Recent data indicate that IGF1R/IRS signaling is a potential therapeutic target in BCR-ABL1-negative myeloproliferative neoplasms(MPN);in this pathway,IRS2 is involved in the malignant transformation induced by JAK2^(V617F),and upregulation of IGF1R signaling induces the MPN ***157,a synthetic compound designed as an IGF1R-IRS1/2 inhibitor,has been shown to induce antineoplastic effects in solid ***,we aimed to characterize the molecular and cellular effects of NT157 in JAK2^(V617F)positive MPN cell lines(HeL and SeT2)and primary patient hematopoietic *** JAK2^(V617F)cell lines,NT157 decreased cell viability,clonogenicity,and cell proliferation,resulting in increases in apoptosis and cell cycle arrest in the G2/M phase(peatment inhibited IRS1/2,JAK2/STAT,and NFκB signaling,and it activated the AP-1 complex,downregulated four oncogenes(CCND1,MYB,WT1,and NFKB1),and upregulated three apoptotic-related genes(CDKN1A,FOS,and JUN)(ped genotoxic stress in a JAK2/STAT-independent ***157 inhibited erythropoietin-independent colony formation in cells from polycythemia vera patients(pese findings further elucidate the mechanism of NT157 action in a MPN context and suggest that targeting IRS1/2 proteins may represent a promising therapeutic strategy for MPN.

Amine-rich carbon nitride nanoparticles:Synthesis,covalent functionalization with proteins and application in a fluorescence quenching assay

Carbon nitride nanoparticles (CNNPs) have been employed as fluorescent sensing tools owing to their unique features, e.g. low cost production, high stability in water and high photoluminescence quantum yield. Here, an easy and versatile synthetic approach was exploited to design fluorescent nanoparticles with surface functionalities suitable for covalent binding to bioligands. High hydrophilic, brightly fluorescent CNNPs, rich of superficial amines, were obtained from the thermal condensation of urea and lysine (CNNP^Lys) and by tuning the precursor ratio and the heating time. Structure and size of the functionalized nanoparticles were characterized through infrared (IR) spectroscopy, transmission electron microscopy (TeM) and dynamic light scattering (DLS). Their optical properties were studied by ultraviolet-visible (UV-Vis) and fluorescence spectroscopy. The superficial primary amino groups, furnished by the lysine co-precursor, enabled for covalently linking CNNP^Lys to model proteins. The CNNP^Lys-protein conjugates excited under UV irradiation emit in the 400-450 nm visible range (quantum yield 24%). The applicability of CNNP^Lys as novel fluorescent probes was demonstrated by a fluorescence quenching assay, in which gold nanoparticles (GNPs) were attached to Staphylococcal protein A and employed to quench the CNNP^Lys fluorescence by Forster resonant energy transfer (FReT). The quenching occurred upon formation of the specific binding between the GNP-linked protein A and CNNP^Lys-tagged immunoglobulins, while the inhibition of the binding resulted in the recovery of CNNP^Lys luminescence. The synthetic strategy, based on combining a "conjugated polymer"-forming unit (urea) and a co-precursor able to provide the desired functional group (lysine), allows designing innovative materials for the development of new generation fluorescence biosensors in which easily functionalized fluorophores are needed.

In autoparasitoids, females are generally primary endoparasitoids of Hemiptera, while males are hyperparasitoids developing in or on conspecific females or other primary parasitoids. Female-host acceptance can be influenced by extrinsic and/or intrinsic factors. In this paper, we are concerned with intrinsic factors such as nutritional status, mating status, etc. We observed the behavior of encarsia pergandiella Howard （Hymenoptera： Aphelinidae） females when parasitizing primary （3rd instar larvae ofBemisia tabaci Gen- nadius [Homoptera： Aleyrodidae]） and secondary hosts （3rd instar larvae and pupae of eretmocerus rnundus Mercet [Hymenoptera： Aphelinidae]） for a period of 1 h. Females had different reproductive （virgin or mated younger） and physiological （fed elder or mated elder） status. Virgin females killed a large number of secondary hosts while investing a long time per host. However, they did not feed upon them. Mated females killed a lower number of secondary hosts and host feeding was observed in both consuming primary and secondary hosts. It was common to observe host examining females of all physiological statues tested repeatedly stinging the same hosts when parasitizing, killing or rejecting them. Fed elder females parasitized more B. tabaci larvae than e. mundus larvae or pupae, while investing less time on the primary host than on the secondary host. They also para- sitized more B. tabaci larvae than mated elder females, while investing less time per host. The access of females to honey allowed them to lay more eggs.

Bloating and functional gastro-intestinal disorders: Where are we and where are we going?

Bloating is one of the most common and bothersome symptoms complained by a large proportion of patients. This symptom has been described with various definitions, such as sensation of a distended abdomen or an abdominal tension or even excessive gas in the abdomen, although bloating should probably be defined as the feeling (e.g. a subjective sensation) of increased pressure within the abdomen. It is usually associated with functional gastrointestinal disorders, like irritable bowel syndrome, but when bloating is not part of another functional bowel or gastrointestinal disorder it is included as an independent entity in Rome III criteria named functional bloating. In terms of diagnosis, major difficulties are due to the lack of measurable parameters to assess and grade this symptom. In addition, it is still unclear to what extent the individual patient complaint of subjective bloating correlates with the objective evidence of abdominal distension. In fact, despite its clinical, social and economic relevance, bloating lacks a clear pathophysiology explanation, and an effective management endorsement, turning this common symptom into a true challenge for both patients and clinicians. Different theories on bloating etiology call into questions an increased luminal contents (gas, stools, liquid or fat) and/or an impaired abdominal empting and/or an altered intra-abdominal volume displacement (abdomino-phrenic theory) and/or an increased perception of intestinal stimuli with a subsequent use of empirical treatments (diet modifications, antibiotics and/or probiotics, prokinetic drugs, antispasmodics, gas reducing agents and tricyclic antidepressants). In this review, our aim was to review the latest knowledge on bloating physiopathology and therapeutic options trying to shed lights on those processes where a clinician could intervene to modify disease course.

Bone marrow derived stem cells for the treatment of end-stage liver disease

end-stage disease due to liver cirrhosis is an important cause of death worldwide. Cirrhosis results from progressive, extensive fibrosis and impaired hepatocyte regeneration. The only curative treatment is liver transplantation, but due to the several limitations of this procedure, the interest in alternative therapeutic strategies is increasing. In particular, the potential of bone marrow stem cell(BMSC) therapy in cirrhosis has been explored in different trials. In this article, we evaluate the results of 18 prospective clinical trials, and we provide a descriptive overview of recent advances in the research on hepatic regenerative medicine. The main message from the currently available data in the literature is that BMSC therapy is extremely promising in the context of liver cirrhosis. However, its application should be further explored in randomized, controlled trials with large cohorts and long follow-ups.

Performance of Geometric Multigrid Method for Two-Dimensional Burgers’equations with Non-Orthogonal,Structured Curvilinear Grids

evaluating the influences of fine-scale habitat heterogeneity on the composition,diversity,structure and functioning of forests is critical to understand how tropical forests will respond to climate change and devise forest management strategies that will enhance biodiversity conservation and aboveground biomass ***,we hypothesized that topographic and soil factors determine fine-scale habitat differentiation,which in turn shape community composition,species richness,structure and aboveground biomass at the local scale in tropical *** test this hypothesis,we selected two areas(each 100×100 m)with contrasting fine-scale topographic conditions where all trees,palms and lianas with a diameter at breast height ≥ 10 cm were tagged and identified to *** each selected area,100 subplots of 10×10 m were *** mainly found that higher topographic variability caused higher habitat differentiation with changes in species composition and community structure,but did not change species *** habitat-scale analyses indicated that,in the less heterogeneous area,the distribution of species was more uniform along a fine-scale topographical gradient with no variation in convexity,which induced changes in structure and aboveground biomass,but not in species *** nonsignificant relationship between species richness and aboveground biomass may be attributable to species redundancy or functional *** study suggests that environmental filtering is a fundamental process for shaping community assembly and forest functioning along a local topographical gradient in tropical forests.

Mindfulness interventions during pregnancy:A narrative review

Pregnancy is a period of major transformations in a woman’s life;increased stress,and mood and sleep disorders are *** review evaluates mindfulness interventions during pregnancy and their ability to help manage stress,anxiety,depression,emotional regulation,level of mindfulness and sleep quality.A search of english language scientific literature relevant to mindfulness interventions for pregnant women was conducted using PubMed,Scopus and Web of Science,without restriction on publication *** criteria were randomized clinical trials with pregnant women,using mindfulness as an intervention for at least three weeks,in one of our main areas of interest,and using only validated scales to measure *** hundred and thirty studies were identified in our searches of research databases,and thirteen were included in our *** found a large diversity of mindfulness programs,heterogeneity among the instruments used to evaluate outcomes,and inconsistency in the gestational periods used in the *** interventions were beneficial for stress,anxiety and *** was also effective when applied in pregnant women with a history of depression or experiencing *** emotional regulation and the level of mindfulness,there were signs of improvement,but more studies are *** of the studies evaluated sleep *** review provides information about current mindfulness programs,an overview of the effects of mindfulness interventions,a description of the measurements used so far,and recommendations for developing highquality mindfulness protocols for pregnant women.

Arabidopsis MSI1 Is Required for Negative Regulation of the Response to Drought Stress

Arabidopsis MSI1 has fundamental functions in plant development. MSI1 is a subunit of Polycomb group protein complexes and Chromatin assembly factor 1, and it interacts with the Retinoblastoma-related protein 1. Altered levels of MSI1 result in pleiotropic phenotypes, reflecting the complexity of MSI1 protein functions. In order to uncover additional functions of MSI1, we performed transcriptional profiling of wild-type and plants with highly reduced MSI1 levels （msil-cs）. Surprisingly, the known functions of MSI1 could only account for a minor part of the transcriptional changes in msil-cs plants. One of the most striking unexpected observations was the up-regulation of a subset of ABA-responsive genes eliciting the response to drought and salt stress. We report that MSI1 can bind to the chromatin of the drought-inducible downstream target RD20 and suggest a new role for MSI1 in the negative regulation of the Arabidopsis drought-stress response.
