n-glycosylation of Wnt3 regulates the progression of hepatocellular carcinoma by affecting Wnt/β-catenin signal pathway

BACKGROUnd Wnt/FZd-mediated signaling pathways are activated in more than 90%of hepatocellular carcinoma(HCC)cell *** a well-known secretory glycoprotein,Wnt3 can interact with FZd receptors on the cell surface,thereby activating the Wnt/β-catenin signaling ***,the n-glycosylation modification site of Wnt3 and the effect of this modification on the biological function of the protein are still *** To investigate the effect of Wnt3 n-glycosylation on the biological function of HCC *** Site-directed mutagenesis was used to verify the Wnt3 n-glycosylation sites,actinomycin d treatment was used to detect the stability of Wnt3 after site-directed mutation,the binding of the n-glycosylation site-directed mutant Wnt3 to FZd7 was observed by laser confocal microscopy,and the effects of the n-glycosylation site-directed mutation of Wnt3 on the Wnt/β-catenin signaling pathway and the progression of HCC cells were detected by western blot and cell function *** Wnt3 has two n-glycosylation-modified sites(Asn90 and Asn301);when a single site at amino acid 301 is mutated,the stability of Wnt3 is weakened;the binding ability of Wnt3 to FZd7 decreases when both sites are mutated simultaneously;and the level of proteins related to the Wnt/β-catenin signaling pathway is *** proliferation,migration and invasion are also weakened in the case of single 301 site and double-site *** These results indicate that by inhibiting the n-glycosylation of Wnt3,the proliferation,migration,invasion and colony formation abilities of liver cancer cells can be weakened,which might provide new therapeutic strategies for clinical liver cancer in the future.

Multi-performance evaluation of high-throughput accelerated corrosion test for high-strength low-alloy 921A steel

The corrosion behavior of high-strength low-alloy 921A steel in a simulated marine atmospheric environment was studied using a high-throughput experimental *** corrosion behavior,corrosion morphology,and corrosion products of 921A steels were analyzed using various techniques,including corrosion mass loss method,polarization curve,white-light interferometry,scanning electron microscopy,energy-dispersive spectrometry,microbeam X-ray fluorescence spectrometry,X-ray diffraction technique,and X-ray photoelectron *** test results indicated that 921A steel exhibits better corrosion resistance than Q450nQR1 steel in simulated harsh atmospheric environments,as evidenced by a lower corrosion mass loss rate throughout the corrosion *** corrosion products of both steels consisted ofα-FeOOH,Fe_(3)O_(4),andγ-FeOOH,withα-FeOOH being more prevalent in the rust layer of 921A steel than in Q450nQR1 *** inner rust layer of 921A steel also exhibited an appositional enrichment region of Cr,ni,Mo,and V,leading to its superior corrosion resistance compared to that of Q450nQR1 *** efficacy of high-throughput accelerated corrosion experimental methods was highlighted for evaluating the corrosion resistance of steel materials in harsh environmental *** findings suggest that 921A steel exhibits better corrosion resistance compared to Q450nQR1 steel and has the potential to be more suitable in harsh marine atmospheric *** characterization of the rust layer structures and composition reveals the parallel enrichment of certain elements in the inner rust layer of 921A steel,which enhances its corrosion resistance.

Synergistically enhanced dielectric, insulating and thermally conductive performances of sandwich PMMA based dielectric films

A tri-layered(B-G-B) dielectric films were simply prepared by alternatively spin-coating boron nitride/polymethylmethacrylate(Bn/PMMA, B) and graphene nanosheets(GnS/PMMA, G) compound solutions. The structure, morphology, thermal conductivity, and particularly the dielectric and relaxation behaviors were systematically studied. Results showed that a good exfoliation and dispersion of Bn and GnS can be simply achieved through intensive mechanical compounding with PMMA in the internal mixer. The dielectric constant and breakdown strength of the obtained 1-3-1(B-G-B) sandwich composite film reached 4.3(@1 kHz) and 458.6kV/mm, being 119 % and 113 % higher than that of pure PMMA, respectively. Furthermore, the overall thermal conductivity of sandwich 1-3-1 composite increased by 112 % due to the addition of thermally conductive Bn and GnS. The sandwich design strategy provided an effective way of achieving good permittivity, high insulation, and improved thermal conductivity simultaneously, showing potential applications in the miniaturization and integration of electronic devices.

Influence of cerium and yttrium addition on strength and electrical conductivity of pure aluminum alloys

To develop pure aluminum alloys with high conductivity and strength, Al-0.2Ce and Al-0.2Ce-0.1y alloys were prepared by rolling and annealing processes in this study. The effects of trace rare earth elements on the strength and electrical conductivity of the pure aluminum alloys were investigated. It is revealed that the addition of Ce and y to pure aluminum can effectively enhance the strength and electrical conductivity of the alloys. In Al-0.2Ce, the addition of Ce can effectively refine the grain size of a-Al, with an average grain size of 90.68 μm in the as-cast state. The grain size of the alloy is further reduced to 87.55 μm by the simultaneous addition of y. The synergistic addition of Ce and y changes the properties of the alloy. The addition of Ce and y also produces the Al_(11)Ce_(3) and Al_(3)y second phases, which have coherent relationship with a-Al. The two-dimensional mismatch degree was calculated to be only 4.43%and 0.85%, respectively, which can provide a certain amount of nucleation substrate for a-Al in the incubation stage. The interfacial match between the L1_(2)structure of Al_(3)y and a-Al was calculated using first-principles simulations. The results indicate that Al_(3)y has a strong bonding effect with a-Al. nanoscale second phases at grain boundaries can be effective in reducing resistivity due to dislocation *** second phases with better matching interfaces to the substrate have no positive effect on the scattering motion of electrons.

In August 1872, British journalist William Simpson (1823-1899) traveled to China to cover the wedding of Emperor Tongzhi of China (reigned 1861-1875) for the Illustrated London news. Simpson, who had previously reported on the Crimean War and the frontlines of the Franco-Prussian War, was now on an “expedition” to *** Illustrated London news, established in1842, was the world’s first news magazine to use illustrations. By the 1860s, it had a larger circulation than any other newspaper of the time, with a readership mostly composed of the middle class.

X-ray excited photodynamic therapy(X-PdT)is the bravo answer of photodynamic therapy(PdT)for deep-seated tumors,as it employs X-ray as the irradiation source to overcome the limitation of light penetration ***,high X-ray irradiation dose caused organ lesions and side effects became the major barrier to X-PdT *** address this issue,this work employed a classic-al co-precipitation reaction to synthesize naLuF_(4):15%Tb^(3+)(nLF)with an average particle size of(23.48±0.91)nm,which was then coupled with the photosensitizer merocyanine 540(MC540)to form the X-PdT system nLF-MC540 with high production of singlet *** system could induce antitumor efficacy to about 24%in relative low dose X-ray irradiation range(0.1-0.3 Gy).In vivo,when nLF-MC540 irradiated by 0.1 Gy X-ray,the tumor inhibition percentage reached 89.5%±5.7%.The therapeutic mechanism of low dose X-PdT was found.A significant increase of neutrophils in serum was found on the third day after *** immunohistochemical staining of tumor sections,the Ly6G^(+),Cd8^(+),and Cd11c^(+)cells infiltrated in the tumor microenvironment were *** the bilat-eral tumor model,the nLF-MC540 with 0.1 Gy X-ray irradiation could inhibit both the primary tumor and the distant tumor ***-tected by enzyme linked immunosorbent assay(ELISA),two cytokines IFn-γand TnF-αin serum were upregulated 7 and 6 times than negative control,*** by enzyme linked immune spot assay(ELISPOT),the number of immune cells attributable to the IFn-γand TnF-αlevels in the group of low dose X-PdT were 14 and 6 times greater than that in the negative control group,***,it conclude that low dose X-PdT system could successfully upregulate the levels of immune cells,stimulate the secretion of cy-tokines(especially IFn-γand TnF-α),activate antitumor immunity,and finally inhibit colon tumor growth.

The obligate biotrophic basidiomycete fungus Puccinia striformis ***(Pst)is the causative agent of wheat stripe(yellow)rust,a disease that results in severe damage to global wheat production(Chen and Kang,2017).This pathogen has a mixed reproductive mode encompassing both asexual and sexual *** asexual stage is accomplished by reinfecting wheat with urediniospores,while the sexual cycle involves five different spore types and two phylogenetically unrelated plant hosts wheat,which is the primary host,and barberry,which is the alternate host(Zhao et al.,2016).This extremely complex lifecycle facilitates large genetic variation among Pst populations,which is conducive to the emergence of new virulent races that can overcome wheat resistance genes,resulting in epidemics(Chen and Kang,2017).Secreted effector proteins,the key virulence factors in fungal pathogens,can not only suppress plant immune systems but also trigger host immune responses following recognition by plant receptors(Toruno et al.,2016).
