Relevance of proteolysis and proteasome activation in fatty liver graft preservation: An Institut Georges Lopez-1 vs University of Wisconsin appraisal

To compare liver proteolysis and proteasome activation in steatotic liver grafts conserved in University of Wisconsin (UW) and Institut Georges Lopez-1 (IGL-1) *** liver grafts from male obese Zücker rats were conserved in UW and IGL-1 solutions for 24 h at 4 °Cand subjected to “ex vivo” normo-thermic perfusion (2 h; 37 °C). Liver proteolysis in tissue specimens and perfusate was measured by reverse-phase high performance liquid chromatography. Total free amino acid release was correlated with the activation of the ubiquitin proteasome system (UPS: measured as chymotryptic-like activity and 20S and 19S proteasome), the prevention of liver injury (transaminases), mitochondrial injury (confocal microscopy) and inflammation markers (TNF 1 alpha, high mobility group box-1 (HGMB-1) and PPAR gamma), and liver apoptosis (TUNEL assay, cytochrome c and caspase 3).RESULTSProfiles of free AA (alanine, proline, leucine, isoleucine, methionine, lysine, ornithine, and threonine, among others) were similar for tissue and reperfusion effluent. In all cases, the IGL-1 solution showed a significantly higher prevention of proteolysis than UW (P d ischemia reperfusion. Livers conserved in IGL-1 presented more effective prevention of ATP-breakdown and more inhibition of UPS activity (measured as chymotryptic-like activity). In addition, the prevention of liver proteolysis and UPS activation correlated with the prevention of liver injury (AST/ALT) and mitochondrial damage (revealed by confocal microscopy findings) as well as with the prevention of inflammatory markers (TNF1alpha and HMGB) after reperfusion. Intracerebral hemorrhage （ICH）, caused by the sudden rupture of an artery within the brain, is a devastating subtype of stroke, which currently has no effective treatment. Intense inflammatory reactions that occur in the peri-hematomal area after ICH are more deleterious than the hematoma itself, resulting in subsequent brain edema and neurologic deterioration. Thus, we developed lipid-coated magnetic mesoporous silica nanoparticles doped with ceria nanoparticles （CeNPs）, abbreviated as LMCs, which have both potent anti-inflammatory therapeutic effects via scavenging reactive oxygen species and help in increasing the efficacy of magnetic resonance imaging enhancement in the peri-hematomal area. LMCs consist of mesoporous silica nanopartide-supported lipid bilayers, which are loaded with large amounts of CeNPs for scavenging of reactive oxygen species, and iron oxide nanoparticles for magnetic resonance imaging contrast. LMCs loaded with CeNPs exhibited strong anti-oxidative and anti-inflammatory activities in vitro. In the rodent ICH model, intracerebraUy injected LMCs reached the peri-hematomal area and were engulfed by macrophages, which were clearly visualized by magnetic resonance imaging of the brain. Moreover, LMCs reduced inflammatory macrophage infiltration, and thus significantly reduced brain edema. Finally, LMC treatment markedly improved neurologic outcomes of the animals with ICH. Thus, LMC is the first nanobiomaterial that successfully showed theragnostic effects in ICH.

Elevating the charge cut-off voltage beyond traditional 4.2 V is a commonly accepted technology to increase the energy density of Li-ion batteries(LIBs) but the risk of Li-dendrites and fire hazard increases as well. The use of ambi-functional additive, which forms stable solid electrolyte interphase(SEI) simultaneously at both cathode and anode, is a key to enabling a dendrites-free and well-working high-voltage LIB. Herein, a novel ambi-functional additive, pentaerythritol disulfate(PEdS), at 1 wt% without any other additive is demonstrated. We show the feasibility and high impacts of PEdS in forming lithium sulfateincorporated robust SEI layers at NCM523 cathode and graphite anode in 1 Ah-level pouch cell under4.4 V, 25 °C and 0.1 C rate, which mitigates the high-voltage instability, metal-dissolution and cracks on NCM523 particles, and prevents Li-dendrites at graphite anode. Improved capacity retention of 83%after 300 cycles is thereby achieved, with respect to 69% with base electrolyte, offering a promising path toward the design of practical high-energy LIBs.

In most sea surveillance radars based on non-coherent integration,the scan-to-scan integration which improves signal to noise ratio and mitigates unwanted echo with a small correlation time is widely used to extract stationary and slow moving *** problems of the conventional scan-to-scan integration are:（a）the target tail is generated through integrating previous scans and current scan with respect to a fast moving target which has a high radar cross section;（b）it is difficult to detect a small and fast moving target due to the attenuated small target *** this contribution,the problems of the conventional scan-to-scan integration are investigated carefully by using time series analysis,and then an efficient scan-to-scan integration/correlation algorithm is proposed to overcome the problems of the existing *** the proposed algorithm,the signal selection logic eliminating the unwanted target tail and recovering the attenuated target echo has been operated according to the difference between the current and previous scans with two *** results demonstrate the proposed algorithm’s performance of detecting a small and fast moving target without the target tail effect.

Enhanced 3d Point Cloud Reconstruction for Light Field Microscopy Using U-Net-Based Convolutional Neural Networks

This article describes a novel approach for enhancing the three-dimensional(3d)point cloud reconstruction for light field microscopy(LFM)using U-net architecture-based fully convolutional neural network(CNN).Since the directional view of the LFM is limited,noise and artifacts make it difficult to reconstruct the exact shape of 3d point *** existing methods suffer from these problems due to the self-occlusion of the *** manuscript proposes a deep fusion learning(dL)method that combines a 3d CNN with a U-Net-based model as a feature *** sub-aperture images obtained from the light field microscopy are aligned to form a light field data cube for preprocessing.A multi-stream 3d CNNs and U-net architecture are applied to obtain the depth feature fromthe directional sub-aperture LF data *** the enhancement of the depthmap,dual iteration-based weighted median filtering(WMF)is used to reduce surface noise and enhance the accuracy of the *** a 3d point cloud involves combining two key elements:the enhanced depth map and the central view of the light field *** proposed method is validated using synthesized Heidelberg Collaboratory for Image Processing(HCI)and real-world LFM *** results are compared with different state-of-the-art *** structural similarity index(SSIM)gain for boxes,cotton,pillow,and pens are 0.9760,0.9806,0.9940,and 0.9907,***,the discrete entropy(dE)value for LFM depth maps exhibited better performance than other existing methods.

Effect of Pectin Lyase Enzyme on Fermentation and drying of Cocoa (Theobroma cacao L.): An Alternative to Improve Raw Material in the Industry of Chocolate

Cocoa (Theobroma cacao), in all its presentations, is consumed all over the world and is one of the main drivers of the economic in several countries. The world’s Cocoa tendency is focused on developing special beans. This category is subject to postharvest processes of utmost importance such as the fermentation and dry, which are currently carried out with traditional and poorly effective devices, which need to be improved to obtain a high quality product. The aim of this study was to evaluate the influence of the pectin lyase enzyme (E.C.4.***.2.10) on the postharvest cocoa process. We evaluated the enzyme dosage (1.0% and 0.5%) in fermentation and its effect on the variables temperature, acidity and drying time by convection at 60°C. The Pectin lyase activity during fermentation does not cause a significant effect on the variables of temperature and acidity;however, the drying process time required to achieve 7.0% moisture was reduced. The enzyme dosage of 1.0% was the best result, the amount of exudate obtained (115 ml) during fermentation and the best degree of fermentation (77% ± 3.8) were increased and further shows a change in porosity facilitating the scale surface and internal moisture diffusion. The drying rate (Nw) expressed in kgwater/m2 * min was determined based on the empirical model of Newton, where the higher speed was obtained during the falling period. In conclusion, enzyme dosage 1% was the best concentration evaluated because weaken grain husk, which allowed an adequate fermentation,and subsequent time drying reduction until 10.8 h.

Inflammatory myofibroblastic tumor （INT） of the stomach in adults is extremely rare, with unpredictable prognosis. We present a 55-year-old woman with a gastric IMT. She experienced sudden abdominal pain 4 d previously. Physical examination showed mild abdominal tenderness in the hypogastrium, but no palpable abnormal abdominal mass. Abdominal CT showed a mass of approximately 8 cm in the gastrocolic ligament. On laparoscopic exploration, unexpected hemoperitoneum of approximately 1.5 L of blood was found, and an exophytic gastric mass of approximately 10 cm, appeared from the anterior wall of the gastric body along the greater curvature. Laparoscopy further showed that non- clotting blood in the abdominal cavity seemed to be from the gastric tumor. After conversion to open surgery for more precise evaluation of the cause of hemoperitoneum and the large friable tumor, gastric wedge resection, including the tumor, was conducted. The final diagnosis was consistent with IMT that originated from the gastric wall.

BACKGROUNd The debate regarding diagnostic classification systems in psychiatry(categorial vs dimensional systems)has essential implications for the diagnosis,prevention and treatment of stress *** previously found a unique pattern of stress reaction in a study executed during the coronavirus disease 2019 pandemic using large representative samples in two countries,and termed it the Complex Stress Reaction Syndrome(CSRS).AIM To investigate CSRS,Type A(psychiatric symptoms,spanning anxiety,depression,stress symptoms,and posttraumatic stress disorder(PTSd)),with or without long-coronavirus disease(COVId)residuals(CSRS,Type B,neuropsychiatric symptoms spanning cognitive deficits and fatigue,excluding systemic symptoms).Our two-tailed hypothesis was that CSRS is a condition related to an unrecognized type of stress reaction in daily life in the general population(Type A)or that it is related to the severe acute respiratory syndrome coronavirus 2 infection and its long-COVId residuals(Type B).METHOdS 977 individuals in four continents(North America,Europe,Australia and the Middle East)completed the online study questionnaire in six languages using the Qualtrics *** study was managed by six teams in six countries that promoted the study on social *** questionnaire assessed anxiety,depression,stress symptoms and PTSd(CSRS,Type A),cognitive deficits and fatigue(CSRS,Type B).The data were analyzed using Proportion Analyses,Multivariate Analysis of Co-Variance(MANCOVA),linear regression analyses and validated clinical cutoff *** The results of the Proportion Analyses showed that the prevalence of 4 symptoms spanning anxiety,depression,stress symptoms,and PTSd was significantly higher than the most prevalent combinations of fewer symptoms across 4 continents,age groups,and *** supports the transdiagnostic argument embedded in the CSRS(Type A).The same pattern of results was found in infected/recovered *** prevalence of the

Growing evidence suggests pollution and other environmental factors have a role in the development of tuberculosis(TB),however,such studies have never been conducted in *** the association between air pollution and specific geographic areas,our objective was to determine the spatial distribution and clustering of TB incident cases in Lima and their co-occurrence with clusters of fine particulate matter(PM2.5)and *** found co-occurrences of clusters of elevated concentrations of air pollutants such as PM2.5,high poverty indexes,and high TB incidence in *** findings suggest an interplay of socio-economic and environmental in driving TB incidence.

A signal-plus-noise （S+N ） energy selection combining （S+N-ESC） scheme for binary phase shift keying （BPSK） signaling is proposed. The S+N-ESC selects the branch with the maximum S+N energy for detection. The advantage of the S+N-ESC is that it requires no channel estimation for diversity branch selection. The bit error rate （BER） performance of the S+N-ESC is analyzed for BPSK signaling in independent and identically distributed （i.i.d.） Rayleigh fading channels. We show that the performance of the S+N-ESC is the same as conventional signal-to-noise ratio selection combining （SNR-SC）,which requires estimation of the fading amplitudes for branch selection.
