Patients with a diagnosis of polycystic ovary syndrome （PCOS） are on the rise. About 4%-12% of women are currently estimated to have this condition. It is hypothesized that PCOS appears in women who have long-standing insulin resistance （1R）, which leads to high androgen and testosterone levels; this ultimately disrupts their menstrual cycles. Some researchers attribute IR to genetic factors, although there have been only minute changes in the human genome in the past 20 000 years. However, even with a stable gene pool, genes can be turned on and offby the environment, food and air quality and toxin exposure.

Leaf beetles Gastrophysa viridula have attracted recently increased research interest from various points of view, since they are：（i） pest insects in rhubarb crops; （ii） potential biocontrol agents of dock plants Rumex spp. in grasslands; and （iii） a model species in ecological studies on insect population dynamics, biochemistry, behavior, biomechanics and biomimetics. The continuous rearing of beetles at standardized conditions may help to unify the fitness state of different individuals, allowing a better comparison of experimental results. The present communication suggests a modular space- and time-saving rearing method ofG. viridula in stackable faunariums under laboratory conditions, which has been successfully established and continuously used over the last 5 years. Several developmental stages were kept in separate boxes, and multiple generations were kept simultaneously, depending on the required number of beetles.

Tumor cells display phenotypic plasticity and heterogeneity due to genetic and epigenetic variations which limit the predictability of therapeutic interventions. Chromatin modifications can arise stochas- tically but can also be a consequence of environmental influences such as the microenvironment of cancer cells. A better understanding of the impact and dynamics of epigenetic modulation at defined chromosomal sites is required to get access to the underlying mechanisms. We investigated the epige- netic modulations leading to cell-to-cell heterogeneity in a tumor cell line model. To this end, we analyzed expression variance in 80 genetically uniform cell populations having a single-copy reporter randomly integrated in the genorne. Single-cell analysis showed high intraclonal heterogeneity. Epige- netic characterization revealed that expression heterogeneity was accompanied by differential histone marks whereas contribution of DNA methylation could be excluded. Strikingly, some clones revealed a highly dynamic, stochastically altered chromatin state of the transgene cassette which was accompanied with a metastable expression pattern. In contrast, other clones represented a robust chromatin state of the transgene cassette with a stable expression pattern. Together, these results elucidate locus-specific epigenetic modulation in gene expression that contributes to phenotypic heterogeneity of cells and might account for cellular plasticity.

Background： Grain legumes represent a valuable energy source in pig diets due to their high starch content. The present study was conducted to determine the content and apparent ileal digestibility（AID） of starch in different grain legume cultivars for pigs by means of both a polarimetric and enzymatic method for starch ***： Three experiments were conducted with six barrows each which were fitted with ileal T-cannulas. In total, 18 diets including six different cultivars of faba beans（Vicia faba L.） and peas（Pisum sativum L.）, five different cultivars of lupins（Lupinus luteus L., Lupinus angustifolius L.）, and one diet with a soybean meal（SBM） were ***： The starch content of faba beans and peas was greater（P ＆lt; 0.05） when determined polarimetrically than enzymatically（438 vs. 345 g/kg dry matter（DM） in faba beans and 509 vs. 390 g/kg DM in peas, respectively）.Considerable lower starch contents were obtained in lupins and SBM, with 82 and 48 g/kg DM（analyzed polarimetrically） and ＆lt;1.1 and 3 g/kg DM（analyzed enzymatically）, respectively. Mean values for contents of neutral detergent fiber（NDF） and acid detergent fiber（ADF） in grain legumes ranged from 111 and 79 g/kg DM in peas to248 and 207 g/kg DM in lupins, respectively. Contents of condensed tannins in the colored flowered faba bean cultivars ranged from 2.1 to 7.4 g/kg DM. The AID of starch was greater（P ＆lt; 0.05） in pea than in faba bean cultivars,and using the polarimetric starch determination method resulted in greater（P ＆lt; 0.05） digestibility values than using enzymatic starch analysis（84 vs. 80% in faba beans and 86 vs. 83% in peas）. Moreover, AID of starch differed（P ＆lt; 0.05）within pea cultivars and starch digestibility in faba beans decreased linearly（P ＆lt; 0.05） as the content of condensed tannins increased. The aim of this study was to evaluate whether unburned carbon particles present in fly ash can help in the retention of S, Cl, Br, As, Se, Cu, Ni, Zn, Ga, Ge, Rb, and Pb out of flue gas during the coal combustion at fluidised-bed power station where the coal was combusted along with limestone. The competitive influence of 10%-25% CaO in fly ashes on the distribution of studied elements was studied as well to be clear which factor governs behaviour of studied elements. Except of S （with significant association with CaO） and Rb and Pb （with major affinity to A1203） the statistically significant and positive correlation coefficients were calculated for the relations between unburned carbon content and Br （0.959）, Cl （0.957）, Cu （0.916）, Se （0.898）, Ni （0.866）, As （0.861）, Zn （0.742）, Ge （0.717）, and Ga （0.588） content. The results suggest that the unburned carbon is promising material in terms of flue gas cleaning even if contained in highly calcareous fly ashes.

AIM： To investigate the biological effects of cishydroxyproline （CHP） on the rat pancreatic carcinoma cell line DSL6A, and to examine the underlying molecular mechanisms. METHODS： The effect of CHP on DSL6A cell proliferation was assessed by using BrdU incorporation. The expression of focal adhesion kinase （FAK） was characterized by Western blotting and immunofluorescence. Induction of endoplasmic reticulum （ER） stress was investigated by using RT-PCR and Western blotting for the glucose-related protein-78 （GRP78） and growth arrest and DNA inducible gene （GADD153）. Cell viability was determined through measuring the metabolic activity based on the reduction potential of DSL6A cells. Apoptosis was analyzed by detection of caspase-3 activation and cleavage of poly（ADP-ribose） polymerase （PARP） as well as DNA laddering. RESULTS： In addition to inhibition of proliferation, incubation with CHP induced proteolytic cleavage of FAK and a delocalisation of the enzyme from focal adhesions, followed by a loss of cell adherence. Simultaneously, we could show an increased expression of GRP78 and GADD153, indicating a CHP-mediated activation of the ER stress cascade in the DSL6A cell line. Prolonged incubation of DSL6A cells with CHP finally resulted in apoptotic cell death. Beside L-proline, the inhibition of intracellular proteolysis by addition of a broad spectrum protease inhibitor could abolish the effects of CHP on cellular functions and the molecular processes. In contrast, impeding the activity of apoptosis-executing caspases had no influence on CHP-mediated cell damage.CONCLUSION： Our data suggest that the initiation of ER stress machinery by CliP leads to an activation of intracellular proteolytic processes, including caspaseindependent FAK degradation, resulting in damaging pancreatic carcinoma cells.
