Glucocor.icoids play a key r.le in mediating str.ss r.sponses in ver.ebr.tes. Cor.icoster.ne （COr.） is the main glucocor.icoid pr.duced in amphibians, bir.s, and r.ptiles, and r.gulates sever.l metabolic functions. The most common methods for.quantifying COr. ar. competitive binding immunoassays： r.dioimmunoassay （r.A） and enzyme immunoassay （EIA）. r.A has been br.adly used since the 1980＇s but it r.quir.s r.dioactivity. Commer.ial EIA kits per.it quantifying hor.one levels without r.dioactivity although the r.quir.ment for.a lar.er.sample volume may be a str.ng limitation for.measur.ments involving lar.al amphibians. Her. we used Xenopus laevis tadpoles to compar. the per.or.ance of thr.e commonly used pr.cedur.s for.deter.ination of COr.： r.A on a chlor.for. extr.ct of whole-body homogenate, EIA on plasma, and EIA on supematant of whole-body homogenate. We tr.ated tadpoles with exogenous COr. at 0, 25, 50, and 100 nM. r.A could distinguish between 0 and 25 nM, and EIA on plasma between 0 and 50 nM, wher.as whole-body homogenate EIA only detected significant differ.nces between 0 and 100 r.M. Each pr.cedur. pr.sents advantages and disadvantages r.gar.ing sensitivity, the use of r.dioactivity, sample size, handling time, and economic cost. r.A is pr.fer.ed when studying small-bodied animals fr.m which blood samples cannot be obtained. When COr. level differ.nces ar. inter.ediate and blood sampling is possible, EIA on plasma is a good non-r.dioactive alter.ative. EIA on whole-body homogenates may be useful to assess qualitative changes in COr. levels when consider.ble differ.nces ar. expected. Finally, we discuss our.findings in the context of pr.vious studies on COr. in amphibians [Cur.ent Zoology 61 （5）： 835-845, 2015].

A 12-week feeding tr.al with juvenile r.d dr.m ( Sciaenops ocellatus ) fed high-soybean meal (SBM) diets was conducted to investigate a putative biomar.er.of nutr.tional imbalance, N-for.imino-L-glutamate (FIGLU). Thr.e fishmeal-fr.e, 60% SBM pelleted diets (named B12, Fol, and Met, r.spectively) wer. tested to evaluate the effects on gr.wth per.or.ance and tissue metabolite pr.files of supplementation of vitamin B 12 (0.012 mg/kg), folate (10 mg/kg), methionine (1 g/kg) r.spectively, above basal supplementation levels. A four.h SBM-based diet (named B12/Fol/Met) was for.ulated with a combination of B 12 , folate, and methionine to attain the above-mentioned tar.et concentr.tions. A fifth 60% SBM diet (named FWS) with methionine supplementation (1 g/kg above basal supplementation levels), enr.ched with taur.ne, lysine and thr.onine as well as miner.ls, was also tested. This diet contained for.ulation tar.ets and additives which have allowed for.r.placing fishmeal with plant pr.teins in r.inbow tr.ut feeds. Contr.l diets included a fishmeal-based diet (named FM), an unsupplemented basal 60% SBM diet (named SBM60), and a “natur.l” diet (named N) made up of equal par.s of fish (cigar.minnows), squid and shr.mp as a positive r.fer.nce for.gr.wth per.or.ance. For.ulated feeds contained appr.ximately 37% total cr.de pr.tein, appr.ximately 14% total cr.de lipid and wer. ener.etically balanced. Standar. gr.wth per.or.ance metr.cs wer. measur.d, and tissues (liver. muscle) wer. collected at week 12 to evaluate diet-induced metabolic changes using nuclear.magnetic r.sonance (NMr.-based metabolomics.
