Aim:Rhynchophylline(Rhy) is the major tetracyclic oxindole alkaloid isolated from Uncaria species, and as the important active component in the Gambirplant(Gouteng) which modulates many physiological functions including cardiovascular ***,the mechanism of Rhy’s actions is *** we report some novel findings about the action of Rhy on smooth muscle cells(SMCs) freshly isolated from human mesenteric ***:The tension changes of mesenteric artery were recorded by Chart5.0 software via Powerlab *** freshly isolated smooth muscle cells were used to record K currents by a whole-cell, amphotericin-perforated configuration of patch-clamp ***:*** BK macroscopic currents and spontaneous transient outward currents(STOCs,transient BK currents) in human mesenteric artery SMCs were voltage-dependent and extracellular *** superimposed stochastically onto macroscopic BK currents,and both of them were significantly inhibited by iberiotoxin(IbTX,200 nmol/L),a specific BK ***(4 mmol/L) activated ryanodine receptor(RyR) to produce a remarkable enhancement in both forms of BK *** inhibitor ryanodine(10μmol/L) significantly inhibited BK macroscopic currents, while abolished STOCs in human mesenteric artery ***(20-100μmol/L) significantly suppressed the two forms of BK currents in a dose-dependent manner(n=6),and the IC50(fitted by Boltzmann function) was(39.06±0.27)μmol/*** the concentration of 60μmol/L(the most potent concentration),Rhy significantly decreased STOCs amplitude by 39.16%±9.10%,area by 54.2%±10.21%,half-width by 40.40%±9.43%and frequency by 66.08%±7.01%,respectively(n=6),while it had little effect on STOCs *** macroscopic currents were also remarkably suppressed by 32.80%±9.34%at the membrane potential of +20 mV(n=7).RyR activator caffeine(4 mmol/L) failed to reactivate both forms of BK currents after Rhy treatment(n=4).*** addition,Rhy re

INTRODUCTIONThe incidence of primary non-function(PNF)of grafted liver in the early postoperative stage is 2%-23%[1-4],its main cause is the ischemic-rechemic injure[5,6].In this experiment,anisodamine was added into the preserving fluid and the grafted liver was rewarmed at different temperatures to protect the cell membranc and prevent ischemic-reperfusive injury.

Effects of Ginkgolide B on action potential and calcium, potassium current in guinea pig ventricular myocytes

AIM: To investigate the effect of Ginkgolide B (GB) on action potential (AP) , delayed rectifier potassium current (IK), and L-type calcium current (ICa-L) in guinea pig ventricular myocytes. METHODS: Single ventricular myocytes were isolated by an enzymatic dissociation method. AP, IK, ICa-L were recorded by whole-cell patch-clamp tech- nique in either current or voltage clamp mode. RESULTS: GB shortened APD in a concentration-dependent manner. GB 0.1, 1, and 10 μmol/L shortened APD50 by 7.9 % (n=5, P>0.05), 18.4 % (n=5, P05), 17.6 % (n=5, P<0.01), 26.4 % (n=5, P<0.01), respectively. GB increased IK in a concentration-dependent manner. GB 0.1, 1, and 10 μmol/L increased IK by 20.1 % (n=6, P<0.05), 43.1 % (n=6, P<0.01), 55.6 % (n=6 , P<0.05); increased IK tail by 10.7 % (n=6, P<0.05), 25.1 % (n=6, P<0.05), and 37.7 % (n=6, P<0.05), respectively at testing potential of +50 mV and shift the I-V curve of Ik upward. But GB had no significant effect on ICa-L at above concentrations. CONCLUSION: GB significantly shortened APD in a concentration-dependent manner which mainly due to increase of IK.

Sequential-release of anticancer drugs microcapsulated with ethylcellulose

Objective To approach the sequential release of antitumor drugs and promote the effect of *** Adriamycin (ADM) and carboplatin (CBP) were respectively microcapsulated with ethylcellulose by organic phase separation. The morphology and sizes of the microcapsules were observed and measured with light microscope and scanning electromicroscope. The contents and the release rates of ADM and CBP in microcapsules were measured with fluorescence spectrophotometer and high-efficiency phantom chromatic (HPC) spectrum respectively. The antitumor sensitivity test in vitro was devised with MTT *** The microcapsules of ADM and CBP were spherical in shape with diameters of 196±64 μm and 214±48 μm respectively. The contents of one-layer and two-layer CBP and ADM microcapsules were 51.4%, 35.7% and 39.8% respectively, with the release rates in vitro of 62.4%/day, 54.8%/day and 48.2% /8h. The results of drug sensitivity test in vitro demonstrated that the current preparation has never affected the stability and antitumor activity of CBP and *** Microcapsules with different drugs and different thickness of material have different release rate. Combined arterial chemoembolization with different microcapsules could approach the sequential release and promote the effect of chemotherapy.

OBJECTIVE: To explain the association between vascular dementia and the cranial MRI manifestations, and recognize the value of cranial MRI in the early diagnosis of vascular dementia and the assessment of disease conditions. DATA SOURCES: Pubmed database was searched to identify articles about the cranial MRI manifestations of patients with vascular dementia published in English from January 1992 to June 2006 by using the of 'MRI, vascular dementia'. Others were collected by searching the name of journals and title of articles in the chinese full-text journal database. STUDY SELECTION: The collected articles were primarily checked, those correlated with the cranial MRI manifestations of patients with vascular dementia were selected, while the obviously irrelative ones were excluded, and the rest were retrieved manually, the full-texts were searched. DATA EXTRACTION: Totally 255 articles were collected, 41 of them were involved, and the other 214 were excluded. DATA SYNTHESIS: MRI can be taken as one of the effective methods for the early diagnosis and disease evaluation of vascular dementia. White matter lesions are the important risk factors of vascular dementia. Vascular dementia is accompanied by the atrophy of related brain sites, but further confirmation is needed to investigate whether there is significant difference. MRI can be used to quantitatively investigate the infarcted sites and sizes of patients with vascular dementia after infarction, but there is still lack of systematic investigation on the association of the infarcted sites and sizes with the cognitive function of patients with vascular dementia. CONCLUSION: Cranial MRI can detect the symptoms of vascular dementia at early period, so that corresponding measures can be adopted to prevent and treat vascular dementia in time.

Clinical manifestations and cerebral angiographic findings of moyamoya disease

Objective To study the clinical features and angiographic findings of moyamoya disease (MMD) as well as their relationship. Methods A total of 22 MMD patients received routine digital substraction angiography (DSA). The clinical manifestations and angiographic findings were analyzed. Results Clinical manifestations varied and each patient often had multiple symptoms,including cerebral infarction in 9 patients with an average age of 23.6 (13-39 years) and cerebral hemorrhage in 7 patients with an average age of 31.2 (28-46 years). Angiographic examination found that all the diseased sides showed MMD blood vessels. The patients who received encephalo-myo-arterio-synangiosis (EMAS) had better prognosis than those without receiving the treatment. Conclusion Cerebral infarction is frequent in children and adolescents with MMD,whereas cerebral hemorrhage is common in adults. DSA is a golden criterion for diagnosing MMD. Surgical treatment,EMAS blood supply reconstruction in particular,should be prescribed.

Histological changes following surgically-assisted rapid tooth movement through resistance reduction and distraction osteogenesis in dogs

Objective To investigate the histological changes of rapid tooth movement in dogs treated by resistance reduction and distraction osteogenesis,aiming to establish an animal model and further to reveal the remodeling mechanism of rapid tooth movement. Methods A total of 8 local hybrid dogs were selected as subjects for this study. The second pre-molar was extracted on both sides. The experimental side underwent alvelor surgery for resistance reduction and a home-made tooth-borne intraoral distraction device was installed for rapid tooth movement,while for the other side (control side) only tooth-borne intraoral distraction device was used for rapid tooth movement. The longest active force-delivery span was 2 weeks,followed by 6-week retention. The distance between the moved tooth and anchor unit was recorded weekly,and radiography was performed for each side before and after distraction. The surrounding tissues including periodontal ligament and alveolar bone were sectioned for histological analysis. Results The average distance of tooth movement was 3.55mm on the experimental side and 1.11mm on the control side. The rate of tooth movement was notably higher (P<0.01) and no significant apical root resorption was detected by X-ray on the experimental side. The active alvelor bone remodeling was found on the tension and pressure sides. However,there was no significant difference between the experimental side and the control side after the retention period. Conclusion The rate of orthodontic tooth movement can be accelerated through resistance reduction and periodontal distraction without any unfavorable effects but at minimal anchorage loss.
