The gastrointestinal(GI)microbiome in fish plays significant roles in health and disease *** investigation was accomplished to enumerate,characterize and identify the potential probiotic bacteria from three Indian Major Carps(IMCs),viz.,rohu(Labeo rohita),catla(Catla catla)and mrigal(Cirrhinus mrigala)using culture dependent ***,105 pathogen-inhibitory bacteria(out of 1216 isolates)were detected from three IMCs by double layer assay.16S rRNA partial gene sequence analyses and BLAST search in the NCBI GenBank unveiled that 94.29%of the pathogen inhibitory bacteria were bacilli(99 strains)and Bacillus licheniformis by far the most common(28%).The primarily selected 27 pathogen-inhibitory strains(cumulative inhibition score≥13)produced extracellular enzymes,while 15 of them produced all the six exo-enzymes studied(amylase,protease,lipase,cellulase,phytase and xylanase).Gut stability of the strains became apparent by their ability to grow in fish mucus and tolerance to diluted ***,14 strains were noticed asγ-hemolytic and susceptible to the commonly used ***,intra-peritoneal injection withγ-hemolytic strains did not induce any pathological signs or mortalities in fish,and thus were considered as *** 14γ-hemolytic isolates were represented by the genus Bacillus(13)and Stenotrophomonas(1),which might form probiotic consortia for prospective use in carp culture.

Bed roughness and grain sorting-an experimental study over fine to medium sand beds

The relation between grain-size distribution of the bed and in suspension was critically examined under a uniform flow velocity of 50 cm/s over two beds: one of mainly fine sands and the other of medium sands. Two sections-one 2.85 m downstream and the other 6.35 m downstream in the experimental channel-were selected for sampling to study the grain-sorting pattern in the vertical direction along the direction of transport. The shape and type of the grain-size distribution pattern were critically studied with height above the bed. The change in the distribution pattern has been attributed to the change of local bed roughness causing scouring against the protruded relatively coarse grains on the bed. Such trends are important to predict the nature of river bed topography. The sand of Bed-1 initially exhibits a log-skew-Laplace distribution at different heights of suspension. The distribution pattern, however,changes but this changing pattern is not consistent along the upstream side. For Bed-2, which initially exhibits a log-normal distribution, the same pattern persists from the height of suspension at 5 cm up to20 cm. Such consistency in log-normality is also observed at the downstream points of measurement. It is generally expected that the mean grain-size would reduce with increases of suspension height but the results of the experiments, in some occasions, differ significantly from the gradual fining upward *** result has been attributed to local changes of bed roughness arising from the protruded relatively coarse grains causing eddies, scouring, and turbulent phenomena which moves coarse particles higher in suspension adding a coarse tail to the distribution increasing the mean grain-size.

Surface chemistry of carbon nanoparticles functionally select their uptake in various stages of cancer cells

Relationship of the surface physicochemical characteristics of nanoparticles with their interactions with biological entities may provide critical information for nanomedicinal application. Here, we report the systematic synthesis of sub-50 nm carbon nanoparticles （CNP） presenting neutral, anionic, and cationic surface functionalities. A subset of CNPs with -10, 20, and 40 nm hydrodynamic sizes were synthesized with neutral surface headgroups. For the first time, the cellular internalization of these CNPs was systematically quantified in various stages of breast cancer cells （early, late, and metastatic）, thereby providing a parametric assessment of charge and size effects. Distinct activities were observed when these systems interacted with cancer cells in various stages. Our results indicated that metastatic breast cancer could be targeted by a nanosystem presenting anionic phosphate groups. On the contrary, for patients in late stage of cancer, drugs could be delivered with sulfonate functionalized carbon nano- particles, which have higher probability of intracellular transport. This study will facilitate the better understanding of nanoparticle-biological entity interaction, and the integration of this knowledge with pathophysiology would promote the engineering of nanomedicine with superior likelihoods of crossing the endocytic ＂barrier＂ for drug delivery inside cancerous cells.

An anisotropic propagation technique for synthesizing hyperbranched polyvillic gold nanoparticles

Of late, many synthesis processes have been studied to develop irregular nano-rnorphologies of gold nanostructures for biomedical applications in order to increase the efficacy of nanoparticle theranostics, tune the plasmonic absorbance spectra, and increase the sensitivity of biomolecule detection through surface enhanced Raman spectroscopy. Here we report, a novel, non-seed mediated versatile single pot synthesis method capable of producing hyperbranched gold ＂nano-polyvilli＂ with more than 50-90 branching nanowires propagating from a single origin within each structure. The technique was capable of achieving precise tuning of the branch propagation where the branching could be controlled by varying the duration of incubation, temperature, and hydrogen ion concentration.

Liquid chromatography-tandem mass spectrometry method for the estimation of adefovir in human plasma:Application to a pharmacokinetic study

An analytical method based on solid phase extraction was developed and validated for analysis of adefovir in human plasma. Adefovir-d4 was used as an internal standard and Synergi MAX RP80A （150rnmx4.6mm. 41am） column provided the desired chromatographic separation of compounds followed by detection with mass spectrometry. The method used simple isocratic chromato- graphic condition and mass spectrometric detection in the positive ionization mode. The calibration cuives were linear over the range of 0.50-42.47 ng/mL with the lower limit of quantitation validated at 0.50 ng/mL. Matrix effect was assessed by post-column infusion experiment to monitor phospholipids and post- extraction addition experiment was performed. The degree of matrix effect for adefovir was determined as 7.5% and ion-enhancement in five different lots of human plasma was 7.1% and had no impact on study samples analysis with 4.5 rain run time. The intra- and inter-day precision values were within 7.7% and 7.8%, respectively, for adefovir at the lower limit of quantification level. Validated bioanalytical method was successfully applied to clinical sample analysis.

Optimal timing of same-admission orthotopic heart transplantation after left ventricular assist device implantation

AIM To investigate the impact of timing of same-admission orthotopic heart transplant(OHT) after left ventricular assist device(LVAD) implantation on in-hospital mortality and post-transplant length of *** Using data from the Nationwide Inpatient Sample from 1998 to 2011, we identified patients 18 years of age or older who underwent implantation of a LVAD and for whom the procedure date was available. We calculated in-hospital mortality for those patients who underwent OHT during the same admission as a function of time from LVAD to OHT, adjusting for age, sex, race, household income, and number of comorbid diagnoses. Finally, we analyzed the effect of time to OHT after LVAD implantation on the length of hospital stay *** Two thousand and two hundred patients underwent implantation of a LVAD in this cohort. One hundred and sixty-four(7.5%) patients also underwent OHT duringthe same admission, which occurred on average 32 d(IQR 7.75-66 d) after LVAD implantation. Of patients who underwent OHT, patients who underwent transplantation within 7 d of LVAD implantation("early") experienced increased in-hospital mortality(26.8% vs 12.2%, P = 0.0483) compared to patients who underwent transplant after 8 d("late"). There was no statistically significant difference in age, sex, race, household income, or number of comorbid diagnoses between the early and late groups. Post-transplant length of stay after LVAD implantation was also not significantly different between patients who underwent early vs late OHT. CONCLUSION In this cohort of patients who received LVADs, the rate of in-hospital mortality after OHT was lower for patients who underwent late OHT(at least 8 d after LVAD implantation) compared to patients who underwent early OHT. Delayed timing of OHT after LVAD implantation did not correlate with longer hospital stays post-transplant.

Imaging sentinel lymph nodes （SLN） could provide us with critical information about the progression of a cancerous disease. Real-time high-resolution intrao- perative photoacoustic imaging （PAI） in conjunction with a near-infrared （NIR） probe may offer opportunities for the immediate imaging for direct identification and resection of SLN or collecting tissue samples. In this work a commercially amenable synthetic methodology is revealed for fabricating luminescent carbon nanoparticles with rapid clearance properties. A one-pot ＂green＂ technique is pursued, which involved rapid surface passivation of carbon nanopartides with organic macromolecules （e.g., polysorbate, polyethyleneglycol） in solvent-free conditions. Interestingly, the naked carbon nanoparticles are derived for the first time, from commercial food grade honey. Surface coated particles are markedly smaller （-7 nm） than previously explored particles （gold, single-walled carbon nanotubes, copper） for SLN imaging. The results indicate an exceptionally rapid signal enhancement （-2 min） of the SLN. Owing to their strong optical absorption in the NIR region, tiny size and rapid lymphatic transport, this platform offers great potential for faster resection of SLN and may lower complications caused in axillary investigation by mismarking with dyes or low-resolution imaging techniques.

Simultaneous ramp right heart catheterization and echocardiography in a Reliant Heart left ventricular assist device

Many clinicians caring for patients with continuous flow left ventricular assist devices(CF-LVAD) use ramp right heart catheterization(RHC) studies to optimize pump speed and also to troubleshoot CF-LVAD malfunction. An investigational device,the Reliant Heart Heart Assist 5(Houston,TX),provides the added benefit of an ultrasonic flow probe on the outflow graft that directly measures flow through the CF-LVAD. We performed a simultaneous ramp RHC and echocardiogram on a patient who received the above CF-LVAD to optimize pump parameters and investigate elevated flow through the CF-LVAD as measured by the flow probe. We found that the patient's hemodynamics were optimized at their baseline pump speed,and that the measured cardiac output via the Fick principle was lower than that measured by the flow probe. Right heart catheterization may be useful to investigate discrepancies between flow measured by a CF-LVAD and a patient's clinical presentation,particularly in investigational devices where little clinical experience exists. More data is needed to elucidate the correlation between the flow measured by an ultrasonic probe and cardiac output as measured by RHC.
