Studying the membrane physiology of filamentous fungi is key to understanding their interactions with the environment and crucial for developing new therapeutic strategies for disease-causing ***,their plasma membrane has been inaccessible for a micron-sized patch-clamp pipette for pA current recordings due to the rigid chitinous cell ***,we report the first femtosecond iR laser nanosurgery of the cell wall of the filamentous fungi,which enabled patch-clamp measurements on protoplasts released from hyphae.A reproducible and highly precise(diffraction-limited,submicron resolution)method for obtaining viable released protoplasts was *** release from the nanosurgery-generated incisions in the cell wall was achieved from different regions of the *** plasma membrane of the obtained protoplasts formed tight and high-resistance(GΩ)contacts with the recording *** entire nanosurgical procedure followed by the patch-clamp technique could be completed in less than 1 *** to previous studies using heterologously expressed channels,this technique provides the opportunity to identify new ionic currents and to study the properties of the ion channels in the protoplasts of filamentous fungi in their native environment.

Background Extracellular vesicles(EVs) present in oviductal(OF) and uterine fluid(UF) have been shown to enhance bovine embryo quality during in vitro culture by reducing lipid contents and modulating lipid metabolism-related genes(LmGs), while also influencing cell proliferation, suggesting their involvement on the regulation of different biological pathways. The regulation of signaling pathways related to cell differentiation, proliferation, and metabo-lism is crucial for early embryo development and can determine the success or failure of the pregnancy. Bioactive molecules within EVs in maternal reproductive fluids, such as micro RNAs(miRNAs), may contribute to this regulatory process as they modulate gene expression through post-transcriptional *** This study evaluated miRNA cargo in OF-EVs from the early luteal phase and UF-EVs from the mid-luteal phase, coinciding with embryo transit within oviduct and uterus in vivo, and its possible influence on LmGs and sign-aling pathways crucial for early embryo development. A total of 333 miRNAs were detected, with 11 exclusive to OF, 59 to UF, and 263 were common between both groups. From the 20 differentially expressed miRNAs, 19 up-regulated in UF-EVs(bta-miR-134, bta-miR-151-3p, bta-miR-155, bta-miR-188, bta-miR-181b, bta-miR-181d, bta-miR-224, bta-miR-23b-3p, bta-miR-24-3p, bta-miR-27a-3p, bta-miR-29a, bta-miR-324, bta-miR-326, bta-miR-345-3p, bta-miR-410, bta-miR-652, bta-miR-677, bta-miR-873 and bta-miR-708) and one(bta-miR-148b) in OF-EVs. BACKGROUNd The sphericity of the femoral head is a metric used to evaluate hip pathologies and is associated with the development of osteoarthritis and femoral-acetabular *** To analyze the three-dimensional asphericity of the femoral head of asymptomatic pediatric *** hypothesized that femoral head asphericity will vary significantly between male and female pediatric hips and increase with age in both *** Computed tomography scans were obtained on 158 children and adolescents from a single institution in the United States(8-18 years;50%male)without hip *** femoral measurements including the femoral head diameter,femoral head volume,residual volume,asphericity index,and local diameter difference were used to evaluate femoral head *** in both sexes,the residual volume increased by age(Pdespite significantly smaller femoral head size in older ages(>13 years)in females,there were no sex-differences in residual volume and aspherity *** were no age-related changes in mean diameter difference in both sexes(P=0.07)with no significant sex-differences across different age groups(P=0.06).in contrast,there were significant increases in local aspherity(maximum diameter difference)across whole surface of the femoral head and all quadrants except the inferior regions in males(P=0.03).There were no sex-differences in maximum diameter difference at any regions and age group(P>0.05).increased alpha angle was only correlated to increased mean diameter difference across overall surface of the femoral head(P=0.024).CONCLUSiON There is a substantial localized asphericity in asymptomatic hips which increases with age *** 2d measured alpha angle can capture overall asphericity of the femoral head,it may not be sensitive enough to represent regional asphericity patterns.

The simultaneous investigation on the parameters affecting the flow of electrically conductive fluids such as volumetric radiation,heat absorption,heat generation,and magnetic field(mF)is very vital due to its existence in various sectors of industry and *** present research focuses on mathematical modeling to simulate the cooling of a hot component through power-law(PL)nanofluid convection *** temperature reduction of the hot component inside a two-dimensional(2d)inclined chamber with two different cold wall shapes is *** formulation of the problem is derived with the lattice Boltzmann method(LBm)by code writing via the FORTRAN *** variables such as the radiation parameter(0–1),the Hartmann number(0–75),the heat absorption/generation coefficient(−5–5),the fluid behavioral index(0.8–1.2),the Rayleigh number(103–105),the imposed mF angle(0°–90°),the chamber inclination angle(−90°–90°),and the cavity cold wall shape(smooth and curved)are *** findings indicate that the presence of radiation increases the mean Nusselt number value for the shear-thickening,Newtonian,and shear thinning fluids by about 6.2%,4%,and 2%,*** most cases,the presence of nanoparticles improves the heat transfer(HT)rate,especially in the cases where thermal conduction dominates *** is the lowest cooling performance index and mF effect for the cavity placed at an angle of 90°.The application in the design of electronic coolers and solar collectors is one of the practical cases of this numerical research.

Numerical modeling of mass Transfer in the interaction between River Biofilm and a Turbulent Boundary Layer

in this article dedicated to the modeling of vertical mass transfers between the biofilm and the bulk flow, we have, in the first instance, presented the methodology used, followed by the presentation of various results obtained through analyses conducted on velocity fields, different fluxes, and overall transfer coefficients. due to numerical constraints (resolution of relevant spatial scales), we have restricted the analysis to low Schmidt numbers (Sc=0.1, Sc=1, and Sc=10) and a single roughness Reynolds number (Re*=150). The analysis of instantaneous concentration fields from various simulations revealed logarithmic concentration profiles above the canopy. in this zone, the concentration is relatively homogeneous for longer times. The analysis of results also showed that the contribution of molecular diffusion to the total flux depends on the Schmidt number. This contribution is negligible for Schmidt numbers Sc≥0.1, but nearly balances the turbulent flux for Sc=0.1. in the canopy, the local Sherwood number, given by the ratio of the total flux (within or above the canopy) to the molecular diffusion flux at the wall, also depends on the Schmidt number and varies significantly between the canopy and the region above. The exchange velocity, a purely hydrodynamic parameter, is independent of the Schmidt number and is on the order of 10% of in the present case. This study also reveals that nutrient absorption by organisms near the wall depends on the Schmidt number. Such absorption is facilitated by lower Schmidt numbers.

Solar-assisted photocatalytic water splitting using defective UiO-66 solids from modulated synthesis
