AIM To investigate the protective effects of Ampelopsis grossedentata(AMP) on dextran sulfate sodium(DSS)-induced colitis in mice based on systems pharmacology *** Systems pharmacology approach was used to predict the active ingredients, candidate targets and the efficacy of AMP on ulcerative colitis(UC) using a holistic process of active compound screening, target fishing, network construction and analysis. A DSSinduced colitis model in C57 BL/6 mice(n = 10/group) was constructed and treated with 5-aminosalicylic acid(100 mg/kg/d) and AMP(400 mg/kg/d) to confirm the underlying mechanisms and effects of AMP on UC with western blot analyses, polymerase chain reaction, histological staining and *** The therapeutic effects of AMP against DSS-induced colitis were determined in the beginning, and the results showed that AMP significantly improved the disease in general observations and histopathology analysis. Subsequent systems pharmacology predicted 89 corresponding targets for the four candidate compounds of AMP, as well as 123 candidate targets of UC, and protein-protein interaction networks were constructed for the interaction of putative targets of AMP against UC. Enrichment analyses on TNF-α and RANKL/RANK, a receptor activator of NF-κB signaling pathways, were then carried out. Experimental validation revealed that inflammation-related signaling pathways were activated in the DSS group, and AMP significantly suppressed DSS-induced high expression of IRAK1, TRAF6, IκB and NF-κB, and inhibited the elevated expression levels of TNF-α, IL-1β, IL-6 and *** AMP could exert protective effects on UC via suppressing the IRAK1/TRAF6/NF-κB-mediated inflammatory signaling pathways.

To investigate the therapeutic effect of Jianpi Qingchang decoction (JPQCD) on dextran sulfate sodium (DSS)-induced ulcerative colitis (UC) in ***57BL/c mice were injected intragastrically with 5% DSS instead of drinking water for 7 d, and their body weight, diarrhea severity and fecal bleeding were monitored, while the mice in the control group were treated with standard drinking water, without DSS. After 7 d, the DSS drinking water was changed to normal water and the DSS group continued with DSS water. The control and DSS groups were given normal saline by intragastric injection. The 5-aminosalicylic acid (5-ASA) group was treated orally with 5-ASA at a dose of 100 mg/kg daily. The JPQCD group was treated orally with JPQCD at a dose of 17.1 g/kg daily. On day 14, the colon length was measured, the colorectal histopathological damage score was assessed, and protein levels of interleukin (IL)-1β, IL-8 and tumor necrosis factor-alpha (TNF-α) in colon supernatants were measured by enzyme-linked immunosorbent assay. mRNA expression of IL-1β, IL-8, TNF-α and nuclear factor-kappa B (NF-κB) was detected by real-time quantitative polymerase chain reaction. Western blotting was used to detect the protein expression of NF-κB and inhibitor of kappa *** inflammation occurred in the mice administered DSS, including the symptoms of losing body weight, loose feces/watery diarrhea and presence of fecal blood; all these symptoms worsened at 7 d. The colons of mice treated with DSS were assessed by histological examination, and the results confirmed that acute inflammation had occurred, as evidenced by loss of colonic mucosa and chronic inflammatory cell infiltration, and these features extended into the deeper layer of the colon walls. The expression levels of IL-1β, IL-8 and TNF-α in the DSS group were higher than those in the control group (P < 0.05), and the expression levels of IL-1β, IL-8 and TNF-α in the JPQCD and 5-ASA groups were lower than those in the

Self-incompatibility(SI)is a genetic mechanism that rejects self-pollen and thus prevents inbreeding in some hermaphroditic *** the Brassicaceae,SI involves a pollen-stigma recognition system controlled by a single locus known as the S locus,which consists of two highly polymorphic genes that encode S-locus cysteine-rich protein(SCR)and S-receptor kinase(SRK).When self-pollen lands on the stigma,the S-haplotype-specific interaction between SCR and SRK triggers ***,we show that the GATA transcription factor ***1 suppresses SI responses in Brassica napus and is induced after compatible *** loss-of-function mutant ***1 displays reduced *** contrast,overexpression of ***1 in self-incompatible stigmas leads to a partial breakdown of SI responses,suggesting that ***1 is a stigmatic compatibility ***,the expression levels of SRK and ARC1 are up-regulated in ***1 mutants,and they are down-regulated in ***1 overexpressing *** affects the cellular localization of ***1 through direct protein–protein ***,our findings highlight the fundamental role of ***1 in SI responses in ***,establishing a direct interaction between ***1 and SRK in this process.

介绍了鞍钢鲅鱼圈焦化废水处理工艺运行概况,分析了焦化废水工艺存在的问题,通过提升预处理单元、生化处理系统和深度处理系统的功能,新增膜脱盐处理系统,大大提升了焦化废水系统出水水质,中水回用率达到70%以上。最终膜脱盐处理系统的浓水水质指标达到了《炼焦化学工业污染物排放标准》(GB 16171—2012)中的间排标准,全部回用于烧结混料,净水全部回用于循环水系统,实现了焦化废水零排放的目标。

Generating diverse and factual text is challenging and is receiving increasing *** sampling from the latent space,variational autoencoder-based models have recently enhanced the diversity of generated ***,existing research predominantly depends on summarizationmodels to offer paragraph-level semantic information for enhancing factual *** challenge lies in effectively generating factual text using sentence-level variational autoencoder-based *** this paper,a novel model called fact-aware conditional variational autoencoder is proposed to balance the factual correctness and diversity of generated ***,our model encodes the input sentences and uses them as facts to build a conditional variational autoencoder *** training a conditional variational autoencoder network,the model is enabled to generate text based on input *** upon this foundation,the input text is passed to the discriminator along with the generated *** employing adversarial training,the model is encouraged to generate text that is indistinguishable to the discriminator,thereby enhancing the quality of the generated *** further improve the factual correctness,inspired by the natural language inference system,the entailment recognition task is introduced to be trained together with the discriminator via multi-task ***,based on the entailment recognition results,a penalty term is further proposed to reconstruct the loss of our model,forcing the generator to generate text consistent with the *** results demonstrate that compared with competitivemodels,ourmodel has achieved substantial improvements in both the quality and factual correctness of the text,despite only sacrificing a small amount of ***,when considering a comprehensive evaluation of diversity and quality metrics,our model has also demonstrated the best performance.

AIM: To evaluate the effect of antisense oligonucleotide targeting midkine (MK-AS) on angiogenesis in chick chorioallantoic membrane (CAM) and in situ human hepatocellular carcinoma (HCC). METHODS: An in situ human hepatocellular carcinoma (HCC) model and CAM assay were used in this experiment. The effect of MK-AS on angiogenesis was evaluated by cell proliferation assay and hematoxylin- eosin (HE) staining. RESULTS: MK-AS significantly inhibited human umbilical vein endothelial cells (HUVEC) and in situ human HCC growth. At the same time, MK-AS suppressed the angiogenesis both in human hepatocellular carcinoma cell line (HEPG2)-induced CAM and in situ human HCC tissues. CONCLUSION: MK-AS is an effective antiangiogenesis agent in vivo.
