Under global climate change,high and low temperature extremes can drive shifts in species *** the range of a species,thermal tolerance is based on acclimatization,plasticity,and may undergo selection,shaping resilience to temperature *** this study,we measured variation in cold temperature tolerance of early instar larvae of an invasive forest insect,Lymantria dispar dispar L.(Lepidoptera:Erebidae),using populations sourced from a range of climates within the current introduced range in the Eastern United *** tested for population differences in chill coma recovery(CCR)by measuring recovery time following a period of exposure to a nonlethal cold temperature in 2 cold exposure experiments.A 3rd experiment quantified growth responses after CCR to evaluate sublethal *** results indicate that cold tolerance is linked to regional climate,with individuals from populations sourced from colder climates recovering faster from chill *** this geographic gradient is seen in many species,detecting this pattern is notable for an introduced species founded from a single point-source *** demonstrate that the cold temperatures used in our experiments occur in nature during cold spells after spring egg hatch,but impacts to growth and survival appear *** expect that population differences in cold temperature performance manifest more from differences in temperature-dependent growth than acute *** intraspecific variation in cold tolerance increases our understanding of the role of climatic gradients on the physiology of an invasive species,and contributes to tools for predicting further expansion.

Endosymbiotic bacteria from donkeys milk in the inhibition of human pathogens

donkey milk is known for its beneficial aspects such as low fat and cholesterol, total protein and casein content, high lactose, whey protein, calcium, selenium and vitamin d3 content. It also has nutraceutical and utilitarian properties that can boost invulnerability, regulate digestion and conveniently change gut microbiota. The current study was focused on the effect of probiotic bacterial metabolites from donkey milk against human bacterial pathogens. The effective isolate VITRAAJ14 was found to be non-hemolytic and was tolerant to acid, NaCl and bile salt. Antibacterial activity by cross streak method showed maximum zone of inhibition. Well diffusion method also exhibited effective inhibition against Escherichia coli (MTCC No. 9721), Staphylococcus aureus (MTCC No. 3160) and Salmonella enterica (MTCC No. 8587) with fractions obtained from chloroform: ethyl acetate. The metabolites capable of inhibiting pathogens were identified as oleyl alcohol by Gas chromatography-mass spectrometry (GCMS). Lead compounds detected in GCMS was studied for its functional group by Fourier transform infrared spectroscopy (FTIR), which revealed the presence of alcoholic stretch, carboxylic acid OH stretch, alkene and amide bond. The isolate VITRAAJ14 was identified to be the closest neighbour of Bacillus species (MH100985) by 16S rRNA gene sequencing. To our knowledge, current study is the first-time report on the presence of Bacillus sp. from donkey milk showing antibacterial activity due to its ability to produce oleyl alcohol. Scanning electron microscopic images revealed the presence of rod-shaped morphology of the effective bacterium VITRAAJ14.

Clinical Analysis of the Colorectal Cohort within the Wales Cancer Biobank: A Study of Outcomes and Genetic Screening

Over the last 12 years, the Wales Cancer Biobank (WCB) has consented to more than 2000 patients with colorectal cancer (CRC). From these patients, clinical data has been collected and patients have been followed through their cancer journey. Clinical data from these patients have been analyzed to identify any correlation between disease grade and outcome. In a small cohort, consisting of 407 patients, WCB has performed genetic analysis on patient primary tumor samples, identifying and characterizing mutations in the KRAS, NRAS, BRAF, PIK3CA and TP53 genes. The majority of patients with CRC who were consented to WCB were male with a mean age of 69 years and received surgery as the primary treatment for their disease. Pathology and disease-free survival data confirmed worse prognoses associated with more advanced disease. Heterogeneity within the primary tumor was explored in a subgroup of patients. Analysis of the KRAS and TP53 genes confirmed that more than 40% of CRC patients who were tested, harbored a genetic mutation within these genes in their primary tumor. due to the limited sample size tested, most mutations did not show significant differences in disease-free survival, however, mutation of the BRAF gene did show a decrease in the disease specific survival, in keeping with the published data. Analysis of the patients diagnosed with CRC within the Biobank has provided us with valuable information on the status of CRC disease and treatment within the Welsh population. Over 12 years of consenting, we have witnessed significant changes in the information that researchers are interested in when sourcing samples for translational research. The development of new drugs that are tailored to the genetics of a cancer is emerging and at WCB we are focusing our collections on samples and data that meet the needs of this ever-evolving field.
