Rad7 E3 Ubiquitin ligase Attenuates Polyubiquitylation of Rpn10 and Dsk2 Following DNA Damage in <i>Saccharomyces cerevisiae</i>

During Nucleotide Excision Repair (NER) in the yeast S. cerevisiae, ubiquitylation of Rad4 is carried out by the E3 ubiquitin ligase that includes Rad7-Elc1-Cul3 and is critical to optimal NER. Rad7 E3 activity targets Rad4 for degradation by the proteaseome but, in principle, could also trigger other DNA damage responses. We observed increased nuclear ubiquitin foci (Ub-RFP) formation in S. cerevisiae containing a Rad7 E3 ligase mutant (rad7SOCS) in response to DNA damage by benzo[a]pyrenediolepoxide (BPDE) in dividing cells. Immunoblots reveal that ubiquitin conjugates of Rpn10 and Dsk2 accumulate in greater abundance in rad7SOCS compared to RAD7 in dividing cells in response to BPDE which makes Rpn10 and Dsk2 candidates for being the ubiquitylated species observed in our microscopy experiments. Microscopy analysis with strains containing Dsk2-GFP shows that Dsk2-GFP and Dsk2-GFP/Ub-RFP colocalized in nuclear foci form to an increased extent in a rad7SOCS mutant background in dividing cells than in a RAD7 wild-type strain. Further, Dsk2-GFP in the rad7SOCS strain formed more foci at the plasma membrane following BPDE treatment in dividing cells relative to strains containing RAD7 or a rad7△?deletion mutant. In response to a different agent, UV irradiation, levels of ubiquitylated proteins were increased in rad7SOCS relative to RAD7, and the proteasomal deubiquitylase subunit, Rpn11 was even monoubiquitylated in the absence of damaging agents. Together these data show that Rad7 E3 activity attenuates ubiq-uitylation of proteins regulating the shuttling of polyubiquitylated proteins to the proteasome (Dsk2 and Rpn10) and removal of ubiquitin chains just prior to degradation (Rpn11). Since Rad7 E3 ligase activity has been shown to increase ubiquitylation of its target proteins, yet our results show increased ubiquitylation in the absence of Rad7 E3, we suggest that Rad7 E3 action regulates ubiquitin ligase and deubiquitylase (DUB) activities that act on Rpn10, Dsk2 a

Some probiotic strains of lactobacilli appear to be protective against vulvovaginal candidiasis. The vaginal epithelial cell line (VK2 E6/E7) was used as a model to assess the protective mechanisms of probiotic lactobacilli for cells chal- lenged with Candida albicans. Co-culture of VK2 cells with lactobacillus rhamnosus GR-1 and lactobacillus reuteri RC-14 prior to C. albicans challenge showed reduced adherence of C. albicans to the VK2 cells and inhibition of C. albicans growth. H2O2 concentrations of 0.3 μg/ml, produced by lactobacilli and estrogen-primed VK2 cells, were inhi-bitory to C. albicans growth. C. albicans growth was also inhibited by 10 μg/ml lactic acid. C. albicans infection was increased by 17β-estradiol through induction of hyphal germination. l. reuteri RC-14, but not l. rhamnosus GR-1, H2O2, or lactic acid inhibited estrogen-stimulated hyphal germination. The results of this study support a role for H2O2 and lactic acid from probiotic bacteria in vaginal epithelial protection from candidiasis and a role for 17β-estradiol in the disease by induction of C. albicans hyphal germination.

The conditions for extracting polysaccharides from tea （Camellia sinensis l.） fruit peel （TFPPs） were studied. Three parameters （temperature, time, and liquid/solid ratio） affecting the extraction of TFPP were optimized using response surface methodology （RSM）. Under the optimized conditions, the yield of TFPP was predicted to be 4.98%. The physicochemical properties, in vitro antioxidant activities, and inhibitory effects on α-glucosidase of frac- tionated TFPPs （TFPP-0, TFPP-20, TFPP-40, and TFPP-60） were investigated. We found that the TFPPs were all acid protein-bound heteropolysaccharides, although with different chemical compositions. They had not only re- markable scavenging activity on 2,2＇-azinobis（3-ethylbenzothiazoline-6-sulfonic acid） diammonium salt （ABTS） and reducing activity, but also excellent inhibitory potential against α-glucosidase in vitro. Our results suggest that tea fruit peel could be treated as a potential bioresource for the development of polysaccharide antioxidants.

The work presents an experimental study held with shells of molluscs of the species Anomalocardia brasiliana and Tivela mactroides, commonly found in the Northeast coast of Brazil. A local community living from the collection of these animals was the motivation of the work. This community wishes to install an industry for processing these molluscs; however, as government agencies require making an appropriate allocation of discarded shells, the community needs an environmental project of final destination of waste. Thus, the expectation of the work was to obtain a material to be added to Portland cement to act as filler, and also obtain a limestone that can be used as ink, because the material is composed of 80% calcium carbonate. The shells were ground and passed the sieve 325 mesh （0.044 mm）. The material was characterized by X-ray diffraction and X-ray fluorescence. Mortars specimens were prepared with percentages： 0%, 5%, 10%, 15%, 20% and 30% replacement of Portland cement. Two conditions were analyzed： with shells in a raw and burned state. The obtained results confirm the filler material effect, which suggests its use as a partial replacement of cement. However, due to the short silicas＇s concentration, the pozzolanic effect was not observed.

目的:研究自身免疫性肝炎和药物性肝损伤临床和病理特征的异同,以提供更好的AIH和DIlI的诊断方法。方法:南京市第二医院2015年1月至2017年7月连续275例肝活检患者。根据临床、血清学和组织学特征,55例患者被诊断为DIlI或AIH(24DIlI和31AIH)。结果:实验室检查结果显示,AIH患者AST[132IU/l(57.8-320.5)vs 56.7 IU/l(32-137.6)]、ADA[17.5IU/l(13.4.6-26.6.7)vs 12.8IU/l(7.6-19.9)]和Ig G[17.5g/l(12.6-20.7)vs13.5g/l(10.89-15.5)]的中位值更高而DIlI患者AlB水平更高[42.1g/l 36.62-44.8)vs 37g/l(32,8-40.8)],有趣的是血清免疫学检测结果没有统计学意义(P>0.05)。此外,组织学分析显示41.92%的AIH活检呈现典型AIH特征,包括界面肝炎、淋巴浆细胞浸润、玫瑰花结形成和淋巴细胞穿入(分别为93.55%,70.97%,61.29%和51.61%),相反DIlI患者中仅4.17%表现典型AIH特征。结论:该研究表明,仅依靠临床表现和实验室检查较难准确区分AIH和DIlI,肝活组织检查对区分这两种情况提供了巨大的帮助。

Cryptosporidium spp. infection is one of the causes of diarrhea in people living with HIV/AIDS. The objective of this study is to compare the sensitivity of microscopy and molecular biology to determine the prevalence of Cryptosporidium spp. in Patients living With HIV (PlWH). This is a descriptive cross-sectional study conducted in three care centers for people living with HIV/AIDS in Abidjan. It took place from November 2018 to March 2020. Sociodemographic data were obtained via a questionnaire. Stool and blood samples were collected and analyzed for microscopy and Nested PCR detection of Cryptosporidium spp. Blood samples were analyzed for CD4+ count. A total of 363 stool samples were collected from the three sites. Individuals aged 40 - 50 years (36.52%) were most likely to participate in the study. HIV Type 1 accounted for 86.22% of the study population. The samples collected consisted of 47.65% diarrheal stool. Microscopic examination of the stool yielded a prevalence of 3.86% for Cryptosporidium spp. while the prevalence was 3.96% with molecular identification. No statistically significant difference was observed between these two prevalences (χ2 = 0.26;p = 0.609). CD4+ count was the factor associated with Cryptosporidium spp. infection for both microscopy (OR = 0.887, p = 0.001) and PCR (OR = 0.896, p = 0.001). This study demonstrated that Nested PCR improves the detection of Cryptosporidium spp. in patient diagnosis.

Annual rings from 30 year old vines in a California rootstock trial were measured to determine the effects of 15 different rootstocks on Chardonnay and Cabernet Sauvignon *** traits measuring vegetative growth,yield,berry quality,and nutrient uptake were collected at the beginning(1995 to 1999)and end(2017 to 2020)of the lifetime of a vineyard initially planted in 1991 and removed in 2021.X-ray Computed Tomography(CT)was used to measure ring widths in 103 *** width was modeled as a function of ring number using a negative exponential *** and late wood ring widths,cambium width,and scion trunk radius were correlated with 27 *** of annual ring width shows that scions alter the width of the first rings but that rootstocks alter the decay of later rings,consistently shortening ring width throughout the lifetime of the *** index,juice pH,photosynthetic assimilation and transpiration rates,and instantaneous water use efficiency are correlated with scion trunk ***,our research indicates that rootstocks modulate secondary growth over years,altering physiology and agronomic *** act in similar but distinct ways from climate to modulate ring width,which borrowing techniques from dendrochronology,can be used to monitor both genetic and environmental effects in woody perennial crop species.
