Microglia are resident macrophage cells in the central nervous system that search for pathogens or abnormal neural activities and migrate to resolve the *** effective search and targeted motion of macrophages mean dearly to maintaining a healthy brain,yet little is known about their migration *** this work,we study microglial motion with and without the presence of external *** discover that the cells are promptly attracted by the applied forces(i.e.,mechanotaxis),which is a tactic behavior as yet unconfirmed in ***,in both the explorative and the targeted migration,microglia display dynamics that is strikingly analogous to bacterial run-and-tumble motion.A closer examination reveals that microglial run-and-tumble is more sophisticated,e.g.,they display a short-term memory when tumbling and rely on active steering during runs to achieve mechanotaxis,probably via the responses of mechanosensitive ion *** differences reflect the sharp contrast between microglia and bacteria cells(eukaryotes ***)and their environments(compact tissue ***).Further analyses suggest that the reported migration dynamics has an optimal search efficiency and is shared among a subset of immune cells(human monocyte and macrophage).This work reveals a fruitful analogy between the locomotion of 2 remote systems and provides a framework for studying immune cells exploring complex environments.

A signaling cascade mediating fruit trait development via phosphorylation-modulated nuclear accumulation of JAZ repressor

It is generally accepted that jasmonate-ZIM domain(JAZ)repressors act to mediate jasmonate(JA)signaling via CORONATINE-INSENsiTIVE1(COI1)-mediated ***,we report a cryptic signaling cascade where a JAZ repressor,FvJAZ12,mediates multiple signaling inputs via phosphorylation-modulated subcellular translocation rather than the COI1-mediated degradation mechanism in strawberry(Fragaria vesca).FvJAZ12 acts to regulate flavor metabolism and defense response,and was found to be the target of Fv MPK6,a mitogen-activated protein kinase that is capable of responding to multiple signal ***6 phosphorylates FvJAZ12 at the amino acid residues S179 and T183 adjacent to the PY residues,thereby attenuating its nuclear accumulation and relieving its repression for FvMYC2,which acts to control the expression of lipoxygenase 3(FvLOX3),an important gene involved in JA biosynthesis and a diverse array of cellular *** data reveal a previously unreported mechanism for JA signaling and decipher a signaling cascade that links multiple signaling inputs with fruit trait development.

Tumor necrosis factorα(TNFα)exhibits diverse biological functions;however,its regulatory roles in myogenesis are not fully *** the present study,we explored the function of TNFαin myoblast proliferation,differentiation,migration,and myotube fusion in primary myoblasts and C2C12 *** this end,we constructed TNFαmuscle-conditional knockout(TNFα-CKO)mice and compared them with flox mice to assess the effects of TNFαknockout on skeletal *** indicated that TNFα-CKO mice displayed phenotypes such as accelerated muscle development,enhanced regenerative capacity,and improved exercise endurance compared to flox mice,with no significant differences observed in major visceral organs or skeletal *** label-free proteomic analysis,we found that TNFα-CKO altered the distribution of several muscle development-related proteins,such as Hira,Casz1,Casp7,Arhgap10,Gas1,Diaph1,Map3k20,Cfl2,and Igf2,in the nucleus and *** set enrichment analysis(GSEA)further revealed that TNFαdeficiency resulted in positive enrichment in oxidative phosphorylation and MyoD targets and negative enrichment in JAK-STAT *** findings suggest that TNFα-CKO positively regulates muscle growth and development,possibly via these newly identified targets and pathways.

Magnetic Fields Reduce Apoptosis by Suppressing Phase Separation of Tau-441

The biological effects of magnetic fields(MFs)have been a controversial ***,in recent years,there has been increasing evidence that MFs do affect biological ***,the physical mechanism remains ***,we show that MFs(16 T)reduce apoptosis in cell lines by inhibiting liquid–liquid phase separation(LLPS)of Tau-441,suggesting that the MF effect on LLPS may be one of the mechanisms for understanding the“mysterious”magnetobiological *** LLPS of Tau-441 occurred in the cytoplasm after induction with *** phase-separated droplets of Tau-441 recruited hexokinase(HK),resulting in a decrease in the amount of free HK in the *** cells,HK and Bax compete to bind to the voltage-dependent anion channel(VdaC I)on the mitochondrial membrane.A decrease in the number of free HK molecules increased the chance of Bax binding to VdaC I,leading to increased Bax-mediated *** the presence of a static MF,LLPS was marked inhibited and HK recruitment was reduced,resulting in an increased probability of HK binding to VdaC I and a decreased probability of Bax binding to VdaC I,thus reducing Bax-mediated *** findings revealed a new physical mechanism for understanding magnetobiological effects from the perspective of *** addition,these results show the potential applications of physical environments,such as MFs in this study,in the treatment of LLPS-related diseases.

Shale oil of the Qingshankou Formation of the Gulong Sag,the northern Songliao Basin,represents the first attempt at large-scale development of pure-shale-type shale oil in *** integrating the multiscale refined reservoir characterization with macro-micro-scale mechanical testing,it is clarified that the Gulong shale is characterized by high clay mineral content,high rock plasticity,highly-developed bedding,and prominent mechanical anisotropy.A three-dimensional(3D)fracture propagation model of hydraulic fracturing was built for the Gulong shale,which fully captures the hydraulic fracture distribution pattern affected by the high bedding density,in-situ stress,and fracturing treatment *** research showed that due to influences of bedding,hydraulic fracturing in the Gulong shale forms a complex fracture morphology featuring the main fracture with multiple perpendicular branches that have different lengths(like the outdoor directional TV antenna);however,the vertical propagation of fractures is inhibited,and the fracture height is commonly less than 10 *** limited stimulated reservoir volume(SRV)is the main problem facing the fracturing stimulation of the Gulong shale *** density has vital effects on fracture morphology,so case-specific fracturing designs shall be developed for shale intervals with different bedding development *** reservoirs with welldeveloped bedding,it is suggested to properly increase the perforation cluster spacing and raise the volume and proportions of viscous fluids of the pad,so as to effectively promote vertical fracture propagation and improve reservoir stimulation *** study integrates multi-scale fine reservoir characterization and macro-micro-scale mechanical testing,as well as the construction and numerical simulation of hydraulic fracturing models for high-density layered shale reservoirs,providing a new approach and methodological framework for the fracturing research of high-density layer

The earliest evidence for a microblade adaptation in the remote,high altitude regions of the Tibetan Plateau

Microblade assemblages are among the most common prehistoric archaeological materials found on the Tibetan Plateau(TP)and are thought to indicate large scale migration to and settlement of the *** microblade sites,however,have been systematically excavated,especially in the remotest,highest-elevation regions of the *** timing of the large-scale arrival,spread,and permanent settlement of people on the TP therefore remains *** this paper,we report on a recently excavated site,Locality 3 of the Nwya Devu site(ND3),located at 4600 meters above sea level(masl),near the shore of Ngoin Lake,on the interior *** analyses reveal a fairly typical microblade technological orientation and two types of microblade cores:wedge-shaped and semi-conical,which are similar to those found throughout North *** Optically Stimulated Luminescence(OSL)dating and AMS^(14)C dating,the age of ND3 ranges from 11 to 10 *** date range indicates ND3 is the oldest microblade site yet recorded in the remote,high-elevation regions of the TP and thus provides important information about when and how hunter-gatherers using microblades began exploiting the higher altitudes of the *** together,studies at ND3 and throughout the TP suggest that a microblade adaptation is associated with the first prolonged human occupation of the plateau and that microblades played a significant role in mediating the risks and facilitating the mobility necessary to permanently inhabit the TP.

Atomically dispersed calcium as solid strong base catalyst with high activity and stability

Solid strong base catalysts are highly attractive for diverse reactions owing to their advantages of neglectable corrosion,facile separation,and environmental ***,their widespread applications are impeded by basic components aggregation and low *** this work,we fabricate single calcium atoms on graphene （denoted as Ca1/G） by use of a redox strategy for the first time,producing solid strong base catalyst with high activity and *** precursor Ca（NO3）2is first reduced to CaO at 400℃ by the support graphene,forming CaO/G with conventional basic sites,and the subsequent reduction at 850℃ results in the generation of Ca1/G with atomically dispersed *** characterizations reveal that Ca single atoms are anchored on graphene in tetra-coordination （Ca–C2–N2） where N is in situ doped from Ca（NO3）*** atomically dispersed Ca,along with their anchoring on the support,endow Ca1/G with high activity and stability toward the transesterification reaction of ethylene carbonate with *** turnover frequency value reaches 128.0 h-1on Ca1/G,which is much higher than the traditional counterpart CaO/G and various reported solid strong bases (2.9–46.2 h-1).Moreover,the activity of Ca1/G is well maintained during 5 cycles,while 60% of activity is lost for the conventional analogue CaO/G due to the leaching of Ca.
