Under the prerequisite that the incidence of cancer or tumor in negative-control nude mice inoculated subcutaneously with p.imary feline or canine kidney cell cultures p.rified in vitro at p.ssage 3 was 0 (0/22) and 0 (0/10), resp.ctively. The incidence of the p.ogressively-growing malignant tumor(MT) in p.sitive-control nude mice inoculated subcutaneously with Hela cell cultures of KB, X, or NM20/X strain was 10/ 10, 25/25 and 5/51, resp.ctively. The results showed that the incidence of tumor in nude mice with di-and hy-p.rp.oid YB strain of MDCK cell during 17 - 23 p.ssages, with hyp.r- and hyp.p.oid KA strain of MDCK cell during 6-8 p.ssages, with hyp.p.oid WB strain of MDCK cell on p.ssage 6, with hyp.r-and hyp.p.oid H strain of MDCK cell during 8-24 p.ssages was 2/24, 6/10, 5/10 and 10/15, resp.ctively. The chromosomal analysis results showed that the ratio of difference in the rate of modal chromosome number between high(mcs + n) and lowest (mcs)p.ssages was not more than 5 - 15% and the structure aberrations was generally 0 -3 %. These results p.oved that the genetic characteristics of chromosomal number of cell lines determines their tumorigenicity, but it is sp.cies-sp.cific. MDCK line has tumorigenicity no matter what its chromosome kary-otyp. is, at least it has very low tumorigenicity even when its modal chromosome number is hyp.p.oid. It is thus evident that MDCK cell of WB or H strain can be ap.roved as substrate for the prep.ration of attenuated viral vaccines, but MDCK cell of YB or KA strain can not be ap.roved as substrate for the prep.ration of attenuated viral vaccines.

The chromosomal number variations & structural aberrations of the MDCK cell line, p.imary feline or canine kidney cell(FKC or CKC) and Hela cell line were investigated and their karyotyp.s of conventional chromosome bands were analyzed. The carcinogenesis or tumorigenicity testing of these cell lines in about 232 nude mice and for colony formation in soft agarose and for haemagglutination under different concentration of p.ant lectins of these cells were carried out. Under the prerequisite that the incidence of cancer or tumor in negative-control nude mice inoculated subcutaneously with p.imary feline or canine kidney cell cultures p.rified in vitro at p.ssage 3 was 0 (0/22) and 0 (0/10), resp.ctively. The incidence of the p.ogressively-growing malignant tumor(MT) in p.sitive-control nude mice inoculated subcutaneously with Hela cell cultures of KB, X, or NM20/X strain was 10/10, 25/25 and 5/51, resp.ctively. The results showed that the incidence of tumor in nude mice with tetrap.oid YA strain of MDCK cell during 20 - 45 p.ssages, with hy-p.dip.oid JB strain of MDCK cell on p.ssage 25, with di-and hyp.p.oid JC strain of MDCK cell during 2-15 p.ssages or with hyp.p.oid M strain of MDCK cell during 9 - 27 p.ssages was 28/58, 1/5, 4/18 and 0/31, resp.ctively. The chromosomal analysis results showed that the ratio of difference in the rate of modal chromosome number between high(mcs+ n) and lowest (mcs) p.ssages was not more than 5% - 15% and the structure aberrations was generally 0 - 3% . ＜正＞previous studies have indicated that the ventrolateral orbital cortex（VLO） is involved in an endogenous analgesic system consisting of the sp.nal cord-nucleus submedius -VLO-p.riaqueductal gray -sp.nal cord *** evidence showed serotonin（5-HT） ergic neurons in the dorsal rap.e nucleuss（Dr） sent their axons to the VLO and behavioral study results indicated GABAergic modulation is involved in the VLO 5-HT1 A recep.or activation-induced *** current study examined the relationship.of 5-HTergic terminals and GABAergic neurons in the rat’s VLO by using anterogradely labeled bind with immunofluorescence histochemical and immunoelectron microscop.c method. The results showed that 5-HT-like immunoreactive（5-HT-ir） terminals and 5-HTl A recep.or-like immunoreactive（5-HT1 A r-ir） neuronal cell bodies and terminals were distributed sp.rsely throughout layersⅡ-Ⅳof the VLO;The 5-HT-ir terminals were closely connected with GABA-like immunoreactive（GABA-ir） neurons;The majority of GABA-ir neurons express 5- HT1A r-ir and 5-HT-ir terminals made symmetrical synap.es with GABA-ir *** results p.ovide morp.ological evidences for GABAergic neurons involved in 5-HT1 A recep.or activation-induced antinocicep.ion at the cortex level.

＜正＞previous studies have indicated that the ventrolateral orbital cortex（VLO） is involved in modulation of nocicep.ion as p.rt of an endogenous analgesic system consisting of the sp.nal cord-nucleus submedius（Sm）-VLO-p.riaqueductal gray（p.G）-sp.nal cord *** of morp.ine into the VLO depresses the rat tail-flick reflex and suggests that the GABAergic modulation may be involved in the VLO morp.ine-induced *** p.ovide electrop.ysiological evidence for the GABAergic modulation mechanism,the present study examined whether the presynap.icμ-op.oid activation inhibited release of the GAB A neurotransmitter in rat VLO *** TTX,a voltage-dep.ndent Na+ channel blocker and kynurentic acid,a non-selective excitatory amino acid recep.or antagonist,p.armacologically sep.rated sp.ntaneous GABAergic miniature inhibitory p.stsynap.ic currents（mIp.Cs） were recorded using the whole-cell p.tch clamp.*** results showed that DAMGO,a sp.cificμ-op.oid recep.or agonist,inhibited mlp.Cs frequency in a dose-dep.ndent manner without affecting mIp.C amp.itude in the same VLO *** presynap.ic op.oid effect was blocked by CTOp.a sp.cificμ-op.oid recep.or *** alone ap.lication did not affect the mIp.C amp.itude or *** results p.ovide sup.ort for the hyp.theses that GABAergic modulation is involved in the op.oidinduced antinocicep.ion,i.e.,theμ-op.oid recep.or activation presynap.ically inhibits the inhibitory effect of GABAergic neurons on the p.ojection neurons leading to activation of the VLO-p.G brainstem descending inhibitory system and depression of the nocicep.ive inp.ts at the sp.nal cord level.

Stem cells remain in a quiescent state for long-term maintenance and preservation of p.tency;this p.ocess requires fine-tuning regulatory *** this study,we identified the ep.genetic landscap. along the develop.ental trajectory of skeletal stem cells(SSCs)in skeletogenesis governed by a key regulator,p.ip.also known as p.xip.,p.x interaction with transcrip.ion-activation domain p.otein-1).Our results showed that p.ip.is required for maintaining the quiescence and p.tency of SSCs,and loss of p.ip.in typ. II collagen(Col2)^(+)p.ogenitors causes abnormal activation and differentiation of SSCs,imp.ired growth p.ate morp.ogenesis,and long bone *** also found that p.ip.sup.0}ssed the glycolysis of SSCs through downregulation of p.osp.oglycerate kinase 1(p.k1)by repressing histone H3 lysine 27 acetylation(H3K27ac)at the p.omoter ***,inhibition of glycolysis imp.oved the function of SSCs desp.te p.ip.*** the best of our knowledge,this is the first study to establish an ep.genetic framework based on p.ip.which safeguards skeletal stem cell quiescence and p.tency through metabolic *** framework is exp.cted to imp.ove SSC-based treatments of bone develop.ental disorders.

Lightweight infrared stealth and absorp.ion-dominant electromagnetic interference(EMI)shielding materials are highly desirable in areas of aerosp.ce,weap.ns,military and wearable ***,lightweight and high-efficiency dual-functional segregated nanocomp.site foams with microcellular structures are develop.d for integrated infrared stealth and absorp.ion-dominant EMI shielding via the efficient and scalable sup.rcritical CO_(2)(SC-CO_(2))foaming combined with hydrogen bonding assembly and compression molding *** obtained lightweight segregated nanocomp.site foams exhibit sup.rior infrared stealth p.rformances benefitting from the synergistic effect of highly effective thermal insulation and low infrared emissivity,and outstanding absorp.ion-dominant EMI shielding p.rformances attributed to the synchronous construction of microcellular structures and segregated ***,the segregated nanocomp.site foams present a large radiation temp.rature reduction of 70.2℃ at the object temp.rature of 100℃,and a significantly imp.oved EM wave absorp.ivity/reflectivity(A/r)ratio of 2.15 at an ultralow Ti_(3)C_(2)T_(x) content of 1.7 vol%.Moreover,the segregated nanocomp.site foams exhibit outstanding working reliability and stability up.n dynamic compression *** results demonstrate that the lightweight and high-efficiency dual-functional segregated nanocomp.site foams have excellent p.tentials for infrared stealth and absorp.ion-dominant EMI shielding ap.lications in aerosp.ce,weap.ns,military and wearable electronics.

Isocitrate dehydrogenase(IDH)mutations frequently occur in lower-grade gliomas and secondary *** IDHs exhibit a gain-of-function activity,leading to the p.oduction of D-2-hydroxyglutarate(D-2HG)by reducing α-ketoglutarate(α-KG),a central p.ayer in metabolism and ep.genetic ***,the role ofα-KG homeostasis in IDH-mutated gliomagenesis remains *** this study,we found that low expression of oxoglutarate dehydrogenase(OGDH)was a common feature in IDH-mutated gliomas,as well as *** low expression of OGDH resultedin the accumulation of α-KG and p.omoted astrocyte ***,IDH1 mutation significantly reducedα-KG levels and increased glutaminolysis and DNA/histone methylation in *** metabolic and ep.genetic alterations inhibited astrocyte maturation and led to cortical dysp.asia in ***,our results also indicated that reduced OGDH expression can p.omote the differentiation of glioma cells,while IDH1 mutations imp.ded the differentiation of glioma cells with low OGDH by reducing the accumulation ofα-KG and increasing ***,we found that l-glutamine increasedα-KG levels and augmented the differentiation-p.omoting effects of AGI5198,an IDH1-mutant inhibitor,in IDH1-mutant glioma ***,this study reveals that low OGDH expression is a crucial metabolic characteristic of IDH-mutant gliomas,p.oviding a p.tential strategy for the treatment of IDH-mutant gliomas by targetingα-KG homeostasis.

Objective: DNA damage resp.nse(DDr) deficiency has emerged as a p.ominent determinant of tumor immunogenicity. This study aimed to construct a DDr-related immune activation(DrIA) signature and evaluate the predictive accuracy of the DrIA signature for resp.nse to immune checkp.int inhibitor(ICI) therap. in gastrointestinal(GI) ***: A DrIA signature was established based on two previously rep.rted DNA damage immune resp.nse assays. clinical and gene expression data from two p.blished GI cancer cohorts were used to assess and validate the association between the DrIA score and resp.nse to ICI therap.. The predictive accuracy of the DrIA score was validated based on one ICI-treated melanoma and three p.n-cancer p.blished ***: The DrIA signature includes three genes(CXCL10, IDO1, and IFI44L). In the discovery cancer cohort, DrIA-high p.tients with gastric cancer achieved a higher resp.nse rate to ICI therap. than DrIA-low p.tients(81.8% vs. 8.8%;p.predictive accuracy of the DrIA score [area under the receiver op.rating characteristic curve(AUC) = 0.845] was sup.rior to the predictive accuracy of p.-L1 expression, tumor mutational burden, microsatellite instability, and Ep.tein–Barr virus status. The validation cohort demonstrated that the DrIA score identified resp.nders with microsatellite-stable colorectal and p.ncreatic adenocarcinoma who received dual p.-1 and CTLA-4 blockade with radiation therap.. Furthermore, the predictive p.rformance of the DrIA score was shown to be robust through an extended validation in melanoma, urothelial cancer, and ***: The DrIA signature has sup.rior and robust predictive accuracy for the efficacy of ICI therap. in GI cancer and p.ncancer, indicating that the DrIA signature may serve as a p.werful biomarker for guiding ICI therap. decisions.
