Effect of tubastatin A on NLRP3 inflammasome activation in macrophages under hypoxia/ reoxygenation conditions

BACKGROUNd:There are currently no effective drugs to mitigate the ischemia/reperfusion injury caused by fluid resuscitation after hemorrhagic shock(HS).The aim of this study was to explore the potential of the histone deacetylase 6(HdAC6)-specific inhibitor tubastatin A(TubA)to suppress nucleotide-binding oligomerization domain-like receptor protein 3(NLRP3)inflammasome activation in macrophages under hypoxia/reoxygenation(H/R)***:The viability of RAW264.7 cells subjected to H/R after treatment with different concentrations of TubA was assessed using a cell-counting kit-8(CCK8)***,2.5μmol/L TubA was used with RAW264.7 cells under H/R ***264.7 cells were divided into three groups,namely the control,H/R,and TubA *** levels of reactive oxygen species(ROS)in the cells were detected using fluorescence *** protein expression of HdAC6,heat shock protein 90(Hsp90),inducible nitric oxide synthase(iNOS),NLRP3,gasdermin-d(GSdMd),Caspase-1,GSdMd-N,and Caspase-1 p20 was detected by western *** levels of interleukin-1β(IL-1β)and IL-18 in the supernatants were detected using enzyme-linked immunosorbent assay(ELISA).RESULTS:HdAC6,Hsp90,and iNOS expression levels were significantly higher(Py higher(Py reduced in the TubA group(Pd higher NLRP3,GSdMd,Caspase-1,GSdMd-N,and Caspase-1 p20 expression levels than the control group(Pd significantly lower expression levels than the H/R group(Pd IL-18 levels in the supernatants were significantly higher in the H/R group compared to the control group(Py lower in the TubA group compared to the H/R group(Pd the expression of HdAC6,Hsp90,and iNOS in macrophages

A Comprehensive Study on the Consequences of Gap-Graded Sands Considering the Loss of Fine Particles

Water and sand leakage disasters are likely to occur during construction in water-rich sand layer areas,resulting in ground *** stress-strain action characteristics of discontinuous graded sand under different internal erosion degrees,and the evolution mechanism of water and sand leakage disasters caused by the internal erosion need to be further ***,this paper takes the discontinuous graded sand in a water rich sand layer area in Nanchang City of China as the research *** the influence of different fine particle losses(0,10%,20%and 30%)under the internal erosion of sand,the salt solution method is used to realize the specified loss of fine particles in the internal *** stress-strain behavior after the loss of fine particles due to internal erosion is studied by triaxial shear ***,the physical model test and PFC-CFd method are both used to study the evolution rules of water and sand leakage disaster considered the influence of internal erosion *** show that:(1)under the same confining pressure,the peak failure strength of sand samples decreases along with the increase of fine particle loss.(2)In the water and sand leakage test of saturated sand,a natural filter channel is formed above the observed soil *** greater the loss of fine particles,the steeper and wider the collapse settlement area.(3)The relationship between the cumulative amount of water and sand leakage and time is *** total mass loss of sand increases along with the increase of internal erosion degree.(4)After the soil arch is formed around the damaged opening,the sand continues to converge above the soil arch under the action of water flow,resulting in the dense convergence of contact force chains.

dietary copper requirement of broilers fed a corn-soybean meal diet during 22—42 d of age

This research was to assess the dietary copper(Cu)requirement of broiler chickens fed a practical cornsoybean meal diet during 22—42 d of age.A total of 288 numbered Arbor Acres male broilers at 22 d of age were randomly allotted 6 treatments with 8 replicate cages(6 broilers per cage)per *** were fed a Cu-unsupplemented corn-soybean meal basal diet(control,containing 7.36 mg Cu/kg)or the basal diet added with 3,6,9,12,or 15 mg Cu/kg from CuSO_(4)·5H_(2)O for 21 ***,asymptotic and broken-line models were fitted and the best fitted models were selected to determine dietary Cu *** results revealed that the contents of Cu in serum and liver,m RNA expression levels of Cu-and zinc-containing superoxide dismutase(CuZnSOd)in liver and monoamine oxidase b(MAO B)in heart,as well as protein expression level of CuZnSOd in liver were affected(Py supplemental Cu levels,and the above indices varied linearly and quadratically(Pd according to the best fitted broken-line models(Pdexes were 10.45—13.81 mg/*** was concluded that mRNA expression levels of CuZnSOd in liver and MAO B in heart,as well as liver CuZnSOd protein expression level were new specific sensitive biomarkers for estimating dietary Cu requirements,and the dietary Cu requirement was recommended to be 14 mg/kg to support Cu metabolic needs related to key Cucontaining enzymes in broilers fed the corn-soybean meal diet during 22—42 d of age,which was higher than the dietary Cu requirement(8 mg/kg)for broilers at the corresponding stage suggested by the Chinese Feeding Standard of Chicken.

Luminescence properties and optical sensing behaviors of Sr_(2)GdSbO_(6):Eu^(3+) phosphors

Effects of inorganic nutrients and environmental factors on the removal of n-propylbenzene and isopropylbenzene in seawater by cryptophytes Rhinomonas reticulata S6A

To effectively remove n-propylbenzene(n-PBZ)and isopropylbenzene(i-PBZ)leaked into seawater using Rhinomonas reticulata S6A(a newly isolated marine microalga),the effects of three inorganic nutrients and four environmental factors on their degradation were determined after 7 d of *** show that NaNO_(3) at 300 mg/L caused a higher removal efficiency of both n-PBZ and i-PBZ(44.79%and 39.26%),while for NaH_(2) PO_(4)·H_(2) O,greater removal rates of two PBZs(47.30%and 42.23%)were achieved at 30 and 20 mg/L,***_(3) supplementation(500-750 mg/L)resulted in a large reduction(43.67%-45.04%)in i-PBZ *** change in seawater pH(from 6 to 9)did not affect the elimination of n-PBZ and *** most suitable salinity and temperature were 30 and 25-30℃,respectively,leading to the PBZs removal of~40%.Light intensity exhibited significant influence on elimination of PBZs,and the maximum removal efficiencies of 56.07%(n-PBZ)and 55.00%(i-PBZ)were recorded under 200 and 600μmol/(m^(2)·s),*** addition,the microalga could still remove PBZs when it failed to grow well due to darkness,strong light,low temperature,or low salinity,which might mean that good growth of alga is not always a necessary condition for PBZs ***,attention should be paid to the suitability of nutrient levels and environmental conditions(excluding pH)in seawater when using microalgae for bioremediating PBZs-contaminated seawater.
