Ge.e.ic susce.tibility to me.abolic associate. fatty live. dise.se.(mAFLD) is comple. and poorly characte.ize.. Accurate.characte.ization of the.ge.e.ic background of he.atic fat conte.t would provide insights into dise.se.e.iology and causality of risk factors. We.pe.forme. ge.ome.wide association study (GWAS) on two noninvasive.definitions of he.atic fat conte.t: magne.ic re.onance.imaging proton density fat fraction (mRI-PDFF) in 16,050 participants and fatty live. index (FLI) in 388,701 participants from the.Unite. Kingdom (UK) Biobank (UKBB). He.itability, ge.e.ic ove.lap, and similarity be.we.n he.atic fat conte.t phe.otype. we.e.analyze., and re.licate. in 10,398 participants from the.Unive.sity me.ical Ce.te. Groninge. (UmCG) Ge.e.ics Life.ine. Initiative.(UGLI). me.a-analysis of GWASs of mRI-PDFF in UKBB re.e.le. five.statistically significant loci, including two nove. ge.omic loci harboring CRe.3L1 (rs72910057-T, P = 5.40e.09) and GCm1 (rs1491489378-T, P = 3.16e.09), re.pe.tive.y, as we.l as thre. pre.iously re.orte. loci: PNPLA3, Tm6SF2, and APOe. GWAS of FLI in UKBB identifie. 196 ge.ome.wide significant loci, of which 49 we.e.re.licate. in UGLI, with top signals in ZPR1 (P = 3.35e.13) and FTO (P = 2.11e.09). Statistically significant ge.e.ic corre.ation (rg) be.we.n mRI-PDFF (UKBB) and FLI (UGLI) GWAS re.ults was found (rg = 0.5276, P = 1.45e.03). Nove. mRI-PDFF ge.e.ic signals (CRe.3L1 and GCm1) we.e.re.licate. in the.FLI GWAS. We.identifie. two nove. ge.e. for mRI-PDFF and 49 re.licable.loci for FLI. despite.a diffe.e.ce.in he.atic fat conte.t asse.sme.t be.we.n mRI-PDFF and FLI, a substantial similar ge.e.ic archite.ture.was found. FLI is identifie. as an e.sy and re.iable.approach to study he.atic fat conte.t at the.population le.e..
