A novel alternative for pyrogen detection based on a transgenic cell line

Pyrogen,often as a contaminant,is a key indicator affecting the safety of almost all parenteral drugs(including biologicals,chemicals,traditional Chinese medicines and medical devices).It has become a goal to completely replace the in vivo rabbit pyrogen test by using the in vitro pyrogen test based on the promoted‘reduction,replacement and refinement’principle,which has been highly considered by regulatory agencies from different *** used NF-κB,a central signalling molecule mediating inflammatory responses,as a pyrogenic marker and the monocyte line THP-1 transfected with a luciferase reporter gene regulated by NF-κB as an in vitro model to detect pyrogens by measuring the intensity of a fluorescence ***,we show that this test can quantitatively and sensitively detect endotoxin(lipopolysaccharide from different strains)and nonendotoxin(lipoteichoic acid,zymosan,peptidoglycan,lectin and glucan),has good stability in terms of NF-κB activity and cell phenotypes at 39 cell passages and can be applied to detect pyrogens in biologicals(group A&C meningococcal polysaccharide vaccine;basiliximab;rabies vaccine(Vero cells)for human use,freeze-dried;Japanese encephalitis vaccine(Vero cells),inactivated;insulin aspart injection;human albumin;recombinant human erythropoietin injection(CHO Cell)).The within-laboratory reproducibility of the test in three independent laboratories was 85%,80%and 80%and the interlaboratory reproducibility among laboratories was 83.3%,95.6%and 86.7%.The sensitivity(true positive rate)and specificity(true negative rate)of the test were 89.9%and 90.9%,*** summary,the test provides a novel alternative for pyrogen detection.

Multi-Objective Optimization of VBHF in deep Drawing Based on the Improved QO-Jaya Algorithm

Blank holder force(BHF)is a crucial parameter in deep drawing,having close relation with the forming quality of sheet ***,there are different BHFs maintaining the best forming effect in different stages of deep *** variable blank holder force(VBHF)varying with the drawing stage can overcome this problem at an *** optimization of VBHF is to determine the optimal BHF in every deep drawing *** this paper,a new heuristic optimization algorithm named Jaya is introduced to solve the optimization *** improved“Quasi-oppositional”strategy is added to Jaya algorithm for improving population ***,an innovated stop criterion is added for better ***,the quality evaluation criteria for wrinkling and tearing are ***,the Kriging models are developed to approximate and quantify the relation between VBHF and forming defects under random ***,the optimization models are established and solved by the improved QO-Jaya algorithm.A VBHF optimization example of component with complicated shape and thin wall is studied to prove the effectiveness of the improved Jaya *** optimization results are compared with that obtained by other algorithms based on the TOPSIS method.

Operational requirements analysis method based on question answering of WEKG

The weapon and equipment operational requirement analysis(WEORA) is a necessary condition to win a future war,among which the acquisition of knowledge about weapons and equipment is a great challenge. The main challenge is that the existing weapons and equipment data fails to carry out structured knowledge representation, and knowledge navigation based on natural language cannot efficiently support the WEORA. To solve above problem, this research proposes a method based on question answering(QA) of weapons and equipment knowledge graph(WEKG) to construct and navigate the knowledge related to weapons and equipment in the WEORA. This method firstly constructs the WEKG, and builds a neutral network-based QA system over the WEKG by means of semantic parsing for knowledge navigation. Finally, the method is evaluated and a chatbot on the QA system is developed for the WEORA. Our proposed method has good performance in the accuracy and efficiency of searching target knowledge, and can well assist the WEORA.

Oxidative dehydrogenation of propane has been an ever-growing field for propylene production due to its exothermic properties,of which overoxidation is the major drawback,with CO and even CO_(2) as undesired *** the purpose of getting higher propylene selectivity as well as yield,herein,we report Ni single atoms supported on calcium aluminate as an efficient catalyst candidate for propane oxidative *** from higher valence states of Ni1 species,it shows 2—3 times as much propylene selectivity as that of Ni *** 14.2%C_(3)H_(6) yield with 47.3%propylene selectivity has been achieved on Ni single atom catalyst and a good stability during 20 h test can be obtained as well.

Pitting is a common type of localized corrosion in passive alloys that can cause rapid failure of material or *** the case of stainless steels,non-metallic inclusions have been identified as the most susceptible sites for pitting,and have therefore garnered significant *** review critically ex-amines the issue of how inclusions induce pitting,with a particular focus on three mechanisms:sponta-neous dissolution of inclusions,active dissolution of Cr-depleted regions,and propagation of microcracks at the inclusion-matrix *** researchers have made significant strides in understanding these mechanisms over the past few decades,many gaps and controversies *** such as the ini-tial driving force of inclusion dissolution and factors affecting Cr-depleted regions require further ***,some old concepts and methods need to be revised to arrive at more credible *** review aims to delve deeply into these important issues and provide inspiration for future research.

Superhydrophobic Co/Fe sulfide nanoparticles and single-atoms doped carbon nanosheets composite air cathode for high-performance zinc-air batteries

development of a high-performance bifunctional catalyst is essential for the actual implementation of zinc-air batteries in practical ***,a bifunctional cathode of Co_(3)S_(4)/FeS heterogeneous nanoparticles embedded in Co/Fe single-atom-loaded nitrogen-doped carbon nanosheets is ***-iron sulfides and single atomic sites with Co-N_(4)/Fe-N_(4)configurations are confirmed to coexist on the carbon matrix by EXAFS spectroscopy.3D self-supported super-hydrophobic multiphase composite cathode provides abundant active sites and facilitates gas–liquid-solid three-phase interface reactions,resulting in excellent electrocatalytic activity and batteries performance,i.e.,an OER overpotential(η_(10))of 260 mV,a half-wave potential(E_(1/2))of 0.872 V for ORR,aΔE of 0.618 V,and a discharge power density of 170 mW cm^(−2),a specific capacity of 816.3 mAh g^(−1).DFT analysis shows multiphase coupling of sulfide heterojunction through single-atomic metal doped carbon nanosheets reduces offset on center of electronic density of states before and after oxygen adsorption,and spin density of adsorbed oxygen with same spin orientation,leading to weakened charge/spin interactions between adsorbed oxygen and substrate,and a lowered oxygen adsorption energy to accelerate OER/ORR.

Bioinspired sensing-memory-computing integrated vision systems: biomimetic mechanisms, design principles, and applications

With the explosion of sensory data in the Internet of Things(Io T) era, conventional machine vision systems are becoming increasingly difficult to meet the requirements of high efficiency, low energy consumption, and low latency due to their inherent shortcomings of separate sensing, memory, and computing units. Inspired by the retina and neuromorphic computing, the sensing-memory-computing integrated vision system(SMCVS) that features low power consumption, low latency, and high parallelism has been considered a promising technology to surpass the von Neumann architecture and realize strong artificial ***, novel materials like two-dimensional semiconductors and quantum dots with novel optoelectronic performance provide hardware carriers for implementing integrated sensing-memory-computing architectures,attracting considerable attention. This paper reviews the recent research progress in bioinspired vision systems in terms of biomimetic mechanisms, design principles, computational architectures, and ***, the biomimetic mechanisms are illustrated to guide the design of high-performance artificial visual perception systems. Then the research progress of optoelectronic-synapse-based bioinspired vision systems in the device principles and applications including image filtering, color recognition, visual adaptation, and motion detection are summarized. Finally, the challenges and future developing directions of the SMCVS are provided regarding bionic application, architecture design, and device fabrication.

Effect of Room Temperature Ultrasonic Vibration Compression on the Microstructure Evolution and Mechanical Properties of AZ91 Alloy

To investigate the potential of direct ultrasonic vibration on improving the performance of magnesium alloys,this study first employed the ultrasonic vibration compression(UVC)on the solid solution treated AZ91 alloy,and explored its microstructure evolution and mechanical properties under *** only two seconds,the UVC alloys showed large deformation strains of 34.8–54.4%,and sudden increase of sample temperature to 243℃.Microstructure characterizations proved that UVC promoted the formation of abundant shear bands,fine grains,and the bimodal distribution of Mg17Al12 precipitates consisting of submicron particles located within the shear bands and nano-sized ones within the *** to the unique microstructure,the micro-hardness(and nano-hardness)value of UVC alloy was increased by 37.7%(35%)when compared with the solution-treated ***,the nano-modulus of the developed AZ91 alloy was also significantly increased to 62 GPa by statistical nanoindentation tests,which could be ascribed to increased Mg_(17)Al_(12) precipitates and decreased c/a value to some *** general,this work provides a new insight into the design and preparation of high-performance magnesium alloys by UVC at room temperature.

Seismic safety assessment with non-Gaussian random processes for train-bridge coupled systems

Extensive high-speed railway(HSR)network resembled the intricate vascular system of the human body,crisscrossing *** events,known for their unpredictability,pose a significant threat to both trains and bridges,given the HSR’s extended operational ***,ensuring the running safety of train-bridge coupled(TBC)system,primarily composed of simply supported beam bridges,is *** methods like the Monte Carlo method fall short in analyzing this intricate system ***,efficient algorithm like the new point estimate method combined with moment expansion approximation(NPEM-MEA)is applied to study random responses of numerical simulation TBC *** of the NPEM-MEA’s feasibility is conducted using the Monte Carlo *** analysis confirms the accuracy and efficiency of the method,with a recommended truncation order of four to six for the ***,the influences of seismic magnitude and epicentral distance are discussed based on the random dynamic responses in the TBC *** methodology not only facilitates seismic safety assessments for TBC systems but also contributes to standard-setting for these systems under earthquake conditions.
