将萜烯类吲哚生物碱代谢关键酶———色氨酸脱羧酶 (TDC)的编码基因转到烟草 (NicotianatabacumL .)植物体内 ,标定在不同的亚细胞区室表达。通过蛋白免疫印迹法和色胺在植物体内的累积量测定分析 ,对转基因植物进行筛选。结果表明 ,TDC在叶绿体和胞液中高效表达 ,TDC在叶绿体中的表达水平最高 ,高于在胞液中的表达 ,在内质网和液泡中表达水平很低 ,用蛋白免疫印迹法未检出。

A number of solid tumors contain a distinct subpopulation of cells, termed cancer stem cells (CSCs) which represent the source for tissue renewal and hold malignant potential and which would be responsible for therapy resistance. Today, the winning goal in cancer research would be to find drugs to kill both cancer cells and cancer stem cells, while sparing normal cells. Osteosarcoma is an aggressive pediatric tumor of growing bones that, despite surgery and chemotherapy, is prone to relapse. We have recently selected from human osteosarcoma MG63 cells a cancer stem-like cell line (3AB-OS), which has unlimited proliferative potential, high levels of stemness-related markers, and in vivo tumorforming capacity in xenograft assays. Here, we have shown that 3AB-OS cells can differentiate in vitro into endoderm-, mesoderm-and ectoderm-derived lineages. Cell differentiation is morphological, molecular and functional. We propose that this model system of 3AB-OS differentiation in vitro might have a number of useful purposes, among which the study of molecular mechanisms of osteosarcoma origin, and the analysis of factors involved in specification of the various cell lineages. We still do not know either what are the shared and distinguishing characters between CSCs and normal stem cells, or what is the reason why the cancer stem cells, like the normal stem cells, have the ability to differentiate toward the derivatives of the primary germ layers. It is possible that each of the differentiation capability may be exploited by CSCs to supply their needs of growing and surviving in hostile microenvironment.

Introduction: D-Aspartate is an endogenous amino acid involved in LH and testosterone release in humans. In this study we investigate the impact of nutritional supplementation of sodium D-aspartate on the improvement of sperm quality in sub-fertile patients and the rate of pregnancies that occurred with their partners. Materials and Methods: A group of 30 patients affected by oligo-asthenozoospermia and a group of 30 patients affected by asthenozoospermia were treated with a daily dose of sodium D-aspartate for 90 days. After which, the change in spermatozoa concentration and their motility and the pregnancies that occurred with their partners were recorded. Results: We found that the supplementation of D-aspartate significantly increased the concentration and the motility of spermatozoa. In oligo- asthenozoospermic patients the increase of sperm concentration was found to be 2.0-fold, P Conclusions:Treatment of sub-fertile patients with sodium D-aspartate improved the number and the motility of the spermatozoa and consequently improved the rate of pregnancies of their partners.

Background and study aims:Celiac disease can manifest with nonspecific symptoms,including functional gastrointestinal disorders such as *** aim of our study was to assess the usefulness of duodenal endoscopic markers of villous atrophy for the selection of dyspeptic patients for histological *** and methods:Esophagogas troduodenoscopy was performed in dyspeptic patients,in patients considered to be at risk of having celiac disease,and in healthy *** least three duodenal biopsies were performed for histological assessment of villous atrophy in all patients and *** looked for the following four duodenal endoscopic markers of celiac disease:reduction in the number of folds,scalloping of folds,mosaic-pattern mucosa,and nodular ***:A total of 175 people were enrolled(75 patients with dyspepsia;75 patients who were “ at risk” of having celiac disease;and 25 healthy volunteers,or “ controls”) .Of the dyspeptic patients,four had endoscopic markers of celiac disease with no histologically confirmed villous atrophy,while one patient without endoscopic markers was found to have Marsh type I villous *** the patients at risk of having celiac disease,16 had at least one endoscopic marker and 10/16 were found to have histological villous *** this group,the sensitivity and specificity of the endoscopic markers were 100% and 90.8% respectively.“ At-risk” patients with two or more endoscopic markers all had histologically confirmed villous *** endoscopic markers nor villous atrophy were found in any of the control ***:Additional endoscopic markers are valuable for diagnosis in patients with clinical symptoms suggestive of celiac *** contrast,endoscopic markers of villous atrophy are not useful for selecting a subgroup of dyspeptic patients for screening for celiac disease by duodenal histological *** patients should be screened using other protocols.

Goals: To assess epidemiologic features and predictive factors of mortality of acute upper gastrointestinal bleeding (UGIB). Study: During a 6-month period, a prospective population based study including all the UGB occurring in a geographic area of 3 million people was conducted. Data from cirrhotic patients were compared with those of noncirrhotic patients. Results: A total of 2,133 UGIB were recorded, 21.9%in cirrhotic patients (n = 468). Endoscopic hemostasis was performed in 46.5%and 8.3%in cirrhotic and noncirrhotic patients, respectively (P dictive factors of mortality were observed in both patient groups: a prothrombin level less than 40%, an UGIB occurring in inpatients, a concomitant digestive carcinoma, a hematemesis revealing the UGIB, a recent use of steroid drugs, and age over 60 years. Four other predictive factors of mortality were also identified in noncirrhotic patients. Conclusions: Although epidemiologic features, clinical course, management, and prognosis of UGIB were quite different in cirrhotic and noncirrhotic patients, the majority of predictive factors of mortality were the same in both patient groups. These data underline the major role of debilitated status and hepatic failure in the prognosis of UGIB in cirrhotic patients.
