The exploration and identification of safe and effective vaccines for the SARS-CoV-2 pandemic have captured the world's attention and remains an ongoing issue due to concerns of balancing protection against emerging variants of concern while also generating long-lasting immunity. Here, we report the synthesis of a novel messenger ribonucleic acid encoding the spike protein in a lipid nanoparticle formulation (STI-7264) that generates robust humoral and cellular immunity following immunization of C57Bl6 mice. In an effort to improve immunity, a clinically focused lymphatic drug delivery device (MuVaxx) was engineered to modulate immune cells at the injection site (epidermis and dermis) and draining lymph node (LN) and tested to measure adaptive immunity. Using MuVaxx, immune responses were elicited and maintained at a 10-fold dose reduction compared to traditional intramuscular (IM) administration as measured by anti-spike antibodies, cytokine-producing CD8 T cells, neutralizing antibodies against the Washington (wild type) strain and South African (Beta) variants, and LN-resident spike-specific memory B cells. Remarkably, a 4-fold-elevated T cell response was observed in MuVaxx-administered vaccination compared to that of IM-administered vaccination. Thus, these data support further investigation into STI-7264 and lymphatic-mediated delivery using MuVaxx for SARS-CoV-2 and VoC vaccines.

目的探讨转录沉默子8(TIS8)基因在调控肝再生进程中的作用。方法野生小鼠(WT鼠)随机分为70%肝切除术组(PH组,每小组5只)和假手术组(SH组,分组同前)。TIS8基因敲除鼠(TIS8鼠)亦实施PH,随即分为术后48h组和术后72h组,每组5只。使用实时RT-PCR法测定PH和SH后WT鼠不同时相肝组织TIS8的mRNA水平；Western blot检测PH后48 h和72h WT鼠和TIS8鼠肝组织中TIS8蛋白质的表达水平并结合肝细胞复制周期进行比较。结果WT鼠TIS8基因的诱导表达始于PH后6～12h,继而下调,但于48h表达再次上调。WT鼠TIS8蛋白质的表达存在于肝静止期,PH后2～6h表达减弱,但12-36h表达再次升高,而48～72h表达略减弱；然而,TIS8鼠术后48h和72 h TIS8蛋白质的表达均明显减弱(P＜0．01),说明,肝再生过程中TIS8蛋白质表达的时段恰好对应于肝细胞的有丝分裂进程。结论正常肝再生的有效进程中,TIS8的表达是必需的；TIS8参与了肝细胞有丝分裂进程的调控。

The most common method of shipping cells between institutes and companies is sending them frozen, usually treated with anti-freeze solution (most commonly DMSO because it is less toxic than many alternatives), and then packaging them in dry ice for shipment. However many countries place restrictions on dry ice shipments. An alternative to shipping frozen cell vials is to send flasks of growing cells in media. This also has problems because cells in media have limited viability and the flasks can leak. Here we report on an alternative method for shipping viable cells at ambient temperature without dry ice or in media filled flasks. In this study we report on the development and properties of HemSol?. This is an inexpensive, eco-friendly and protects cell integrity at ambient temperature while maintaining viability. We have previously shown that HemSol? protects platelet and RBC function in cold storage and circulating tumor cells up to 6 days. Therefore we wanted to know if HemSol? could also be used to transport live cells. Since HemSol? is a liquid, we experimented with encasing the cells with HemSol? and gelatin so as to prevent dry ice shipment of cells and circumvent the shipping of cells in media. We performed mock shipping experiments where cells were stored in HemSol? gel kept at room temperature on a lab benchtop and cells stored in dry ice was also kept on lab benchtop for up to 2 days. After the mock shipping period, we analyzed cells for their functions. Our results show that cells in HemSol? gel have greater than 95% viability and restored biological functions in 2 hours, whereas, cells shipped in dry ice required more than 24 hours to recover and needed media change to remove the DMSO.

Background:This study aimed to determine the effect of different carbohydrate(CHO)doses on exercise capacity in patients with McArdle disease—the paradigm of“exercise intolerance”,characterized by complete muscle glycogen unavailability—and to determine whether higher exogenous glucose levels affect metabolic responses at the McArdle muscle cell(in vitro)***:Patients with McArdle disease(n=8)and healthy controls(n=9)underwent a 12-min submaximal cycling constant-load bout followed by a maximal ramp test 15 min after ingesting a non-caloric *** a randomized,double-blinded,cross-over design,patients repeated the tests after consuming either 75 g or 150 g of CHO(glucose:fructose=2:1).Cardiorespiratory,biochemical,perceptual,and electromyographic(EMG)variables were ***,glucose uptake and lactate appearance were studied in vitro in wild-type and McArdle mouse myotubes cultured with increasing glucose concentrations(0.35,1.00,4.50,and 10.00 g/L).Results:Compared with controls,patients showed the“classical”second-wind phenomenon(after prior disproportionate tachycardia,myalgia,and excess electromyographic activity during submaximal exercise,all p<0.05)and an impaired endurance exercise capacity(-51%ventilatory threshold and55%peak power output,both p<0.001).Regardless of the CHO dose(p<0.05 for both doses compared with the placebo),CHO intake increased blood glucose and lactate levels,decreased fat oxidation rates,and attenuated the second wind in the ***,only the higher dose increased ventilatory threshold(+27%,p=0.010)and peak power output(+18%,p=0.007).In vitro analyses revealed no differences in lactate levels across glucose concentrations in wild-type myotubes,whereas a doseresponse effect was observed in McArdle ***:CHO intake exerts beneficial effects on exercise capacity in McArdle disease,a condition associated with total muscle glycogen *** of these benefits are dose dependent.

介绍了超高温激光共聚焦显微镜(HT-CLSM)的工作原理,并利用HT-CLSM实现了高钢级管线钢微观组织转变的原位观察。首先,选用不同合金成分体系的X70和X80管线钢作为研究对象,在模拟手工电弧焊焊接工艺下实现了微观组织变化的实时观察,直观展现了贝氏体相变过程,并分析了合金元素对贝氏体相变前晶粒尺寸的影响。其次,在管线钢TMCP成型保温过程中,研究了不同合金元素对X80管线钢晶粒尺寸的影响。最后,在模拟手工电弧焊焊接和熔化极气体保护自动焊焊接工艺下研究了不同冷速对X80管线钢贝氏体相变的影响。HT-CLSM可以为管线钢的合金成分设计以及生产工艺、焊接工艺的研究提供原位观察方法。

Recognition of a pathogen by the plant immune system often triggers a form of regulated cell death traditionally known as the hypersensitive response(HR).This type of cell death occurs precisely at the site of pathogen recognition,and it is restricted to a few *** research has shed light on how plant immune receptors are mechanistically ***,two central key questions remain largely unresolved:how does cell death zonation take place,and what are the mechanisms that underpin this phenomenon?Consequently,bona fide transcriptional indicators of HR are lacking,which prevents deeper insight into its mechanisms before cell death becomes macroscopic and precludes early or live *** this study,to identify the transcriptional indicators of HR we used the paradigmatic Arabidopsis thaliana–Pseudomonas syringae pathosystem and performed a spatiotemporally resolved gene expression analysis that compared infected cells that will undergo HR upon pathogen recognition with bystander cells that will stay alive and activate *** data revealed unique and time-dependent differences in the repertoire of differentially expressed genes,expression profiles,and biological processes derived from tissue undergoing HR and that of its ***,we generated a pipeline based on concatenated pairwise comparisons between time,zone,and treatment that enabled us to define 13 robust transcriptional HR *** these genes,the promoter of an uncharacterized AAA-ATPase was used to obtain a fluorescent reporter transgenic line that displays a strong spatiotemporally resolved signal specifically in cells that will later undergo pathogen-triggered cell *** valuable set of genes can be used to define cells that are destined to die upon infection with HR-triggering bacteria,opening new avenues for specific and/or high-throughput techniques to study HR processes at a single-cell level.

Mycoviruses are viruses that infect fungi,and hypovirulence-associated mycoviruses have the potential to control fungal ***,it is unclear how mycovirus-mediated hypovirulent strains live and survive in the field,and no mycovirus has been applied for field crop *** this study,we found that a previously identified small DNA mycovirus(SsHADV-1)can convert its host,Sclerotinia sclerotiorum,from a typical necrotrophic pathogen to a beneficial endophytic ***-1 downregulates the expression of key pathogenicity factor genes in *** during *** growing in rapeseed,the SsHADV-1-infected strain DT-8 significantly regulates the expression of rapeseed genes involved in defense,hormone signaling,and circadian rhythm *** a result,plant growth is promoted and disease resistance is *** experiments showed that spraying DT-8 at the early flowering stage can reduce the disease severity of rapeseed stem rot by 67.6%and improve yield by 14.9%.Moreover,we discovered that SsHADV-1 could also infect other *** strains on DT-8-inoculated plants and that DT-8 could be recovered from dead *** findings suggest that the mycoviruses may have the ability to shape the origin of *** discoveries suggest that mycoviruses may influence the origin of endophytism and may also offer a novel strategy for disease control in which mycovirus-infected strains are used to improve crop health and release mycoviruses into the field.

分子蒸馏技术富集柚皮精油的抗氧化成分,经两次分子蒸馏后,得到两种重馏分(1RH、2RH)、两种中间馏分(1DM、2DM)和一种轻馏分(1DL)。分子蒸馏过程中,采用气相色谱-质谱联用技术(GC-MS)实时分析了各馏分的化学组成,确定了33种化合物。通过测定各馏分清除DPPH·和ABTS+·的能力得知,香叶醇和β-石竹烯是主要的抗氧化物质,被富集到重馏分(2RH)中,浓缩比分别为3.7和4.3,同时D-柠檬烯在轻馏分(1DL)中得到明显富集,浓缩比达4.9。抗氧化实验表明重馏分(2RH)的抗氧化能力较原油得到了明显提升,清除DPPH·和ABTS+·的能力分别比原油提高了60%和75%。分子蒸馏技术不仅用于分离脂肪和脂肪酸,而且在植物精油精制及提高附加值方向也具有广阔的应用前景。
