Flow Cytometric Analysis of the Toxicity of Nitrofen in Cultured Keratinocytes

Lactate dehydrogenase (LDH) release test, 3 H-thymidine (3 H-TdR) and 3 H-leucine (3 H-Leu) incoopration tests and flow cytometric analysis (FCM) of cell cycle were empoyed to elucidate cellular and molecular mechanism of nitrofen-induced toxicity in cultured *** results showed that cell morphologic damages were observed after exposure to 1.0 mmol/L and 10.0 mmol/L nitrofen. LDH release increased in a dose- and time-dependent manner. Depressions in 3H -TdR and 3 H-Leu incorpration were found even at 0.01 mmol/L, and increased with the exposure dose. Cell cycle was analyzed from the DNA- histogram with propidium iodde stain. The results showed that there was no pronounced alteration in cell cycle after cells exposed to 0.01 and 0.1 mmol/L nitrofen. At dose of 1.0 mmol/L, S phase cells increased 2 times of that of control. With the increase of dose, G2/M phase cells became to increase about 5 times of that of the control. At 1 .0 mmol/L, time course of cell cycle after exposure was observed. At the beginning of exposure, cells in S phase and G2/M phase were about 8 .7 % and 11 %. Following 24 h incubation with nitrofen, cells in S phase increased to 18.0% with almost no change in G2/M. 72 h after exposure, G2/M phase cells increased to 63 .3%. The forve results demonstrated that S phase and G2/M phase blockage in cultured keratinocytes after exposed to nitrofen seems of importance in the mechanism of nitrofen-induced toxicity.

SOME FACTORS AFFECTING [3H]TYROSINE BINDING TO PLASMA MEMBRANE PREPARATION OF RAT CORPORA LUTEA in vitro

Temperature, pH, enzymes and ions affected the two types of [H]tyrosine binding sites differently. The optimal temperature and pH for the high affinity sites were 24 C and 7.4, and for the low affinity sites 37℃ and 7.0, respectively. The [H]tyrosine binding to the high and low affinity sites was not observed at 55℃, nor at pH 6.0 or pH 8.5. Trypsin reduced [H]tyrosine binding to the high affinity sites, but had no effect on the binding to the low affinity sites. Neuraminidase reduced [H]tyrosine binding to the low affinity sites, but had no effect on the binding to the high affinity sites. None of cycloheximide, kCG, cAMP, and progesterone affected [H]tyrosine binding.

In our present study we build up a new experimental model in the rat forepilepsy and examine the effects of electroacupuncture on epileptiform hippo-campal unit activity. Experiments were carried out on conscious immobilized male rats, CoriariaLactone at a concentration of 30μg/10μl in saline was injected intracerebrove-ntricularly. Electrocorticography (ECoG) and hippocampal unit activity wererecorded simultaneously on the side of brain which received the drug *** some cases, the hippocampal EEG was also recorded. Acupuncture of Da-zhui, Shenzhu, Taichong and Kunlun were stumulated respectively.

Down-regulation of C-type natriuretic peptide receptor by vasonatrin peptide in cardiac myocytes and fibroblasts

AIM: To investigate the regulatory effects of vasonatrin peptide (VNP) on the expression of C-type natriuretic peptide receptor (NPR-C) in cultured neonatal rat cardiac myocytes and fibroblasts. METHODS: Quantitative RT- PCR was undertaken to evaluate the levels of NPR-C mRNA and radioimmunoassay was used to determine the formation of intracellular cGMP. RESULTS: Twenty-four hours hypoxic exposure increased the level of NPR-C mRNA in cardiomyocytes, while did not alter the expression of NPR-C in cardiac fibroblasts. VNP (1×10-8-1×10-6 mol/L) reduced the levels of NPR-C mRNA in cardiac myocytes induced by hypoxia in a concentration-dependent manner, and with high concentration (1×10-6mol/L) also decreased the expression of NPR-C in cardiac fibroblasts and air-control cardiac myocytes. The inhibitory effects of VNP on the expression of NPR-C was mimicked by 8- bromo-cGMP 1×10-6 mol/L (a membrane permeable analog of cGMP). VNP (1×10-8-1×10-6mol/L) increased the formation of intracellular guanosine-3',5'-cyclic monophosphate (cGMP) in both cardiac myocytes and fibroblasts. HS-142-1, the particulate guanylyl cyclase-coupled receptor antagonist, partially abrogated the above effects of VNP. CONCLUSION: Hypoxic exposure for 24 h up-regulated the expression of NPR-C in cultured neonatal rat cardiac myocytes. VNP decreased the expression of NPR-C in cardiac myocytes and fibroblasts under both air- control and hypoxic condition, which was at least partially mediated by guanylate cyclase linked natriuretic peptide receptors through increasing the intracellular cGMP.

Bicoid is one of the important Drosophila maternal genes involved in the control of embryo polarity and larvae segmentation. To clone and characterize the rice bicoid-related genes, one cDNA clone, Rb24 (EMBL accession number: AJ2771380), was isolated by screening of rice unmature seed cDNA library. Sequence analysis indicates that Rb24 contains a putative amino acid sequence, which is homologous to unique 8 amino acids sequence within Drosophila bicoid homeodomain (50% identity, 75% similarity) and involves a lys-9 in putative helix 3. Northern blot analysis of rice RNA has shown that this sequence is expressed in a tissue-specific manner. The transcript was detected strongly in young panicles, but less in young leaves and roots. This results are further confirmed with paraffin section in situ hybridization. The signal is intensive in rice globular embryo and located at the apical tip of the embryo, then, along with the development of embryo, the signal is getting reduced and transfers into both sides of embryo. The existence of bicoid-related sequence in rice embryo and the similarity of polar distribution of bicoid and Rb24 mRNA in early embryo development may implicates a conserved maternal regulation mechanism of body axis presents in Drosophila and in rice.

Experiments were performed on cats under the treatments of chloraloseand flexedil. The cortical evoked potentials (C-CEP) recorded in primarysomatosensory cortex(SI), which were elicited by C-fiber inputs of sapheneousnerve, were used as an index of slow pain. The effect of electro-acupuncture(EA) of "Zusanli" point on C-CEP and role of cerebral anterior laterialassociation area(ALA) in this effect were observed.

In this artical the effect of stimulating PAG on C-CEP was observed in order to provide some data for the research of slow pain modulation. Experiments were performed on cats under choloralose anaesthesis andimmobitized by gallamine triethiodide. After blocking A-fibers input by anodalcurrent the superficial peroneal nerve was stimulated with strong electricalpulse, and the C-fiber input was elicited selectively. The evoked potentialswere recorded on the back surface of ipsilateral spinal cord (C-SSP) andcontralateral cerebral somatosensory area (C-CEP). PAG was stimulated withshort electrial pulse.

In orderto solve this question, we applied capsaicin topically on sciatic nerve and usedthe techniques of extracellular recording and nerve trunk recording.
