Pediatric functional constipation treatment with Bifi dobacterium-containing yogurt:A crossover,double-blind,controlled trial

AIM: To evaluate the treatment of pediatric functional chronic intestinal constipation (FCIC) with a probiotic goat yogurt. METHODs: A crossover double-blind formula-controlled trial was carried out on 59 students (age range: 5-15 years) of a public school in Belo Horizonte, MG, Brazil, presenting a FCIC diagnostic, according to Roma Ⅲ criteria. The students were randomized in two groups to receive a goat yogurt supplemented with 109 colony forming unit/mL Bifidobacterium longum (***) (probiotic) daily or only the yogurt for a period of 5 wk (formula). Afterwards, the groups were intercrossed for another 5 wk. defecation frequency, stool consistency and abdominal and defecation pain were ***: Both treatment groups demonstrated improvement in defecation frequency compared to baseline. However, the group treated with probiotic showed most signif icant improvement in the f irst phase of the study. An inversion was observed after crossing over, resulting in a reduction in stool frequency when this group was treated by formula. Probiotic and formula improved stool consistency in the f irst phase of treatment, but the improvement obtained with probiotic was significantly higher (P = 0.03). In the second phase of treatment, the group initially treated with probiotic showed worseningstool consistency when using formula. However, the difference was not signif icant. A signif icant improvement in abdominal pain and defecation pain was observed with both probiotic and formula in the first phase of treatment, but again the improvement was more signif icant for the group treated with B. longum during phase I (P ssover study were analyzed, significant differences were observed between probiotic yogurt and yogurt only for defecation frequency (P = 0.012), defecation pain (P = 0.046) and abdominal pain (P = 0.015).

Objective:To determine an algorithm for molecular diagnosis of visceral leishmaniasis(vL)by kinetoplast DNA(kDNA)(Rv1/Rv2)and internal transcriber spacer(ITs1)(LITsR/L5.8 s)polymerase chain reaction(PCR),complemented by ITs 1 PCR restriction fragment length polymorphism(RFLP),using peripheral blood or bone marrow aspirate from patients with suspected ***:Biological samples were submitted to the gold standard for the diagnosis of vL and molecular diagnosis represented by ITs 1 PCR,kDNA PCR,and ITs 1 PCR *** samples were obtained from seven groups:groupⅠ,82 samples from patients with confirmed vL;groupⅡ,16 samples from patients under treatment for vL;groupⅢ,14 samples from dogs with canine visceral leishmaniasis(CvL);groupⅣ,a pool of six experimentally infected sandflies(Lutzomya longipalpis);group v,18 samples from patients with confirmed tegumentary leishmaniasis(TL)and groupsⅥandⅦwere from control groups without ***:The following gold standard and molecular examination results were obtained for each of the seven groups:groupⅠ:parasitologic and immunochromatographic tests showed a sensitivity of 76.3%(61 of 80)and 68.8%(55 of 80),respectively,and a sensitivity of 97.6%(80 of 82)and 92.7%(76 of 82)by ITs1 and kDNA PCR,*** ITs1 PCR RFLP(HaeⅢ)analysis of the 80 positive samples,52.5%(42 of 80)generated three fragments of 180,70,and 50 bp,corresponding to the pattern of Leishmania infantum infantum;groupⅡ:negative for the parasitologic methods and positive for IrK39(100%,16 of 16),presented 12.5%(2 of 16)of positivity by ITs 1 PCR and 25.0%(4 of 16)by kDNA PCR;groupⅢ:positive in the parasitologic and serologic tests(100%,14 of 14),presented 85.7%(12 of 14)of positivity by ITs1 PCR and kDNA ***1 PCR RFLP showed that 83.3%(10 of 12)of the canine samples contained parasites with profiles similar to ***;groupⅣpresented amplifications by ITs1 PCR and kDNA ***1 PCR products were analyzed by RFLP,generating a profile similar to

The coconut mite, Aceria guerreronis Keifer (Acari: Eriophyidae), is a major pest of coconut plantations (Cocos nucifera L.) worldwide. Here, we conducted a bioguided phytochemical approach using toxicity and repellency bioassays of nonpolar extract and its fractions of Vitex gardneriana Schauer (Lamiaceae) leaves to this pest. Nonpolar crude extract was fractionated by column chromatography using solvents with increased polarity and binary mixtures, resulting in five semipurified groups. The biomonitoring bioassay provided active fractions and led to the isolation and characterization of the bioactive compound squalene, a biosynthetic precursor of 20-hydroxyecdysone, which plays an important role in plant defense against arthropods. The LCsub>50sub> of the crude extract of v. gardneriana for A. guerreronis was estimated to be 0.185 mg·mLsup>-1sup> and LCsub>80sub> = 4.123 mg·mLsup>-1sup>. Also, the extract was highly repellent to this pest for up to 24 h. The fractions of v. gardneriana, and also squalene, caused mortality to A. guerreronis. The potential of v. gardneriana fractions/squalene as biopesticides for controlling A. guerreronis in coconut plantations span>is discussed herein.

Microstructural evolution during unsteady-state horizontal solidification of Al-si-Mg(356) alloy

The increasing demand for reducing vehicle weight in the automotive and aerospace industries has raised the need to develop improved structural aluminum-based alloys. Thus, horizontal solidification experiment with the Al-7%si-0.3%Mg（mass fraction） alloy was carried out. A water-cooled horizontal directional solidification device was developed and used. Microstructural characterization was carried out using traditional techniques of metallography, optical microscopy and sEM microscopy. The Thermo-Calc software was used to generate the solidification path of the investigated alloy with addition of 0.17% Fe（mass fraction）. The effects of the thermal parameters such as the growth rate（vL）, cooling rate（TC） and solidification local time（tsL） on the formation of the macrostructure and on the dendritic microstructure evolution were evaluated. A columnar to equiaxed transition（CET） was found for vL and TC values from 0.82 to 0.98 mm/s and from 1.71 to 2.55 ℃/s, respectively. The microstructure was characterized by the measurement of the primary and secondary dendrite arm spacings（λ1 and λ2, respectively）. Experimental laws of λ1 =f（vL, TC） and λ2 =f（tsL） were proposed. It is observed that the interdendritic region is composed of the following eutectic mixture： a（Al）＋si＋p-Al8 Mg3 Fe si6＋q-Mg2 si.

The Integration of Graphic Disciplines in Civil Engineering Courses through Computer Graphics

This paper presents the partial results of a project in progress that aims to improve the teaching and learning of Graphic Geometry modules taught in Engineering and Civil Engineering courses at the Federal University of Pernambuco through the development of education methodologies and content integration of these disciplines using computational technologies. This study covers and analyzes the contents of Tridimensional Graphic Geometry, Technical Drawing 3 and descriptive Geometry in the Bsc course in Civil Engineering, in order to create an integrated assessment model across these disciplines from the use of specific computer graphic softwares. It is proposed to use computer graphic for：（i） concept formation, （ii） completing and/or reviewing the content, （iii） creation of a database with virtual geometric models and their applications in Engineering and for the studies of their representations, geometric properties, etc., providing digital images of everyday forms and objects. In order to begin the diagnosis of the current situation, we gave an open questionnaire to 65 students from the 16 groups of three-dimensional graphical geometry, 20 students from the two courses in Technical Drawing 3 and 35 students from the three descriptive Geometry classes. The analysis of the data suggests that students better understand the two-dimensional representations of three-dimensional models through their modeling with the computer graphics software used, and the association of abstract concepts with concrete content. From the analysis of the data we can verify that when our students enter the foundation Engineering courses, the main difficulty in the disciplines of drawing is the three-dimensional visualization. The hypothesis is that the student will better understand the two-dimensional representations of three-dimensional models through their modeling with the computer graphic software used and the association of abstract concepts with concrete content.

Immobilization of Lipase from Candida rugosa on Mesoporous MCM 41

The use of enzymatic route for production of biofuels is growing up due the mild reaction conditions that this method provides, as well as reducing sOx emission. To reduce costs, it’s necessary to immobilize the enzyme, making possible to use it continuously as biocatalyst. The aim of this work was to measure the influence of the mass of support and pH used for immobilization of commercial lipase from Candida rugosa acquired by sigma laboratory. The immobilization method chosen was adsorption on mesoporous and hydrophobic support MCM 41, this has been treated with nitric acid 10% v/v to remove any organic residue. Then, 20 ml of enzymatic solution in phosphate buffer (pH 6.0, 7.0 and 8.0;50 mM) and 1 g/L was placed under constant stirring with 0.30 and 0.45 g of support. Aliquots were taken from the reaction medium and analyzed by spectrophotometry at 10 minutes intervals. a.v.lume of 0.2 ml of supernatant was put with 1.8 ml of substrate p-NFL at 0.18 g/L, and the absorbance at 410 nm was analyzed. In four cases there was a sharp reduction of supernatant’s activity at first 10 minutes, that ratifies the big affinity of the enzyme for the support and the negative influence of pH about the activity. Using the calibration curve, it was possible to calculate the final activity of each immobilization batch. This work suggests the occurrence of diffusional effects, which means that the enzyme mobility was restricted due the excessive amount of support, and then, it lost a part of accessibility to substrate, reflecting in not expressive activity values, and changing the state of ionization of the components of the system.

Analytical determination of Benzophenone-3 in sunscreen Preparations Using Boron-Doped Diamond Electrodes

A new electroanalytical procedure was developed for the determination of Benzophenone-3 (BENZO) in commercial sunscreen as the active ingredient. The procedure is based on the use of electrochemical methods as cyclic and square-wave voltammetry, with boron-doped diamond (BDD) electrodes. The reduction of BENZO in Britton-Robinson buffer (0.1 mol●L–1) using this type of electrode gives rise to one irreversible peak in –1.30 v (versus Ag/AgCl) in presence of cationic surfactant cetyltrimethylammonium bromide (CTABr). The proposed electrochemical method was successfully applied to the analysis of commercially available pharmaceutical preparations.
