Phytochemical analysis of Berberis lyceum methanolic extract and its antiviral activity through the restoration of MAPK signaling pathway modulated by HCV NS5A

Objective:To evaluate the antiviral activity and phytochemicals of selected plant extracts and their effect on the mitogen-activated protein kinase(MAPK)signaling pathway modulated by hepatitis C virus(HCV)nonstructural protein 5 A(NS5A).Methods:A total of ten plant extracts were initially screened for their toxicities against Hep G2 *** non-toxic plants were tested for their inhibitory effect on the expression of HCV NS5A at both m RNA and protein levels using real-time PCR and Western blotting assays,*** differential expression of the genes associated with MAPK pathway in the presence of NS5A gene and plant extract was measured through real-time ***,the identification of secondary metabolites was carried out by phytochemical and HPLC ***:The phytochemical profiling of Berberis lyceum revealed the presence of alkaloids,phenols,saponins,tannins,flavonoids,carbohydrates,terpenoids,steroids,and ***,quercetin,myricetin,gallic acid,caffeic acid,and ferulic acid were identified through HPLC *** methanolic extract of Berberis lyceum strongly inhibited HCV RNA replication with an IC50 of 11.44μg/m ***-PCR and Western blotting assays showed that the extract reduced the expression of HCV NS5A in a dosedependent *** lyceum extract also attenuated NS5A-induced dysregulation of the MAPK signaling ***:Our findings suggest that Berberis lyceum extract strongly inhibits HCV propagation by reducing HCV NS5A-induced perturbation of MAPK signaling.

Objective:To evaluate the effects of three oxidized palm oil diets(OPD)on female rat reproductive ***:Forty-four female Wistar rats presenting five consecutive and regular estrous cycles were divided into 4 *** rats were fed with:a standard diet,70%of standard diet+30%oxidized palm oil diet(OPD1),OPD1+5 g of boiled yolk egg(OPD2)and OPD1+10%sucrose(OPD3)for 125 days,*** the feeding period,morphometric,estrous cycle,sexual behavior,gestation,biochemical and histomorphometric parameters were ***:All OPDs significantly increased abdominal circumference,body mass index and Lee index coupled to an irregularity and lengthening of the estrous *** significantly decreased appetite and consumption behaviours,quantic pregnancy index,fertility rate,implantation sites and index,serum progesterone and high-density lipoprotein levels,increased pre-implantation losses,anti-implantation activities,serum estradiol,triglycerides,total and low-density lipoprotein-cholesterol levels,and impaired brain and ovaries oxidative *** examinations revealed increases in the number of atresic and primary follicles and decreases in secondary,tertiary,Degraaf,total and corpus luteum follicles in ovaries coupled to a neurodegeneration of hypothalamic anteroventral periventricular neurons in the OPD groups compared to the standard diet ***:The three OPDs induce obesity and impair the female reproductive function,especially OPD2 and *** findings contribute to a better understanding of the adverse effects of palm oil bleaching on the reproductive function in female rats,which could be useful in the management of women with obesity-related sexual dysfunction.

Acute and Reproductive Toxicity of the Aqueous Extract of the Dry Seeds of Aframomum daniellii on the Female Wistar Rat Strain

This work was designed to investigate the acute and reproductive toxicity activity of the aqueous extract of the dry seeds of Aframomum daniellii on the female rats. The acute toxicity of the aqueous extract of Aframomum daniellii (A. daniellii) was evaluated with 6 female rats which were divided into 2 groups (1 Test group and the Control group) of 3 female rats each. The control group received distilled water (10 mL/kg/po) and the test group received a single dose of extract of A. daniellii at the dose of 2000 mg/kg. The reproductive toxicity was evaluated using 45 adult female rats which were divided into 5 groups. Group I, received distilled water (1 mL/100 g/po, neutral control);group II, received Clomiphene citrate (600 μg/kg/po, positive control);Groups III, IV and V (trials) received aqueous extract of A. daniellii at doses of 100, 200 and 400 mg/kg/po respectively. The animals were treated daily for 14 days. From the 6th day of treatment, the rats were mated with males of proven fertility for 8 days. On day 22, after laparotomy and delivery, the number of implantation sites, corpora lutea, resorption sites and pups were recorded. Concerning the acute toxicity, it was observed that, after the single dose of 2000 mg/kg administration of the aqueous extract of the dry seeds of A. daniellii, no deaths were recorded. Concerning the reproductive toxicity, no implantation and gestation were observed when compared to the control. However, the aqueous extract of A. daniellii caused a significant (p A daniellii is weakly toxic, but could negatively affect some reproductive parameters.

Sexual dysfunction is a group of diseases, disorders or difficulties experienced by men or women during any stage of normal sexual activity. In Africa in general and Cameroon in particular, statistical data concerning the prevelance and the main causes of male sexual dysfunction are rare due to socio-cultural and religious burdens and sometimes self-medication. The objective of this work was to determine the prevalences, comorbidities and the risk factors of the main sexual dysfunction in some hospitals in city of Douala. Through a descriptive retrospective study carried out from Novembre 2019 to June 2021, sociodemographic parameter, reasons of consultation, risk factors and type of male sexual dysfunction (erectile dysfunction or erectile dysfunction (ED), ejaculation disorders (EjD), libido disorders (LD) and disorders of sexual activity with pain or painful sexual intercourse (PSI)) were collected in the medical files of patients who have made urological consultation between 2016 and 2020 at the Deido District hospital, Laquintinie Douala Hospital and Douala General hospital respectively. At the end of our data collection, out of a total of 24995 consultations, 2743 (10.98%) patients were suffering from at least one male sexual dysfunction. Moreover, sexual disorders were the 2nd reason of urological consultation (13.69%) after urinary disorders (33.85%). Among sexual disorders, erectile (76%) and ejaculatory (20%) disorders were the predominant pathologies recorded with the prevalences of 9.79 et 2.62% respectively, and were mostly represented in patients from [41 - 50] for erectile dysfunction, and [18 - 30] years old for ejaculative dysfunction. In addition, the number of patients with ejaculatory and erectile dysfunctions was more important in patients from [18 - 30] and [31 - 40] years old respectively. In recent years, the rate of infertility has not stopped increasing in the world. The aim of the present study was to evaluate the effects of the aqueous extract of Schumanniophyton magnificum (Rubiaceae) on cyclicity, ovulation and gestation in mature rats. Methods: After a qualitative phytochemical analysis of these aqueous extracts, the experimental studies carried out were based on the evaluation of the pro-fertility effects of this extract in mature rats. For this purpose, 35 rats were used for the estrous cyclicity test and treated for 21 days at the end of which vaginal smears were taken and the duration, as well as the frequency of the appearance of the phases of the cycle, were evaluated. The ovulation test was performed on 80 female rats, which were divided into two groups of 40 animals and treated respectively in the morning and evening with distilled water, β-oestradiol or plant extract at doses of 200, 400 and 800 mg/kg. At the end of estrus, the rats were sacrificed, the ovaries were removed and weighed, the hemorrhagic points counted and the blood samples were taken for hormonal studies. The last phase of the study consisted in evaluating the effects of these plant extracts on the evolution of gestation. Thus, 42 mature rats were treated during the periods from the 1st to the 10th day (1st stage), and from the 11th to the 17th day (2nd stage). At the end of these two phases, a laparotomy was performed and the number of implantation sites and corpus luteum was counted. And finally, at parturition, from the 18th to the 22nd day (3rd stage), the number of living pups was performed and the gestational parameters were calculated. Results: Administration at the beginning of prœstrus allowed a significant increase (p S. magnificum extract. The aim of the study was to bring scientific evidence of the curative action of the stem bark aqueous extract of Schumanniophyton magnificum (ASMa) on the sexual maturation and fertility of the immature female Wistar rat. Forty immature female rats were randomized and divided into 4 groups of ten animals each and orally treated with the ASMa at doses of 0 (distilled water), 200 mg/kg, 400 mg/kg and 800 mg/kg /BW/day for 30 consecutive days. Body weight and food intake were recorded throughout the experimental period. The precocity of the puberty onset in treated animals was evaluated through the determination of their age at vaginal opening. At the end of the experimental period, 5 animals in each group were sacrificed and blood samples were collected for hormonal assay. Their ovaries and uteri were removed, blotted, weighted and prepared for biochemical analysis. The remaining rats (5 per group) were crossed with males of proven fertility. After laparotomy (ten days after mating) and delivery, fertility and gestational parameters were recorded. It was noticed that, body weight gain increased significantly at all doses although there was no significant difference in food intake. The sexual maturation of treated animal was reduced to 5 days when compared to control. This was associated with the simultaneous elevation of FSH and LH (p < 0.0001) at dose of 800 mg/kg and FSH alone (p mg/kg. However, the ovarian cholesterol significantly decreased (p < 0.0001) at all doses while the uterine protein significantly increased (p < 0.05;p mg/kg and 800 mg/kg. The animal treated at doses of 400 mg/kg and 800 mg/kg exhibited an early fertilization (2 - 3 days) when compared to the control one (9 - 14 days). The number of implantation site significantly increased (p < 0.05;p mg/kg and 800 mg/kg) after laparotomy as well as the number of alive fetuses after delivery and gestational rate (80% and 100%) respectively. Antifertility Effects of Aqueous Roots Extract of Alchornea cordifolia(Euphorbiaceae) on Female Albino Rats

Aqueous roots extract of Alchornea cordifolia was studied to evaluate its traditional antifertility claim. Antifertility effect of Alchornea cordifolia was evaluated according to the stages of pregnancy of female rats. In the first stage, forty female rats were randomly divided into four groups (10 rats/group) and respectively treated with 1 ml distilled water (control), 100, 200 and 400 mg/kg body weight (BW) per day of Alchornea cordifolia. After laparotomy and at parturition, sites of implantation and resorption on the uterine horns were determined. The ovaries and uterus were collected and weighed. Blood samples were collected for hormonal assay. In the second and third stage of pregnancy, treatment schedule and antifertility parameters were assessed as above for the first stage. Treated groups received daily 400 mg/kg bw of extract of Alchornea cordifolia while Control group received distilled water. It was observed in the first stage of pregnancy that, the number of resorption sites significantly (p < 0.05) decreased at 100 and 200 mg/kg. No implantation site was observed at dose 400 mg/kg after laparotomy as well as the number of pups at parturition at all doses. The body weight of ovaries and uterine horns at dose 400 mg/kg significantly (p < 0.05) decreased when compared to the control. The extract dose dependently decreased significantly the concentrations of serum progesterone. In the second stage of pregnancy after laparotomy and at parturition, the number of live fetus and the survival ratio significantly (p < 0.001) decreased and were associated with the vaginal discharge and bleeding of animals. In the third stage of pregnancy, the extract caused an early parturition associated with a decreased number of live fetus and the survival ratio with p The results indicate that aqueous roots extract of Alchornea cordifolia could induce inhibitory effects on reproductive functions in female albino rats.
