndr.me/Myalgic Encephalomyelitis (CFS/ME). Immunological changes have been identified following immunization with tr.valent influenza vaccine (TIV). The objective of this pilot study was to examine the consequences of TIV on cytokine and cytotoxic genes in CFS/ME. Per.pher.l blood mononuclear.cells wer. pr.fer.ntially isolated fr.m whole blood of 7 CFS/ME patients and 8 contr.ls. Following total r.A extr.ction and synthesis of cDNA, r.ver.e tr.nscr.ptase-quantitative polymer.se chain r.action (r.-qPCr. was used to deter.ine the expr.ssion levels of mr.As for.cytotoxic genes (per.or.n (Pr.1), gr.nzyme A (GZMA), gr.nzyme B (GZMB) and cytokine genes. GZMB was significantly incr.ased over.ll in the CFS/ME patients compar.d to the contr.ls. GZMA was significantly incr.ased 28 days after.vaccination while Pr.1 was r.duced pr.vaccination but incr.ased 14 days post-vaccination in the CFS/ME patients. Ther. wer. no significant changes in cytokine genes pr. or.post vaccination. Administr.tion of TIV may incr.ase the expr.ssion of lytic genes in CFS/ME and this may contr.bute to the incr.ase in cytotoxic activity we obser.ed in these patients post vaccination.
