A study of the NW Kakamas domain in South Africa/Namibia provides a new,unified lithostratigraphy and evolutionary history applicable to the whole Namaqua *** Mesoproterozoic history ranges from~1350 Ma to 960 Ma,but isotopic evidence suggests it was built upon pre-existing Paleoproterozoic continental crust that extended west from the Archaean *** eastern Namaqualand,early rift-related magmatism and sedimentation at~1350 Ma occurred in a confined ocean *** tectonic reversal and subduction at~1290–1240 Ma led to establishment of the Areachap,Konkiep and Kaaien *** the Kakamas domain,widespread deposition of pelitic sediments occurred at~1220 Ma(Narries Group).These contain detrital zircons derived from proximal crust with ages between~2020 Ma and 1800 Ma(western Palaeoproterozoic domains)and 1350–1240 Ma(eastern early Namaqua domains),suggesting pre-sedimentation *** pelites underwent granulite grade metamorphism at~1210 Ma(peak conditions:4.5–6 kbar and 770–850°C),associated with voluminous,predominantly S-type granitoid orthogneisses between~1210 Ma and 1190 Ma(Eendoorn and Ham River Suites)and low-angle ductile(d_(2))deformation which continued until~1110 Ma,interspersed with periods of *** enduring P-T regime is inconsistent with the expected crustal over-thickening associated with the generally-accepted collision-accretion Namaqualand ***,we propose the Namaqua Sector is a‘hot orogen’developed in a wide continental back-arc with subduction west of the present-day *** observed high geotherm resulted from thinned back-arc lithosphere accompanied by an influx of mantle-derived *** d_(2)deformation resulted from"bottom-driven"tectonics and viscous drag within the crust by convective flow in the underlying asthenospheric *** extended tectonothermal regime ceased at~1110 Ma when SW-directed thrusting stacked the Namaqua domains into their current positions,constrained in

Streamlining the preparation of “endotoxin-free” ClearColi cell extract with autoinduction media for cell-free protein synthesis of the therapeutic protein crisantaspase

An“endotoxin-free”***-based cell-free protein synthesis system has been reported to produce therapeutic proteins rapidly and ***,preparation of the most complex CFPS reagent–the cell extract–remains time-consuming and labor-intensive because of the relatively slow growth kinetics of the endotoxin-free ClearColiTMBL21(dE3)*** we report a streamlined procedure for preparing *** cell extract from ClearColi™using auto-induction *** this work,the term auto-induction describes cell culture media which eliminates the need for manual induction of protein *** Clearcoli™cells in autoinduction media significantly reduces the hands-on time required during extract preparation,and the resulting“endotoxinfree”cell extract maintained the same cell-free protein synthesis capability as extract produced with traditional induction as demonstrated by the high-yield expression of crisantaspase,an FdA approved leukemia *** is anticipated that this work will lower the barrier for researchers to enter the field and use this technology as the method to produce endotoxin-free ***-based extract for CFPS.

The mode of abscisic acid （ABA） action, and its relations to drought adaptive responses in particular, has been a captivating area of plant hormone research for much over a decade. The hormone triggers stomatal closure to limit water loss through transpiration, as well as mobilizes a battery of genes that presumably serve to protect the cells from the ensuing oxidative damage in prolonged stress. The signaling network orchestrating these various responses is, however, highly complex. This review summarizes several significant advances made within the last few years. The biosynthetic pathway of the hormone is now almost completely elucidated, with the latest identification of the ABA4 gene encoding a neoxanthin synthase, which seems essential for de novo ABA biosynthesis during water stress. This leads to the interesting question on how ABA is then delivered to perception sites. In this respect, regulated transport has attracted renewed focus by the unexpected finding of a shoot-to-root translocation of ABA during drought response, and at the cellular level, by the identification of a B-galactosidase that releases biologically active ABA from inactive ABA-glucose ester. Surprising candidate ABA receptors were also identified in the form of the Flowering Time Control Protein A （FCA） and the Chloroplastic Magnesium Protoporphyrin-IX Chelatase H subunit （CHLH） in chloroplast-nucleus communication, both of which have been shown to bind ABA in vitro. On the other hand, the protein（s） corresponding to the physiologically detectable cell-surface ABA receptor（s） is （are） still not known with certainty. Genetic and physiological studies based on the guard cell have reinforced the central importance of reversible phosphorylation in modulating rapid ABA responses. In a context of growing efforts to develop sustainability strategies, energy-related issues occupy central stage in the built environment. Thus, the energy performance of housings has improved radically over the past decades. Yet other types of buildings, in particular commercial centers, haven’t received the same level of interest. As a result, there is a need for effective and practical measures to decrease their energy consumption, both for heating and electricity. The objective of the paper is to demonstrate that it is possible, through coherent strategies, to integrate energy issues and bioclimatic principles into the design process of commercial centers. It analyzes the exemplary case study of Marin Commercial Center (Switzerland). The interdisciplinary approach, based on integrated design strategies, aimed at increasing the energy efficiency while keeping the cost comparable to the market cost. The main design principles include natural ventilation, nighttime cooling with energy recovery and natural lighting, as well as optimization of mechanical systems. The results of the simulations show that Marin Center attains the best energy performance observed so far among Swiss commercial centers. It also meets the Swiss Minergie standard. The paper thus questions traditional design processes and outlines the need for interdisciplinary evaluation and monitoring approaches tailored for commercial centers. Even though most crucial decisions are taken during the early stages, all phases of the process require systematic optimization strategies, especially operating stages. Recommendations include legal measures, in particular in the fields of ventilation and air-conditioning, education, professional development and technology transfer, and financial incentives for the replacement of energy intensive installations.

Strigolactones （SLs） are known not only as plant hormones, but also as rhizosphere signals for establishing symbiotic and parasitic interactions. The design of new specific SL analogs is a challenging goal in understanding the basic plant biology and is also useful to control plant architectures without favoring the development of parasitic plants. Two different molecules （23 （3＇-methyI-GR24）, 31 （thia-3＇-methyl-debranone-like molecule）） already described, and a new one （AR36）, for which the synthesis is presented, are biologically compared with the well-known GR24 and the recently identified CISA-1. These different structures emphasize the wide range of parts attached to the d-ring for the bioactivity as a plant hormone. These new compounds possess a common dimethylbutenolide motif but their structure varies in the ABC part of the molecules： 23 has the same ABC part as GR24, while 31 and AR36 carry, respectively, an aromatic ring and an acyclic carbon chain. detailed information is given for the bioactivity of such derivatives in strigolactone synthesis or in perception mutant plants （pea rmsl and rms4, Arabidopsis max2 and, max4） for different hormonal functions along with their action in the rhizosphere on arbuscular mycorrhizal hyphal growth and parasitic weed germination.

In plants, phosphate (Pi) homeostasis is regulated by the interaction of PHR transcription factors with stand-alone SPX proteins, which act as sensors for inositol pyrophosphates. Here, we combined different methods to obtain a comprehensive picture of how inositol (pyro)phosphate metabolism is regulated by Pi and dependent on the inositol phosphate kinase ITPK1. We found that inositol pyrophosphates are more responsive to Pi than lower inositol phosphates, a response conserved across kingdoms. With CE-ESI-MS we could separate different InsP7 isomers in Arabidopsis and rice, and identify 4/6-InsP7 and a PP-InsP4 isomer hitherto not reported in plants. We found that the inositol pyrophosphates 1/3-InsP7, 5-InsP7 and InsP8 increase severalfold in shoots after Pi resupply and that tissue-specific accumulation of inositol pyrophosphates relies on ITPK1 activities and MRP5-dependent InsP6 compartmentalization. Notably, ITPK1 is critical for Pi-dependent 5-InsP7 and InsP8 synthesis in planta and its activity regulates Pi starvation responses in a PHR-dependent manner. Furthermore, we demonstrate that ITPK1-mediated conversion of InsP6 to 5-InsP7 requires high ATP concentrations and that Arabidopsis ITPK1 has an AdP phosphotransferase activity to dephosphorylate specifically 5-InsP7 under low ATP. Collectively, our study provides deeper insights into Pi-dependent changes in nutritional and energetic states with the synthesis of regulatory inositol pyrophosphates.
