Mesenchymal Stem Cells Express Low Levels of Cardiomyogenic Genes and Show Limited Plasticity towards Cardiomyogenic Phenotype

Mesenchymal stem cell differentiation towards osteogenic, chondrogenic and adipogenic lineages have been extensively described and reproduced in the literature. In contrast, cardiomyogenic differentiation still remains largely controversial. In this study the authors aim to shed new light into this unclear phenomenon and test whether BMMSC （bone marrow mesenchymal stem cells） and ATMSC （adipose tissue derived mesenchymal stem cells） are able to differentiate into functional cardiomyocytes, investigating two differentiation protocols. AT and BMMSC behaved differently when cultured in differentiation media and presented lower levels of proliferation and alkaline phosphatase production, expression of cardiomyocyte-specific transcription factors such as GATA-4, Nkx2-5 and proteins such as ct and 13 Myosin Heavy Chains. Furthermore, MSC started to express higher levels of Connexin-43 and c~ sarcomeric actinin protein. Unfortunately, though, MSC did not present cardiomyocyte-like electrophysiological properties. In order to analyze a possible explanation for such limited plasticity, the authors decided to address the issue using a quantitative approach. Gene expression was quantified by Real time PCR, and, for the first time, the authors show that a possible explanation for limited plasticity of MSC is that even though differentiated cells presented differential gene expression, the levels of key cardiomyogenic genes did not reach expression levels presented by adult cardiomyocytes, nor were maintained along differentiation, reaching peaks at 4 days of stimulation, and decaying thereafter.

Immunization with rPb27 Protects Mice from the Disruption of VEGF Signaling in Paracoccidioides brasiliensis Infection

VEGF （Vascular endothelial growth factor） signaling is critical for endothelial cell survival and maintenance of the vasculature. Deregulation of VEGF signaling contributes to the physiopathology of many diseases. However, the ways in which infection with Paracoccidioides brasiliensis affects VEGF signaling and the influence of immunization with rPb27 （recombinant protein Pb27） on VEGF signaling have not yet been studied. Animals were immunized with rPb27 and subsequently infected with a virulent strain of P. brasiliensis. The fungal load was evaluated by measuring CFU （colony-forming unit） and histology was performed to evaluate the inflammatory reaction. At the two time points analyzed, the PC （positive control） and TREAT （treated） animals had decreased levels of pulmonary VEGF compared to basal levels. However, in the immunized （Pb27） and treated mice （Pb27 ＋ TREAT）, VEGF expression remained unchanged after infection. In the case of VEGFR-2, the Pb27 and Pb27 ＋ TREAT groups showed increased levels of expression. Regarding the levels of the eNOS enzyme, only the Pb27 group did not reduce the expression levels relative to baseline. The immunization with rPb27 kept VEGF signaling, NO production and increased VEGFR-2 expression, after infection with P. brasiliensis. Thus, the authors infer that immunization with rPb27 protects mice from the disruption of VEGF signaling in Paracoccidioides brasiliensis infection.

Polygenic Profile and Magnitude Impact of Inflammatory Response in Soccer Athletes

The practice of soccer involves carrying-out actions of high intensity, which demand a great generation of eccentric strength, which in turn results in an increase in the inflammatory response after training practice and game matches. The study aimed to investigate, in combination and individually, the association of 28 polymorphisms with the inflammatory responses of soccer athletes. The sample consisted of 47 male under-20 soccer athletes who belong to clubs in the first division of Brazilian soccer. Blood samples were collected at Pre, and 03, 24, and 48 hours after the training session to evaluate the inflammatory responses (hematological analyzes (hemogram), creatine phosphokinase (CK), high sensitivity quantitative C-reactive protein (CRP), tumor necrosis factor-alpha, (TNFα), interleukin 6 (IL-6) and insulin-like growth factor 1 (IGF-1)). DNA was obtained through scraping of buccal cells, where a sterile swab was rubbed on the inner side of the mouth of each participant 06 times. The database was built using the TruSeq DNA PCRFree kit (Illumina®) and the Covaris equipment for shearing genomic DNA (gDNA) by ultrasound. Of the analyzed SNPs, 09 (ACTN3 rs1815739, COL5A1 rs12722, COL5A1 rs3196378, HGF rs5745697, IGF1 rs35767, IL-6 rs1800795, MMP3 rs679620, SLC30A8 rs13266634, SOX15 rs4227) were individually associated with biomarkers and 07 SNPs, (COL5A1 rs12722, COL5A1 rs3196378, COL5A1 rs1800012, HGF rs5745697, IGF1 rs35767, IL-6 rs1800795 and MMP3 rs679620) analyzed in combination, explained 16% to 40% of the variation of inflammatory responses in soccer athletes. The results suggest that the genotypic profile can be taken into account for a more individualized distribution of the training load, along with the elaboration of recovery strategies for high-level athletes between training sessions and games of high physical and physiological demand.

The Brazilian state of Acre has an extensive natural reserve of bamboo,making it one of the largest in loco gene *** aim of this study was to characterize the structure and genetic diversity of Guadua weberbaueri *** two populations,one native(FAPB)and the other anthropized(FAPBA),using ISSR *** results show that the FAPB population exhibited higher values for all estimates of population ***,the FAPBA population also showed high heterozygosity,corroborated by estimated gene flow(Nm=3.9)between the *** study of the association between Nei’s genetic distances and the geographic distances between the populations were significantly correlated(r=0.45,p=0.01),corroborated by the dendrogram revealing two distinct groups corresponding to the collection sites,without mixing classes between populations in the same *** for the coancestry coefficient,pairs of individuals in the first distance class were positive and significant,indicating that plants that are geographically closer share common alleles with a frequency greater than by chance,which means that there is a tendency that geographically closer individuals are *** presented similar genetic structure when the geographical distance between them was up to 56 m for FAPB and up to 156 m for *** was concluded that anthropized environments exhibit less genetic diversity than native environments,inferring risks for species conservation if appropriate and planned management techniques are not adopted.

Impacts of land use and land cover changes on hydrological processes and sediment yield determined using the SWAT model

Land use and land cover (LULC) changes strongly affect local hydrology and sediment *** current study focused on a basin in the Brazilian Amazon and had the following three objectives:(1) to perform an effective diagnosis of flow and sediment yield,(2) to evaluate the impacts of LULC changes over the last 40 years on the hydro-sedimentological variables,and (3) to investigate the impacts of the possible trends or breaking points in the flow,surface runoff,and sediment yield *** Soil and Water Assessment Tool (SWAT) model validation showed that the simulated results were consistent with the data measured in the dynamic reproduction of flow ***,changes in LULC altered surface runoff,sediment yield,and flow according to the Mann-Kendall and Pettitt non-parametric *** was also observed that the sub-basins in which pastureland is predominant are more susceptible to increased surface runoff and sediment *** to trend and homogeneity tests,these sub-basins had break points in the time series of these *** the other hand,in the sub-basins whose land cover is predominantly forest,the time series is homogeneous and *** current study highlighted the impacts that 40 years of LULC changes in an Amazonian basin had on hydro-sedimentological variables,which,in turn,can play an important role in changing the hydrological cycle,consequently these changes can also impact diverse human activities in the region,such as agribusiness,livestock,energy production,food security,and public water ***,it is concluded that the SWAT model is a good estimator of hydro-sedimentological processes in Amazonian basins and can be used by decision makers in the management of water and environmental resources.

In this work,the impact of the doping process on the photoluminescence emission of CaWO_(4) as a function of the concentration of Eu^(3+) cation(0.01 mol%,0.02 mol%,0.04 mol%,0.06 mol%,0.08 mol%,and 0.10 mol%) is discussed in ***_(1-x)WO~4:xEu^(3+) samples were successfully synthesized by a simple coprecipitation method followed by microwave *** blue shift in the absorption edge confirms the quantum confinement effect and the band gap energy covers the range from 3.91 to 4.18 eV,as the amount of Eu^(3+) cations *** experimental results are sustained by first-principles calculations,at the density functional theory level,to decipher the geometry and electronic properties,thereby enabling a more accurate and direct comparison between theory and experiment for the Ca_(1-x)WO_(4):xEu^(3+) structure.

Metabolic Syndrome in People with HIV/AIDS

Background: Highly Active Antiretroviral Therapy (HAART) has changed the clinical picture of HIV infection by reducing morbidity and mortality rates in the population. However, alterations in lipid metabolism leading to hypertriglyceridemia, hypercholesterolemia, insulin resistance, hyperglycemia and redistribution of body fat, which are risk factors for cardiovascular diseases, have emerged. Metabolic Syndrome (MS) is a complex disorder represented by a set of cardiovascular risk factors commonly associated with central adiposity and insulin resistance. Aim: Current paper evaluates the prevalence of MS in patients with HIV/AIDS using HAART from a reference Center in southern Brazil. Methods: Samples comprised patients who had the infection for at least five years and were undergoing antiretroviral therapy. Metabolic syndrome was identified according to the National Cholesterol Education Program Expert Panel on Detection, Evaluation, and Treatment of High Blood Cholesterol in Adults (NCEP-ATPIII). A physical examination was performed by evaluating percentage of body fat by bio-impedance and measuring blood pressure, determination of Body Mass Index and Waist-Hip Ratio, glycaemia, total cholesterol, HDL cholesterol, LDL cholesterol and triglycerides. Results: 184 patients were evaluated. MS prevalence was 30% (55 patients), with 30 (16.3%) males and 25 (13.7%) females. Conclusions: Brazil was among the first country profoundly impacted by the HIV/AIDS epidemic but today, Brazil has less than 1% adult HIV prevalence, implemented treatment and prevention programs early in the epidemic. Whereas there is currently a significant increase in the survival of HIV patients by HAART, the patients reveal a higher prevalence of Metabolic Syndrome in this specific population requiring political strategy of care to this population.

Homocysteine and Cardiovascular Risk Factors in Overweight or Obese Children and Adolescents

Introduction: Among the extrinsic factors, homocysteine (Hy) stands out, which is an intermediate amino acid of the intracellular metabolism of methionine involved in the process of cellular oxidation, which promotes the installation of atheromatous plaques and, therefore, is considered as an emerging cardiovascular risk factor. Objective: To evaluate the plasma homocysteine levels (Hy) in overweight or obese children and adolescents and their relation with cardiovascular risk factors. Methods: A cross-sectional study was conducted from July 2011 to May 2012 with overweight or obesity children and adolescents aged 2 to 18 years followed at the Center for Childhood Obesity (IOC), Campina Grande-PB. A structured form was used to record demographic, socioeconomic and clinics variables and the patients underwent laboratory tests to define their lipid and glucose profiles and measurement of plasma Hy levels. Results: The study evaluated a total of 165 children and adolescents with mean age of 12.5 (±2.5) years;the majority were female (57.0%). Regarding the lipid profile, there was more individuals with low HDL cholesterol (88.5%). Plasma Hy levels were high in 24.2% of the sample. The mean Hy levels ranged from 4.3 to 18.9 μmol/L, being higher in males, obese adolescents and also in patients with high insulin levels and resistance. Conclusions: The results shown in this study emphasize the importance of detecting and controlling the plasma Hy levels as an independent cardiovascular risk factor, and the need for further studies to evaluate the clinical and biological factors related to alterations in its metabolism.

Evaluation of the genotoxic and cytotoxic potentials is a part of the analysis process where plant products with therapeutic properties must be submitted for being safely employed, mainly through long time points, doannesia princeps, also known as ＂cotieira＂ has been used on popular medicine as laxative and several diseases treatment. Aiming to analyze genotoxic potential ofd. princeps leaves extract, Allium cepa assay and micronucleus test were employed. No genotoxic activity presented J. princeps leaves extract; however, extracts cytotoxic activity over A. cepa meristematic cells and mice marrow cells was observed. J. princeps leaves extract presented antiproliferative activity at concentrations and systems employed indicating their therapeutic potential for cellular cycle inhibition. Moreover, results showed that in regard to its mutagenic effect, usage of J. princeps leaves tea on popular medicine has been a safe procedure, as well.
