Reliability of permanent downhole systems: Minimum sample and quality index

Permanent downhole monitoring systems are responsible for measuring pressure and temperature time series and enable uninterrupted reservoir characterization during the oil field production period, playing a key role in the oil and gas industry. Located in hostile pressure and temperature environments (i) close to the reservoir, in the case of the PDG (Permanent Downhole Gauge) sensor, and (ii) at the wellhead, in the case of the TPT (Pressure and Temperature Transducer) and PT (Pressure Transducer), its data are transmitted from the subsea environment to the Floating Production Storage and Offloading (FPSO), where the Master Control System (MCS) provides the information in engineering format. This information fulfills its function in the FPSO plant and finally is stored in an onshore data historian. Such complexity, importance, and maintenance difficulty of this system make it necessary to control and manage its reliability. Therefore, the objective of this work is to increase the availability and maximize the useful life of the downhole permanent monitoring system through the reliability calculation, using the Weibull estimate with 2 parameters, and the application of an index quality of statistical inferences. The proposed method for estimating reliability uses a database containing information from permanent downhole monitoring systems of the PDG, TPT, and PT type, from January 1st, 2008 to January 9th, 2014, and considers only the failures that occur until the arrival of the data in the MCS. From the reliability results, it can be observed that stratifications of this database could generate samples with a smaller number of observations, thus inferring reliability even with a small number of samples. The deepening of this method results in the definition of the minimum sample that allows removing reliability inferences without statistical significance and a quality index that allows classifying the reliability estimates of stratified sets of the largest sample o

Treatment of spinal cord injury with biomaterials and stem cell therapy in non-human primates and humans

This work explores the potential of a triple combination of meropenem(MEM),a novel metallo-blactamase(MBL)inhibitor(indole-2-carboxylate 58(InC58)),and a serine-b-lactamase(SBL)inhibitor(avibactam(AVI))for broad-spectrum activity against carbapenemase-producing bacteria.A diverse panel comprising MBL-and SBL-producing strains was used for susceptibility testing of the triple combination using the agar dilution *** frequency of resistance(FoR)to MEM combined with InC58 was *** were sequenced and tested for cross resistance,fitness,and the stability of the resistance *** with the double combinations of MEM plus an SBL or MBL inhibitor,the triple combination extended the spectrum of activity to most of the isolates bearing SBLs(oxacillinase-48(OXA-48)and Klebsiella pneumoniae carbapenemase-2(KPC-2))and MBLs(New delhi metallo-blactamases(NDMs)),although it was not effective against Verona integron-encoded metallo-blactamase(VIM)-carrying Pseudomonas aeruginosa(***)and OXA-23-carrying Acinetobacter baumannii(***).The FoR to MEM plus InC58 ranged from 2.22×10^(-7)to 1.13×10^(-6).The resistance correlated with mutations to ompC and comR,affecting porin C and copper permeability,*** mutants manifested a fitness cost,a decreased level of resistance during passage without antibiotic pressure,and cross resistance to another carbapenem(imipenem)and a b-lactamase inhibitor(taniborbactam).In conclusion,compared with the dual combinations,the triple combination of MEM with InC58 and AVI showed a much wider spectrum of activity against different carbapenemaseproducing bacteria,revealing a new strategy to combat b-lactamase-mediated antimicrobial resistance.

To establish the chemical profile, and to evaluate the antibacterial and modulatory activity of the ethanolic extracts of the stalk’s inner bark and heartwood of Secondatia *** of total phenols and flavonoids was determined by the Folin-Ciocalteu method and aluminum chloride, respectively. Phenolic compounds were identified and quantified by HPLC-DAD (High Performance Liquid Chromatography-Diodearray detector) and the Infrared Spectroscopy was performed using the measure by Attenuated Total Reflectance with Fourier Transform (ATR-FTIR). Antibacterial assays for determination of the Minimum Inhibitory Concentration (MIC) and modification of aminoglycosides were performed by *** spectra showed similar characteristics, having among its main absorption bands hydroxyl group (OH). The antibacterial activity showed clinically significant results for the strains of Staphylococcus aureus and Escherichia coli. In modulation assay, synergic and antagonistic effect for both extracts was observed. Heartwood extract in combination with antibiotics showed a significant MIC reduction at 19.8% (P < 0.000 1)-79.3% (P < 0.01).ConclusionsThis study is the first report of chemical and biological information of Secondatia floribunda suggesting that it is clinically relevant source of a new antibacterial therapy, especially due to the presence of significant levels of phenolic compounds.

Trauma to the peripheral nervous system often results in loss of motor and sensory functions of the affected area of the body,leading to a series of functional impairments(Allodi et al.,2012).Injuries to peripheral nerves initiate a series of complex events,known as Wallerian degeneration,which allows the injured axons to regenerate and reinnervate their targets(Allodi et al.,2012).Damage to a nerve induces multiple alterations,which include:axonal degeneration,breakdown of myelin sheath,Schwann cells(SC)proliferation and conversion to a repair phenotype which express cytokines and chemokines that allow the infiltration and activation of macrophages(Mietto et al.,2015;Jessen and Mirsky,2016).

Injection of bone marrow mesenchymal stem cells by intravenous or intraperitoneal routes is a viable alternative to spinal cord injury treatment in mice

In spite of advances in surgical care and rehabilitation, the consequences of spinal cord injury （SCI） are still challenging. Several experimental therapeutic strategies have been studied in the SCI field, and recent advances have led to the development of therapies that may act on the inhibitory microenvironment. Assorted lineages of stem cells are considered a good treatment for SCI. This study investigated the effect of systemic transplantation of mesenchymal stem cells （MSCs） in a compressive SCI model. Here we present results of the intraperitoneal route, which has not been used previously for MSC administration after compressive SCI. We used adult female C57BL/6 mice that underwent laminectomy at the T9 level, followed by spinal cord compression for 1 minute with a 30-g vascular clip. The animals were divided into five groups： sham （anesthesia and laminectomy but without compression injury induction）, MSC i.p. （intraperitoneal injection of 8×10^5 MSCs in 500 μL of DMEM at 7 days after SCI）, MSC i.v. （intravenous injection of 8 × 10^5 MSCs in 500μL of DMEM at 7 days after SCI）, DMEM i.p. （intraperitoneal injection of 500μL of DMEM at 7 days after SCI）, DMEM i.v. （intravenous injection of 500 μL of DMEM at 7 days after SCI）. The effects of MSCs transplantation in white matter sparing were analyzed by luxol fast blue staining. The number of preserved fibers was counted in semithin sections stained with toluidine blue and the presence of trophic factors was analyzed by immunohistochemistry. In addition, we analyzed the locomotor performance with Basso Mouse Scale and Global Mobility Test. Our results showed white matter preservation and a larger number of preserved fibers in the MSC groups than in the DMEM groups. Furthermore, the MSC groups had higher levels of trophic factors （brain-derived neurotrophic factor, nerve growth factor, neurotrophin-3 and neurotrophin-4） in the spinal cord and improved locomotor performance.
