In vitro calcification studies on bioprosthetic and decellularized heart valves under quasi-physiological flow conditions

The lifespan of biological heart valve prostheses available in the market is limited due to structural alterations caused by calcium phosphate deposits formed from blood plasma in contact with the tissues.The objective of this work is to present a comparative methodology for the investigation of the formation of calcium phosphate deposits on bioprosthetic and tissue-engineered scaffolds in vitro and the influence of mechanical forces on tissue mineralization.Based on earlier investigations on biological mineralization at constant supersaturation,a circulatory loop simulating dynamic blood flow and physiological pressure conditions was developed.The system was appropriately adapted to evaluate the calcification potential of decellularized(dcV)and glutaraldehyde-fixed(GAV)porcine aortic valves.Results indicated that dcV calcified at higher,statistically nonsignificant,rates in comparison with GAV.This difference was attributed to the tissue surface modifications and cell debris leftovers from the decellularization process.Morphological analysis of the solids deposited after 20 h by scanning electron microscopy in combination with chemical microanalysis electron-dispersive spectroscopy identified the solid formed as octacalcium phosphate(ca8(PO4)6H2·5H2O,OcP).OcP crystallites were preferentially deposited in high mechanical stress areas of the test tissues.Moreover,GAV tissues developed a significant transvalvular pressure gradient increase past 36 h with a calcium deposition distribution similar to the one found in explanted prostheses.In conclusion,the presented in vitro circulatory model serves as a valuable prescreening methodology for the investigation of the calcification process of bioprosthetic and tissue-engineered valves under physiological mechanical load.

Quality assessment of surgery for colorectal cancer:Where do we stand?

Quality assurance in surgery has been one of the most important topics of debate among colorectal surgeons in the past dec.de.It has produc.d new surgical standards that led in part to the impressive oncological outcomes we see in many units today.Total mesorectal excision,complete mesocolic excision(cME),and the Japanese d3 lymphadenectomy are now benchmark techniques embrac.d by many surgeons and widely recommended by surgical societies.However,there are still ongoing discrepancies in outcomes largely based on surgeon performance.This is one of the main reasons why many countries have shifted colorectal cancer surgery only to high volume centers.defining markers of surgical quality is thus a perquisite to ensure that standards and oncological outcomes are met at an institutional level.With the evolution of cME surgery,various quality markers have been described,mostly based on measurements on the surgical specimen and lymph node yield,while others have proposed radiological markers(i.***.arterial stumps)measured on postoperative scans as part of the routine cancer follow-up.There is no ideal marker;however,taken together and assembled into a new score or set of criteria may become a future point of reference for reporting outcomes of colorectal cancer surgery in research studies and defining subspecialization requirements both at an individual and hospital level.

Type 2 diabetes mellitus(T2dM)is recognized as a serious public health concern with a considerable impact on human life,long-term health expenditures,and substantial health losses.In this context,the use of dietary polyphenols to prevent and manage T2dM is widely documented.These dietary compounds exert their beneficial effects through several actions,including the protection of pancreatic islet β-cell,the antioxidant capacities of these molecules,their effects on insulin secretion and actions,the regulation of intestinal microbiota,and their contribution to ameliorate diabetic complications,particularly those of vascular origin.In the present review,we intend to highlight these multifaceted actions and the molecular mechanisms by which these plant-derived secondary metabolites exert their beneficial effects on type 2 diabetes patients.

Anti-ganglioside antibodies are associated with delayed/poor clinical recovery in Guillain-Barrèsyndrome,mostly related to halted axon regeneration.cross-linking of cell surface gangliosides by anti-ganglioside antibodies triggers inhibition of nerve repair in in vitro and in vivo paradigms of axon regeneration.These effects involve the activation of the small GTPase Rho A/ROcK signaling pathways,which negatively modulate growth cone cytoskeleton,similarly to well stablished inhibitors of axon regeneration described so far.The aim of this work was to perform a proof of concept study to demonstrate the effectiveness of Y-27632,a selective pharmacological inhibitor of ROcK,in a mouse model of axon regeneration of peripheral nerves,where the passive immunization with a monoclonal antibody targeting gangliosides Gd1a and GT1b was previously reported to exert a potent inhibitory effect on regeneration of both myelinated and unmyelinated fibers.Our results demonstrate a differential sensitivity of myelinated and unmyelinated axons to the pro-regenerative effect of Y-27632.Treatment with a total dosage of 9 mg/kg of Y-27632 resulted in a complete prevention of anti-Gd1a/GT1b monoclonal antibody-mediated inhibition of axon regeneration of unmyelinated fibers to skin and the functional recovery of mechanical cutaneous sensitivity.In contrast,the same dose showed toxic effects on the regeneration of myelinated fibers.Interestingly,scale down of the dosage of Y-27632 to 5 mg/kg resulted in a significant although not complete recovery of regenerated myelinated axons exposed to anti-Gd1a/GT1b monoclonal antibody in the absence of toxicity in animals exposed to only Y-27632.Overall,these findings confirm the in vivo participation of Rho A/ROcK signaling pathways in the molecular mechanisms associated with the inhibition of axon regeneration induc.d by anti-Gd1a/GT1b monoclonal antibody.Our findings open the possibility of therapeutic pharmacological intervention targeting Rho A/Rock pathway in immune neuropathies associated wi

Hepatic artery thrombosis(HAT)is a devastating vascular complication following liver transplantation,requiring prompt diagnosis and rapid revascularization treatment to prevent graft loss.At present,imaging modalities such as ultrasound,computed tomography,and magnetic resonance play crucial roles in diagnosing HAT.Although imaging techniques have improved sensitivity and specificity for HAT diagnosis,they have limitations that hinder the timely diagnosis of this complication.In this sense,the emergence of artificial intelligence(AI)presents a transformative opportunity to address these diagnostic limitations.The development of machine learning algorithms and deep neural networks has demonstrated the potential to enhance the precision diagnosis of liver transplant complications,enabling quicker and more accurate detection of HAT.This article examines the current landscape of imaging diagnostic techniques for HAT and explores the emerging role of AI in addressing future challenges in the diagnosis of HAT after liver transplant.

Machine learning assisted derivation of minimal low-energy models for metallic magnets

We consider the problem of extracting a low-energy spin Hamiltonian from a triangular Kondo Lattice Model(KLM).The non-analytic.dependence of the effective spin-spin interactions on the Kondo exchange excludes the use of perturbation theory beyond the second order.We then introduce a Machine Learning(ML)assisted protocol to extract effective two-and four-spin interactions.The resulting spin model reproduces the phase diagram of the original KLM as a function of magnetic field and single-ion anisotropy and reveals the effective four-spin interactions that stabilize the field-induc.d skyrmion crystal phase.Moreover,this model enables the computation of static and dynamical properties with a much lower numerical cost relative to the original KLM.A comparison of the dynamical spin structure factor in the fully polarized phase computed with both models reveals a good agreement for the magnon dispersion even though this information was not included in the training data set.

dispersed multiphase flows are commonly encountered in a wide variety of natural and industrial applications,from sedimentation in rivers and plankton surfacing in oceanic flows to fluidized bed in chemical reactors and pneumatic conveying systems.In these flows,a dispersed phase(con-sisting of particles,droplets and/or bubbles)is transported by a carrier phase.The interaction between the phases re-sults in complex dynamics(e.g.,transport,spatial clustering,and deformation or break-up)that is rich in physics and poses a wide spectrum of theoretical,computational,and experimental challenges.These challenges are at the very core of the research efforts made by the multiphase flow community over the past dec.des.

A high Q terahertz metamaterial absorber using concentric elliptical ring resonators for harmful gas sensing applications

A nearly perfect metamaterial absorber is proposed that can find utility in terahertz sensing applications.The design consists of two concentric elliptical ring resonators(ERRs)whose parameters are appropriately set to achieve dual band absorption with near perfect absorption.The first absorption band at 3.62 THz having a Q-factor of 51.7 was caused due to the currents in the outer and inner ERR.The second absorption peak at 3.814 THz having a Q factor of 1411.11 was a consequence of currents flowing across the gap between the two concentric resonators.Furthermore,it is observed that the absorption bands are sensitive to the variation in refractive index of the surrounding medium.The sensitivity's in the absorption bands are 3 THz/RIU and 3.59 THz/RIU respectively.A sensor is proposed based on this design to detect harmful gases,which is demonstrated for detection of Methane and chloroform.High Q-factor and high sensitivity of the narrow band makes the design an excellent sensor for detecting variations in the refractive index.
