Mechanisms of Organelle Inheritance in Dividing Plant Cells

Organelles form essential compartments of all eukaryotic cells. Mechanisms that ensure the unbiased inheritance of organelles during cell division are therefore necessary to maintain the viability of future cell generations. Although inheritance of organelles represents a fundamental component of the cell cycle, surprisingly little is known about the underlying mechanisms that facilitate unbiased organelle inheritance. Evidence from a select number of studies, however, indicates that ordered organelle inheritance strategies exist in dividing cells of higher plants. The basic requirement for unbiased organelle inheritance is the duplication of organelle volume and distribution of the resulting organelle populations in a manner that facilitates unbiased partitioning of the organelle population to each daughter cell. Often, partitioning strategies are specific to the organelle, being influenced by the functional requirements of the organelle and whether the cells are mitotically active or re-entering into the cell cycle. Organelle partitioning mechanisms frequently depend on interactions with either the actin or microtubule cytoskeleton. In this focused review, we attempt to summarize key findings regarding organelle partitioning strategies in dividing cells of higher plants. we particularly concentrate on the role of the cytoskeleton in mediating unbiased organelle partitioning.

Functional Characterization and RNAi-Mediated Suppression Reveals Roles for Hexose Transporters in Sugar Accumulation by Tomato Fruit

Hexoses accumulate to high concentrations （-200 mM） in storage parenchyma cells of tomato fruit. Hexoses are sourced from the fruit apoplasm as hydrolysis products of phloem-imported sucrose. Three hexose transporters （LeHT1, LeHT2, LeHT3）, expressed in fruit storage parenchyma ceils, may contribute to hexose uptake by these cells. An analysis of their full-length sequences demonstrated that all three transporters belong to the STP subfamily of monosaccharide transporters that localize to plasma membranes. Heterologous expression of LeHT1 （and previously LeHT2, Gear et al., 2000）, but not LeHT3, rescued a hexose transportimpaired yeast mutant when raised on glucose or fructose as the sole carbon source. Biochemically, LeHT1, similarly to LeHT2, exhibited transport properties consistent with a high- affinity glucose/H^＋ symporter. Significantly, LeHT1 and LeHT2 also functioned as low-affinity fructose/H^＋ symporters with apparent Km values commensurate with those of fruit tissues. A substantial reduction （80-90%） in fruit expression levels of all LeHT genes by RNAi-mediated knockdown caused a 55% decrease in fruit hexose accumulation. In contrast, photoassimilate production by source leaves and phloem transport capacity to fruit were unaffected by transporter knockdown. Collectively, these findings demonstrate that LeHTs play key roles in driving accumulation of hexoses into storage parenchyma cells during tomato fruit development.

Porphyria cutanea tarda as a complication of therapy for chronic hepatitis C

There is a strong association between porphyria cutanea tarda (PCT) and chronic viral hepatitis C. Therapy for chronic viral hepatitis C may improve PCT. However, there are only a few reports of the de novo development of PCT during therapy for chronic viral hepatitis C. we describe the development of PCT in a 56-year-old patient with chronic viral hepatitis C after 12 wk of peginterferon/ribavirin therapy. In addition, the patient was homozygous for the H63D hereditary hemochromatosis gene (HFE ) mutation. The association of PCT with chronic viral hepatitis C and the possible role of hepatic iron overload and ribavirin-induced hemolytic anemia in the development of PCT during therapy for chronic viral hepatitis C are discussed.

Synthesis,and measurement of structural and magnetic properties,of La_(1-x)Na_xFeO_3 (0.0≤x≤0.3) perovskite oxides

Polycrystalline La1-xNaxFeO3 (0.0≤x≤0.3) solid solutions were synthesized by a solution combustion method using glycine as a fuel. The combustion synthesized compounds were characterized by powder X-ray diffraction (XRD), scanning electron microscopy (SEM), diffused reflectance (DR) in the UV-vis, and magnetic measurements. The crystal structure examined by X-ray powder diffraction indicated that the samples were single phase and crystallized in an orthorhombic (space group, Pbnm no.62) structure. The parent and sodium-substituted compounds showed canted anti-ferromagnetic behavior associated with an increase in magnetic moment as Na content increased. The changes in magnetic properties of the materials were correlated to the changes in structural features, as shown by Rietveld structural refinement of the materials.

Dynamic modeling of a three-dimensional braided composite thin plate considering braiding directions

Currently, there are a limited number of dynamic models available for braided composite plates with large overall motions, despite the incorporation of three-dimensional(3D) braided composites into rotating blade components. In this paper, a dynamic model of 3D 4-directional braided composite thin plates considering braiding directions is established. Based on Kirchhoff's plate assumptions, the displacement variables of the plate are expressed. By incorporating the braiding directions into the constitutive equation of the braided composites, the dynamic model of the plate considering braiding directions is obtained. The effects of the speeds, braiding directions, and braided angles on the responses of the plate with fixed-axis rotation and translational motion, respectively, are investigated. This paper presents a dynamic theory for calculating the deformation of 3D braided composite structures undergoing both translational and rotational motions. It also provides a simulation method for investigating the dynamic behavior of non-isotropic material plates in various applications.
