Murine models based on acute myeloid leukemia-initiating stem cells xenografting

Acute myeloid leukemia(AML) is an aggressive malignant disease defined by abnormal expansion of myeloid blasts. Despite recent advances in understanding AML pathogenesis and identifying their molecular subtypes based on somatic mutations, AML is still characterized by poor outcomes, with a 5-year survival rate of only 30%-40%, the majority of the patients dying due to AML relapse. Leukemia stem cells(LSC) are considered to be at the root of chemotherapeutic resistance and AML relapse. Although numerous studies have tried to better characterize LSCs in terms of surface and molecular markers, a specific marker of LSC has not been found, and still the most universally accepted phenotypic signature remains the surface antigens CD34+CD38- that is shared with normal hematopoietic stem cells. Animal models provides the means to investigate the factors responsible for leukemic transformation, the intrinsic differences between secondary post-myeloproliferative neoplasm AML and de novo AML, especially the signaling pathways involved in inflammation and hematopoiesis. However, AML proved to be one of the hematological malignancies that is difficult to engraft even in the most immunodeficient mice strains, and numerous ongoing attempts are focused to develop "humanized mice" that can support the engraftment of LSC. This present review is aiming to in-troduce the field of AML pathogenesis and the concept of LSC, to present the current knowledge on leukemic blasts surface markers and recent attempts to develop best AML animal models.

Borderline resectable pancreatic cancer: Definitions and management

Pancreatic cancer is the fourth leading cause of cancer death in the United States. While surgical resection remains the only curative option, more than 80% of patients present with unresectable disease. Unfortunately, even among those who undergo resection, the reported median survival is 15-23 mo, with a 5-year survival of approximately 20%. Disappointingly, over the past several decades, despite improvements in diagnostic imaging, surgical technique and chemotherapeutic options, only modest improvements in survival have been realized. Nevertheless, it remains clear that surgical resection is a prerequisite for achieving longterm survival and cure. There is now emerging consensus that a subgroup of patients, previously considered poor candidates for resection because of the relationship of their primary tumor to surrounding vasculature, may benefit from resection, particularly when preceded by neoadjuvant therapy. This stage of disease, termed borderline resectable pancreatic cancer, has become of increasing interest and is now the focus of a multiinstitutional clinical trial. Here we outline the history, progress, current treatment recommendations, and future directions for research in borderline resectable pancreatic cancer.

Monotonic, cyclic and post-cyclic performances of single-helix anchor in residual soil of sandstone

Helical anchors are commonly used in Brazil for guyed transmission towers subjected to static and cyclic wind *** most cases,these anchors are installed in tropical residual soil,a micro-structured material in which the shear strength is provided by soil *** installation of a helical anchor,as the helical plate moves downward into the ground,the soil penetrated is sheared and ***,in this type of soil,anchor installation affects the soil shear strength significantly associated with a bonded ***,the cyclic responses of helical anchors in this type of structured soils are rarely *** address this problem,tests were conducted in a Brazilian residual soil to investigate the monotonic,cyclic and post-cyclic performances of single-helix *** tests used two instrumented single-helix anchors installed in this typical residual soil of sandstone,which is frequently observed in large areas in the southern *** testing results indicate that the disturbance caused by the anchor installation affected the monotonic uplift performance *** results of cyclic loading tests also show no significant degradation of helix bearing resistance and reduced displacement accumulation with increasing load *** is perhaps due to the soil improvement caused by previous loading,which then increases the stiffness response of the anchor.

Feature reduction with PCA/ KPCA for gait classification with different assistive devices

Purpose–The purpose of this paper is to propose a gait analysis technique that aims to identify differences and similarities in gait performance between three different assistive devices(ADs).Design/methodology/approach–Two feature reduction techniques,linear principal component analysis(PCA)and nonlinear kernel-PCA(KPCA),are expanded to provide a comparison of the spatio-temporal,symmetrical indexes and postural control parameters among the three different ***,a multiclass support vector machine(MSVM)with different approaches is designed to evaluate the potential of PCA and KPCA to extract relevant gait features that can differentiate between ***–Results demonstrated that symmetrical indexes and postural control parameters are better suited to provide useful information about the different gait patterns that total knee arthroplasty(TKA)patients present when walking with different *** combination of KPCA and MSVM with discriminant functions(MSVM DF)resulted in a noticeably improved *** combination demonstrated that,with symmetric indexes and postural control parameters,it is possible to extract with high-accuracy nonlinear gait features for automatic classification of gait patterns with ***/value–The information obtained with the proposed technique could be used to identify benefits and limitations of ADs on the rehabilitation process and to evaluate the benefit of their use in TKA patients.

The advent of genomic big data and the statistical need for reaching significant results have led genome-wide association studies to be ravenous of a huge number of genetic markers scattered along the whole *** its very beginning,the so-called genotype imputation served this purpose;this statistical and inferential procedure based on a known reference panel opened the theoretical possibility to extend association analyses to a greater number of polymorphic sites which have not been previously assayed by the used *** this review,we present a broad overview of the genotype imputation process,showing the most known methods and presenting the main areas of interest,with a closer look to the most up-to-date approaches and a deeper understanding of its usage in the presentday genomic landscape,shedding a light on its future developments and investigation areas.

Triassic actinopterygian fishes:the recovery after the end-Permian crisis

In the last 15 years,the discovery of several new actinopterygian fish faunas from the Early and Middle Triassic of the Tethys,cast new light on the timing,speed and range of their recovery after the end-Permian *** addition to several new taxa having been described,the stratigraphical and geographical record of many others have been greatly *** fact,most of the new fossiliferous sites are in southern China,thus at the Eastern end of the Tethys,and furthermore a few are somewhat older(Chaohu,Panxian,Luoping)than the major classical Western Tethys sites(Monte San Giorgio).Following these new finds,it is possible to have a better definition of the Triassic recovery ***,after a quite short phase till the end of the Smithian(Olenekian,Early Triassic)in which a rather consistent fauna was present all around the Pangea coasts,a major radiation occurred in the Early-Middle Anisian after the new Middle Triassic fish fauna already appeared in the late Early Triassic,thus occuring well before what was previously supposed from the Alps ***,the new assemblages from southern China point to an early broader differentiation among the basal neopterygians rather than in the‘subholosteans’,the group that was then dominant in the Western Tethys since the Late Ani*** stands that during the Norian a new basal neopterygian radiation gave rise to several new branches that dominated the remaining part of the Mesozoic.

Biostratigraphy versus isotope geochronology: Testing the Urals island arc model

Formation of the Urals volcanic-hosted massive sulphide(VHMS) deposits is considered to be related with the intra-oceanic stage of island arc(s) development in the Upper Ordoviciane Middle Devonian based on the biostratigraphic record of ore-hosting sedimentary rocks. However, the direct Re-Os dating of four known VHMS systems in the Urals gives significantly younger Re-Os isochron ages ranging from355 ± 15 Ma up to 366 ± 2 Ma. To address this discrepancy, we performed SHRIMP U-Pb dating on zircons extracted from rhyodacites(Eifelian biostratigraphic age of 393 -388 Ma) from the footwall of the Alexandrinka VHMS deposit which has a Re-Os isochron age of sulphides of 355 ± 15 *** ^(206) Pb/^(238) U mean age of 374 ± 3 Ma(MSWD ? 1.4 and probability ? 0.11) is considered to be the crystallisation age of the host volcanic rock. This age is ca. 15 Ma younger than the Eifelian(393 -388 Ma)biostratigraphic age and overlaps the Frasniane Famennian boundary(372 ± 2 Ma), characterised by the final stages of Magnitogorsk Arc e East European continent collision. Such an inconsistency with geochronological age may be due to a reburial of conodonts during resedimentation as a result of erosion of older rocks in younger sedimentary sequences.

Important parameters in plasma jets for the production of RONS in liquids for plasma medicine: A brief review

Reactive oxygen and nitrogen species (RONS) are among the key factors in plasma medicine. They are generated by atmospheric plasmas in biological fluids, living tissues and in a variety of liquids. This ability of plasmas to create a delicate mix of RONS in liquids has been used to design remote or indirect treatments for oncological therapy by treating biological fluids by plasmas and putting them in contact with the tumour;Documented effects include selective cancer cell toxicity, even though the exact mechanisms involved are still under investigation. However, the "right" dose for suitable therapeutical activity is crucial and still under debate. The wide variety of plasma sources hampers comparisons. This review focuses on atmospheric pressure plasma jets as the most studied plasma devices in plasma medicine and compiles the conditions employed to generate RONS in relevant liquids and the concentration ranges obtained. The concentrations of H2O2, NO2^-, NO3^- and short-lived oxygen species are compared critically to provide a useful overview for the reader.
