Because of the close interaction between tumors and the immune system,immunotherapies are nowadays considered as the most promising treatment against *** order to define the diagnosis and the subsequent therapy,crucial information about the immune cells at the tumor site is ***,different types or activation status of cells may be indicative for specific and personalized ***,we present a quantitative method to identify ten different immuno-markers in the same tumor cut section,thereby saving precious samples and enabling correlative analysis on several cell families and their activation status in a tumor microenvironment *** designed and fabricated a microfluidic chip with optimal thermomechanical and optical properties for fast delivery of reagents on tissue slides and for fully automatic imaging by integration with an optical *** multiplexing capability of the system is enabled by an optimized cyclic immunofluorescence protocol,with which we demonstrated quantitative sequential immunostaining of up to ten biomarkers on the same tissue ***,we developed high-quality image-processing algorithms to map each cell in the entire *** proofof-concept analyses,we identified coexpression and colocalization patterns of biomarkers to classify the immune cells and their activation *** to the quantitativeness and the automation of both the experimental and analytical methods,we believe that this multiplexing approach will meet the increasing clinical need of personalized diagnostics and therapy in cancer pathology.

Concentration–discharge patterns of weathering products from global rivers

Quantifying the functional relationships relating river discharge and weathering products places key constraints on the negative feedback between the silicate weathering and climate. In this study we analyze the concentration–discharge relationships of weathering products from global rivers using previously compiled time-series datasets for concentrations and discharge from global rivers. To analyze the nature of the covariation between specific discharge and concentrations, we use both a power law equation and a recently developed solute production equation. The solute production equation allows us to quantify weathering efficiency, or the resistance to dilution at high runoff, via the Damkohler coefficient. These results are also compared to those derived using average concentration–discharge *** the power law exponent and the Damkohler coefficient increase and asymptote as catchments exhibit increasingly chemostatic behavior, resulting in an inverse relationship between the two parameters. We also show that using thedistribution of average concentration–discharge pairs from global rivers, rather than fitting concentration–discharge relationships for each individual river, underestimates global median weathering efficiency by up to a factor of ～10. This study demonstrates the utility of long time-series sampling of global rivers to elucidate controlling processes needed to quantify patterns in global silicate weathering rates.

The tumor suppressr p73 is a homolog of p53 and is capable of inducing cell cycle arrest and ***,we identify nerve growth factor receptor(NGFR,p75NTR,or CD271)as a novel negative p73 regulator.p73 activates NGFR transcription,which,in turn,promotes p73 degradation in a negative feedback *** directly binds to p73 central DNA-binding domain and suppresses p73 transcriptional activity as well as p73-mediated apoptosis in cancer ***,we uncover a previously unknown mechanism of NGFR-facilitated p73 degradation through the chaperone-mediated autophagy(CMA)***,our studies demonstrate a new oncogenic function for NGFR in inactivating p73 activity by promoting its degradation through the CMA.

Kikuchi-Fujimoto Disease Associated to the Epstein-Barr Virus. A Type of Rare Necrotizing Lymphadenitis and Its Differential Diagnosis

China's urban environmental management research has often been focused on comparison of pollution concentrations,physical phenomena or summary socioeconomic statistics at a citywide or city-regional scope and scale. This article proposes a scalar approach to evaluate the environmental management practices in post-Mao era China. The scalar approach for the assessment of urban sustainability research is distinctive in four aspects:1). it recognizes scalar complexity of environmental problems;2). it provides a more comprehensive and sophisticated framework to assess urban environmental management;3). it centers on the connection of urban environmental issues at a continuous spatial scale,ranging from individual level everyday practices such as choices of mode of transportation to citywide or urban-regional scales problems such as industrial pollution to global issues such as carbon emission. Using Shanghai as a case study,we aim to demonstrate how a scalar approach can contribute to current debates about urban sustainability in China. Analysis suggests that Shanghai's urban environmental management in the past three decades has been focused primarily on the environmental problems at the intra-urban level. While this "end of pipe" approach has been quite effective in mitigating domestic and industrial pollution that were either accumulated during Mao's era or generated in post-Mao reform era,progresses were much slower in dealing with environmental problems both at a smaller (i.e.,those related to urban residents' everyday practices) scale and at a larger (i.e.,those related to inter-urban/regional or global issues) scale. Analysis further suggests that urban environmental management policies that can prioritize and address urban environmental impacts at continuous geographic scales are urgently needed in China in order to achieve long-term "triple bottom line" sustainability. Introduction: Kikuchi-Fujimoto disease (KFD), also known as histiocytic necrotizing lymphadenitis, is a specific and self-limited disease;its etiology is unknown. Some causal microorganisms have been proposed. The objective of the present article is to emphasize the clinicopathological characteristics of this disease that has been associated to the Epstein-Barr virus and to compare the histological changes with other types of necrotizing lymphadenopathies. Material and Methods: We studied 32 patients of the Surgical Pathology Service with necrotizing lymphadenitis, diagnosed in the years from 2004 to 2012 to found more cases of this rare disease in our Institution. Patients were 18 women and 14 men with an average age of 37 years. Results: The lymph nodes were cervical and axillary ones, some were associated to autoimmune diseases and no cause was identified in others. One of the cases, was diagnosed as KFD, presented morphological changes characteristic of this disease, such as subcapsular lymphoid follicles, zones with cell debris, epithelioid macrophages, clear-cytoplasm histiocytes, and immunoblast-reactive lymphocytes. Immunohistochemical markers were determined, such as CD20, CD2, CD4, CD8, CD68, lysozyme, CD56, granzyme B and EBER, which demonstrated the presence of B, T lymphocytes, histiocytes and cells positive to EBER. Histological changes in KFD occurred in three stages: proliferative stage, necrotizing, and xanthomatous. It is important to identify the histological stages of the disease because a differential diagnosis must be performed in regard to lymphadenopathies with necrosis and diverse types of lymphomas. Conclusion: We present a case of necrotizing lymphadenitis (KFD) associated to the Epstein-Barr virus and in some cases it is not possible to render a specific diagnosis based on morphologic findings, alone, and a diagnosis of necrotizing lymphadenitis may be used.
