Effects of Chuanxiongqin hydrochloride on increasing the fluidity of brain cell membrane and scavenging free radicals in model rats with ischemia/reperfusion injury

BACKGROUND: The fluidity of cell membrane can be affected by various factors. Many experiments have confirmed that the ischemia/reperfusion of organic tissue can increase the contents of free radicals, which lead to high rigidity and low fluidity of cell membrane, and the conditions can be changed by Chuanxiongqin. OBJECTIVE: To observe the effect and mechanism of Chuanxiongqin hydrochloride on the fluidity of brain cell membrane in rat models of ischemia/reperfusion. DESIGN: A completely randomized controlled animal trial. SETTINGS: Institute of Brain Sciences; department of physiology, medical college, datong university. MATERIALS: Twenty male grade Ⅰ Wistar rats of 170-220 g were randomly divided into model group (n =10) and control group (n =10). Chuanxiongqin hydrochloride (molecular mass was 172.2) was purchased from the National Institute for the Control of Pharmaceutical and Biological Products (batch number: 0817-9803); Spin labelers: 5-doxyl-stearlic acid methylester (5DS), 16-doxyl-stearlic acid methylester (16DS), xanthine, xanthine oxidase (XOD) and 5,5-dimeth-1-pyrroline- N-oxide (DMPO) from Sigma Company; Bruker ESP 300 electron paramagnetic resonance (EPR) spectrometer by Bruker Company (Germany). METHODS: The experiments were carried out in the State Key Laboratory of Natural and Biomimetic Drugs, Peking university from June 2001 to July 2002. In the model group, rats were made into models of cerebral ischemia by 30-minute ligation and 2-hour reperfusion of common carotid arteries; The rats in the control group were not made into models. The order parameter (S) and rotational correlation time (τc) were detected with the ESR spectrometer by means of spin labeling. The greater the S and τc, the smaller the fluidity. Meanwhile, the clearance rate of free radicals was detected with ESR spin trapping. The measurement data were compared using the t test. Quantum statistical entropy corresponding to cosmic horizon in five-dimensional spacetime

The generalized uncertainty relation is introduced to calculate the quantum statis-tical entropy corresponding to cosmic horizon. By using the new equation of state density motivated by the generalized uncertainty relation, we discuss entropies of Bose field and Fermi field on the background of five-dimensional spacetime. In our calculation, we need not introduce cutoff. There is no divergent logarithmic term in the original brick-wall method. And it is obtained that the quantum statistical en-tropy corresponding to cosmic horizon is proportional to the area of the horizon. Further it is shown that the entropy corresponding to cosmic horizon is the entropy of quantum state on the surface of horizon. The black hole's entropy is the intrinsic property of the black hole. The entropy is a quantum effect. In our calculation, by using the quantum statistical method, we obtain the partition function of Bose field and Fermi field on the background of five-dimensional spacetime. We provide a way to study the quantum statistical entropy corresponding to cosmic horizon in the higher-dimensional spacetime.

Radiation spectrum of a high-dimensional rotating black hole

This study extends the classical Damour-Ruffini method and discusses Hawking radiation in a (n + 4)-dimensional rotating black hole. Under the condition that the total energy and angular momentum of spacetime are conservative, but angular momentum a = J/M of unit mass of the black hole is variable, taking into consideration the reaction of the radiation of the particle to the spacetime, a new Tortoise coordinate transformation and discuss the black hole radiation spectrum is discussed. The radiation spectrum that satisfies the unitary principle in the general case is derived.

Tests for nonparametric parts on partially linear single index models

Tests for nonparametric parts on partially linear single index models are considered in this paper. Based on the estimates obtained by the local linear method, the generalized likelihood ratio tests for the models are established. Under the null hypotheses the normalized tests follow asymptotically the χ2-distribution with the scale constants and the degrees of freedom being independent of the nuisance parameters, which is called the Wilks phenomenon. A simulated example is used to evaluate the performances of the testing procedures empirically.

The hydrolysis products of tetrahydrofolate model compound 4 were a mixture of 6 and 7 and the rearrangement reaction between 6 and 7 via 5 was in a state of equilibrium, confirmed by (HNMR)-H-1 and IR spectra.

Isoprenaline enhances local Ca2+ release in cardiac myocytes

Aim:Contraction of cardiac myocytes is controlled by the generation and ampli-fication of intracellular Ca2+*** key step of this process is the couplingbetween sarcolemma L-type Ca2+channels（LCCs）and ryanodine receptors（RyRs）in the sarcoplasmic reticulum（SR）.β-Adrenergic stimulation is an important regu-latory mechanism for this coupling *** the details underlied the globallevel,which require local Ca2+release study are still *** present study isto explore the effects of β-adrenergic stimulation on local Ca2+***:Using confocal microscopy combined with loose-seal patch-clamp approaches,effects of isoprenaline(1 μmol·L-1),a β-adrenergic agonist,on local SR Ca2+re-lease triggered by Ca2+influx through LCCs in intact rat cardiac myocytes ***:Isoprenaline increased the intensity of ensemble averagedlocal Ca2+transients,the peak of which displayed a typical bell-shaped voltage-dependence over the membrane voltages ranging from～-40mV to～+35m ***-ther analysis showed that this enhancement could be explained by the increasedcoupling fidelity（which refers the increased probability of RyRs activation upondepolarization）,and the increased amplitude of evoked Ca2+sparks(due to moreCa2+releases through local RyRs).In addition,isoprenaline decreased the firstlatency,which displayed a typical"U"-shaped voltage-dependence,showing theavailable acceleration and synchronization of β-adrenergic stimulation on intrac-ellular calcium ***:Isoprenaline enhances local Ca2+release incardiac *** results underscore the importance of regulation of β-adrenergic stimulation on local intermolecular signals between LCCs and RyRs inheart cells.

Alzheimer disease (AD),an age-related neurodegenerative disorder with progres-sive loss of memory and deterioration of comprehensive cognition,is character-ized by extracellular senile plaques of aggregated β-amyloid (Aβ),and intracellularneurofibrillary tangles that contain hyperphosphorylated tau *** showed that melatonin,an indoleamine secreted by the pineal gland,mayplay an important role in aging and AD as an antioxidant and *** decreases during aging and patients with AD have a more profoundreduction in this *** from clinical trials indicate that melatonin supple-mentation improves sleep,ameliorates sundowning,and slows down the progres-sion of cognitive impairment in Alzheimer's *** efficiently pro-tects neuronal cells from Aβ-mediated toxicity via antioxidant and anti-amyloidproperties:it not only inhibits Aβ generation,but also arrests the formation ofamyloid fibrils by a structure-dependent interaction with Aβ.Our recent studieshave demonstrated that melatonin efficiently attenuates Alzheimer-like *** the exact mechanism is still not fully understood,a direct regulatory influence of melatonin on the activities of protein kinases andprotein phosphatases is ***,melatonin also plays a role inprotecting cholinergic neurons and in ***,the neuroprotectiveeffects of melatonin and the underlying mechanisms by which it exerts its effectsare *** capacity of melatonin to prevent or ameliorate tau and Aβpathology further enhances its potential in the prevention or treatment of AD.

Effect of areca on contraction of colonic muscle strips in rats

AIM: To investigate the effects of areca on the contractile activity of isolated colonic muscle strips in rats and mechanism involved. METHODS: Each strip (LMPC, longitudinal muscle of proximal colon; CMPC, circular muscle of proximal colon; LMDC, longitudinal muscle of distal colon; CMDC, circular muscle of distal colon.) was suspended in a tissue chamber containing 5 mL Krebs solution (37 degrees C), bubbled continuously with 950 mL.L(-1) O(2) and 50 mL.L(-1) CO(2). The mean contractile amplitude (A), the resting tension (T), and the contractile frequency (F) were simultaneously recorded on recorders. RESULTS: Areca dose dependently increased the mean contractile amplitude, the resting tension of proximal and distal colonic smooth muscle strips in rats (P0.05). CONCLUSION: Areca stimulated the motility of isolated colonic smooth muscle strips in rats. The stimulation of areca might be relevant with M receptor partly.
