Transgenic mice are widely used to delete or overexpress genes in a cell specific manner to advance knowledge of bone biology,function and *** numerous Cre models exist to target gene recombination in osteoblasts and osteoclasts,few target osteocytes specifically,particularly mature *** goal was to create a spatial and temporal conditional Cre model using tamoxifen to induce Cre activity in mature osteocytes using a bac construct containing the 5’and 3’regions of the Sost gene(Sost ERT2 Cre).Four founder lines were crossed with the Ai9 Cre reporter *** founder line showed high and specific activity in mature *** and organs were imaged and fluorescent signal *** no activity was observed in 2 day old pups,by 2 months of age some osteocytes were positive as osteocyte Cre activity became spontaneous or‘leaky’with *** percentage of positive osteocytes increased following tamoxifen injection,especially in males,with 43%to 95%positive cells compared to 19%to 32%in *** signal was observed in any bone surface cell,bone marrow,nor in muscle with or without tamoxifen *** spontaneous signal was observed in any other ***,with tamoxifen injection,a few positive cells were observed in kidney,eye,lung,heart and *** other organs,28 in total,were negative with tamoxifen ***,with age,a muscle phenotype was apparent in the Sost-ERT2 Cre ***,although this mouse model may be useful for targeting gene deletion or expression to mature osteocytes,the muscle phenotype may restrict the use of this model to specific applications and should be considered when interpreting data.

This article is devoted to the study of a quasilinear Schrodinger equation coupled with an elliptic equation on the metric g. We first prove that, in this context, the propagation of regularity holds which ensures local wellposedness for initial data small enough in H1/2 and belonging to the besov space b3/2 2,1. In a second step, we establish Strichartz estimates for time dependent rough metrics to obtain a lower bound of the time existence which only involves the b1＋ε 2,∞ norm on the initial data.

The vanadium redox flow battery(VRFb)has been receiving great attention in recent years as one of the most viable energy storage technologies for large-scale ***,higher concentrations of vanadium species are required in the H_(2)O-H_(2)SO_(4) electrolyte in order to improve the VRFb energy *** might lead to unwanted precipitation of vanadium compounds,whose nature has not been accurately characterized *** this purpose,this study reports the preparation ofⅤ^((Ⅱ)),ⅤV^((Ⅲ)),Ⅴ^((Ⅳ))andⅤ^((Ⅴ))supersaturated solutions in a 5 M H_(2)SO_(4)-H_(2)O electrolyte by an electrolytic method,from the only vanadium sulfate compound commercially available(VOSO_(4)).The precipitates obtained by ageing of the stirred solutions are representative of the solids that may form in a VRFb operated with such supersaturated *** solid phases are identified using thermogravimetric analysis,X-ray diffraction and *** report that dissolvedⅤ^((Ⅱ)),Ⅴ^((Ⅲ))andⅤ^((Ⅳ))species precipitate as crystals of VSO_(4),V_(2)(SO_(4))3 and VOSO_(4) hydrates and not in their anhydrous form;conversely V^((Ⅴ))precipitates as an amorphous V_(2) O_(5) oxide partially *** measured hydration degrees(respectively 1.5,9,3 and 0.26 mol of H_(2)O per mol of compound)might significantly affect the overall engineering of VRFb operating with high vanadium concentrations.

Effect of 2LMISEN<sup>®</sup>on Long-Term Hippocampal Neurons Culture as a Screening Senescent Cells Model: p16<sup>INK4A</sup>and Caspase 3 Quantification

The aim of the present study was to investigate the effect of one capsule of the micro-immunotherapy medicine (MIM) 2LMISEN®compared to vehicle, in a neuronal aging model. Senescence and apoptosis of hippocampal neurons were evaluated by measuring p16INK4a and caspase 3 levels, respectively. The data presented is a single observation. Mice hippocampal neuron cultures were treated with MIM (11 mM) or vehicle (11 mM) from 22 days in vitro (DIV) until 27 DIV. After incubation, hippocampal neuron cultures were fixed at 15 (control condition), 22, 25 and 27 DIV and then incubated with primary antibodies p16INK4a, MAP2 and Caspase 3. Quantification of p16INK4a and Caspase 3-positive neurons was done using Developer software. We found that vehicle had no effect on p16INK4a expression, whereas MIM was able to decrease p16INK4a levels at 22, 25 and 27 DIV in a statistically significant manner. The MIM had no significative effect on Caspase 3 expression. Our preliminary results showed that the MIM capsule significantly reduced neuronal senescence and not apoptosis.

A hallmark of systemic lupus erythematosus(SLE)is the breaking of b-cell tolerance with the generation of high-affinity autoantibodies;however,the antibody-independent features of the b-cell compartment in SLE are less *** this study,we performed an extensive examination of b-cell subsets and their proinflammatory properties in a Chinese cohort of new-onset SLE *** observed that SLE patients exhibited an increased frequency of transitional b cells compared with healthy donors and rheumatoid arthritis *** from SLE patients potently promoted the survival of transitional b cells in a type I IFN-dependent manner,which can be recapitulated by direct IFN-α***,the effect of IFN-αon enhanced survival of transitional b cells was associated with NF-κb pathway activation and reduced expression of the pro-apoptotic molecule *** b cells from SLE patients harbored a higher capacity to produce proinflammatory cytokine IL-6,which was also linked to the overactivated type I IFN *** addition,the frequency of IL-6-producing transitional b cells was positively correlated with disease activity in SLE patients,and these cells were significantly reduced after short-term standard ***,the current study provides a direct link between type I IFN pathway overactivation and the abnormally high frequency and proinflammatory properties of transitional b cells in active SLE patients,which contributes to the understanding of the roles of type I IFNs and b cells in the pathogenesis of SLE.

In pulsar timing, timing residuals are the differences between the observed times of arrival and predictions from the timing model. A comprehensive timing model will produce featureless resid- uals, which are presumably composed of dominating noise and weak physical effects excluded from the timing model （e.g. gravitational waves）. In order to apply optimal statistical methods for detecting weak gravitational wave signals, we need to know the statistical properties of noise components in the residuals. In this paper we utilize a variety of non-parametric statistical tests to analyze the whiteness and Gaussianity of the North American Nanohertz Observatory for Gravitational Waves （NANOGrav） 5- year timing data, which are obtained from Arecibo Observatory and Green bank Telescope from 2005 to 2010. We find that most of the data are consistent with white noise; many data deviate from Gaussianity at different levels, nevertheless, removing outliers in some pulsars will mitigate the deviations.

The cumulative effects of periodic redox cycling on the mobility of As,Fe,and S from alluvial sediment to groundwater were investigated in bioreactor *** particular sediments from the alluvial floodplain of the Mekong Delta River were investigated: Matrix A (14 m deep) had a higher pyrite concentration than matrix b (7 m deep) *** present in matrix b but absent in matrix *** the reactors,the sediment suspensions were supplemented with As(III) and SO4^2-,and were subjected to three full-redox cycles entailing phases of nitrogen/CO2,compressed air sparging,and cellobiose *** differences in As concentration and speciation were observed upon redox *** support the fact that initial sediment composition is the main factor controlling arsenic release and its speciation during the redox ***,a high pyrite content associated with a low SO4^2- content resulted in an increase in dissolved As concentrations,mainly in the form of As(III),after anoxic half-cycles;whereas a decrease in As concentrations mainly in the form of As(V),was instead observed after oxic *** addition,oxic conditions were found to be responsible for pyrite and arsenian pyrite oxidation,increasing the As pool available for *** same processes seem to occur in sediment with the presence of gypsum,but,in this case,dissolved As were sequestered by biotic or abiotic redox reactions occurring in the FeeS system,and by specific physico-chemical condition (***).The contrasting results obtained for two sediments sampled from the same core show that many complexes and entangled factors are at work,and further refinement is needed to explain the spatial and temporal variability of As release to groundwater of the Mekong River Delta (Vietnam).

PHOX2b mutations in three Chinese patients with congenital central hypoventilation syndrome

Congenital central hypoventilation syndrome （CCHS, OMIM #209880） is a rare autosomal dominant disorder of the autonomic nervous system （ANS） characterized by an abnormal autonomic ventilatory response to progressive hypercarbia and sustained hypoxemia. Patients typically present in the newborn period with hypoventilation or apnea asleep, awake, or both, without any associated cardiac, pulmonary, neuromuscular or brainstem lesions. Rarely, some patients may present at a later age and are diagnosed to have late onset central hypoventilation syndrome （LOCHS）.1 Other features of ANS dysfunction such as feeding difficulty due to oesophageal dysmotility, severe constipation in the absence of Hirschsprung disease, poor regulation of basal body temperature, episodes of profuse sweating, pupillary and ocular abnormalities, decreased beat-to-beat variability of heart rate, attenuated response of heart rate to exercise, abnormal fluctuation of blood pressure, decreased perception to pain, and decreased perception to anxiety may be variably present but not essential for diagnosis Furthermore, this central hypoventilation can occur as an isolated feature or in association with a number of neurocristopathies, notably Hirschsprung disease （Haddad syndrome, OMIM #209880） and tumours of the sympathetic nervous system particularly neuroblastoma, ganglioneuro- blastoma, and ganglioneuroma, which were found in 20% and 5%--10% of CCHS patients, respectively.2 Studies of genes pertinent to the early embryologic development of the neural crest cells, specifically the endothelin and the RET-GDNF signaling pathways, have recently led to the identification of PHOX2b as the major disease causing gene for CCHS.2-6 PHOX2b was mapped to chromosome 4p12 and consists of 3 exons. It encodes a highly conserved paired-like homeobox transcription factor of 314 amino acids linked to the RET-GDNF signaling

background:Cholera is endemic in Guinea,having suffered consecutive outbreaks from 2004 to 2008 followed by a lull until the 2012 *** we describe the temporal-spatial and behavioural characteristics of cholera cases in Conakry during a three-year period,including the large-scale 2012 ***:We used the national and African Cholera Surveillance Network(Africhol)surveillance data collected from every cholera treatment centre in Conakry city from August 2011 to December *** prevalence of suspect and confirmed cholera cases,the case fatality ratio(CFR),and the factors associated with suspected cholera were described according to three periods:pre-epidemic(A),epidemic 2012(b)and post epidemic(C).Weekly attack rates and temporal-spatial clustering were calculated at municipality level for period *** was confirmed by culture at the cholera national reference ***:A total of 4559 suspect cases were reported:66,4437,and 66 suspect cases in periods A,b and C,*** the 204 suspect cases with culture results available,6%,60%,and 70%were confirmed in periods A,b,and C,*** 0.3%,the CFR was significantly lower in period b than in periods A(7.6%)and C(7.1%).The overall attack rate was 0.28%in period b,ranging from 0.17%to 0.31%across ***,a cluster of cases was identified in two districts in the northern part of *** 14%,rice water stools were less frequent in period A than in period b and C(78%and 84%).Dehydration(31%vs 94%and 89%)and coma(0.4%vs 3.1%and 2.9%)were lower during period b than in periods A and *** treatment of drinking water was less frequent in period A,while there were more reports of recent travel in period ***:The epidemic dynamic and the sociological description of suspect cases before,during,and after the large-scale epidemic revealed that the Vibrio cholerae was already present before the ***,it appeared that infected individuals react
