Plant triterpenoid saponins function as susceptibility factors to promote the pathogenicity of Botrytis cinerea

The current spotlight of cancer therapeutics is shifting towards personalized medicine with the widespread use of monoclonal antibodies(mAbs).Despite their increasing potential,mAbs have an intrinsic limitation related to their inability to cross cell membranes and reach intracellular *** offers promising solutions to overcome this limitation,however,formulation challenges *** challenges are the limited loading capacity(often insufficient to achieve clinical dosing),the complex formulation methods,and the insufficient characterization of mAb-loaded ***,we present a new nanocarrier consisting of hyaluronic acid-based nanoassemblies(HANAs)specifically designed to entrap mAbs with a high efficiency and an outstanding loading capacity(50%,w/w).HANAs composed by an mAb,modified HA and phosphatidylcholine(PC)resulted in sizes of~100 nm and neutral surface *** modeling identified the principal factors governing the high affinity of mAbs with the amphiphilic HA and *** composition and structural configuration were analyzed using the orthogonal techniques cryogenic transmission electron microscopy(cryo-TEM),asymmetrical flow field-flow fractionation(AF4),and small-angle X-ray scattering(SAXS).These techniques provided evidence of the formation of core-shell nanostructures comprising an aqueous core surrounded by a bilayer consisting of phospholipids and amphiphilic *** vitro experiments in cancer cell lines and macrophages confirmed HANAs’low toxicity and ability to transport mAbs to the intracellular *** reproducibility of this assembling process at industrial-scale batch sizes and the long-term stability was *** conclusion,these results underscore the suitability of HANAs technology to load and deliver biologicals,which holds promise for future clinical translation.

In vivo oxidative stress alters thiol redox status of peroxiredoxin 1 and 6 and impairs rat sperm quality

Oxidative stress, the imbalance between the production of reactive oxygen species （ROS） and antioxidant activity is a major culprit of male infertility. Peroxiredoxins （PRDXs） are major antioxidant enzymes of mammalian spermatozoa and are thiol oxidized and inactivated by ROS in a dose-dependent manner. Their deficiency and/or inactivation have been associated with men infertility. The aim of this study was to elucidate the impact of oxidative stress, generated by the in vivo tert-butyl hydroperoxide （tert-BHP） treatment on rat epididymal spermatozoa during their maturation process. Adult Sprague-Dawley males were treated with 300 -moles tert-BHP/kg or saline （control） per day intraperitoneal for 15 days. Lipid peroxidation （2-thibarbituric acid reactive substances assay）, total amount and thiol oxidation of PRDXs along with the total amount of superoxide dismutase （SOD）, motility and DNA oxidation （8-hydroxy-deoxyguanosine） were determined in epididymal spermatozoa. Total amount of PRDXs and catalase and thiol oxidation of PRDXs were determined in caput and cauda epididymis. While animals were not affected by treatment, their epididymal spermatozoa have decreased motility, increased levels of DNA oxidation and lipid peroxidation along with increased PRDXs （and not SOD） amounts. Moreover, sperm PRDXs were highly thiol oxidized. There was a differential regulation in the expression of PRDX1 and PRDX6 in the epididymis that suggests a segment-specific role for PRDXs. In conclusion, PRDXs are increased in epididymal spermatozoa in an attempt to fight against the oxidative stress generated by tert-BHP in the epididymis. These findings highlight the role of PRDXs in the protection of sperm function and DNA integrity during epididymal maturation.

This article presents an update on the variable prognostic significance of different sperm pathologies in patients with severe male factor infertility due to morphology and motility disorders. Severe asthenozoospermia is one of the leading causes of male infertility as spermatozoa cannot reach the oocyte and/or penetrate normally. Identifying structural causes of sperm immotility was of great concern before the advent of intracytoplasmic sperm injection （ICSI）, because immotility was the limiting factor in the treatment of these patients. In these cases, in vitro methods are used to identify live spermatozoa or stimulate sperm motility to avoid selection of non-viable cells. With these advances, fertilization and pregnancy results have improved dramatically. The identification of genetic phenotypes in asthenozoospermia is important to adequately inform patients of treatment outcomes and risks. The one sperm characteristic that seriously affects fertility prognosis is teratozoospermia, primarily sperm head and neck anomalies. Defects of chromatin condensation and acrosomal hypoplasia are the two most common abnormalities in severe teratozoospermia. The introduction of microscopic methods to select spermatozoa and the development of new ones to evaluate sperm quality before ICSI will assure that ultrastructural identification ofsperm pathologies will not only be of academic interest, but will also be an essential tool to inform treatment choice. Herein, we review the differential roles played by sperm components in normal fertilization and early embryo development and explore how assisted reproductive technologies have modified our concepts on the prognostic significance of sperm pathologies affecting the head, neck, mid-piece and tail.

The increased demand of electronic devices promotes the development of advanced and more efficient energy storage devices, such as batteries. Lithium-ion batteries (LIBs) are the most studied battery systems due to their high performance. Among the different battery components, the separator allows the control of lithium ion diffusion between the electrodes. To overcome some drawbacks of liquid electrolytes, including safety and environmental issues, solid polymer electrolytes (SPEs) are being developed. In this work, a UV photocurable polyurethane acrylate (PUA) resin has been blended with lithium bis(trifluoromethanesulfonyl) imide (LiTFSI) up to 30 wt% LiTFSI content to reach a maximum ionic conductivity of 0.0032 mS/cm at room temperature and 0.09 mS/cm at 100 ℃. Those values allowed applying the developed materials as photocurable SPE in Swagelok type Li/C-LiFePO_(4) half-cells, reaching a battery discharge capacity value of 139 mAh.g^(−1) at C/30 rate. Those results, together with the theoretical studies of the discharge capacity at different C-rates and temperatures for batteries with LiTFSI/PUA SPE demonstrate the suitability of the developed photocurable SPE for LIB applications.

Convergence of the turkey gut microbiota following cohabitation under commercial settings

Organic coating of 1-2-nm-size silicon nanoparticles： Effect on particle properties

Photoluminescent silicon nanoparticles 1-2 nm in size were synthesized by a wet chemical procedure and derivatized with propylamine （NH2SiNP）. Surface NH2 groups were used as linkers for additional poly（ethylene glycol）（PEG） and folic acid （Fo） attachment （PEG-NHSiNP and Fo-NHSiNP, respectively） to enable efficient targeting of the particles to tumors and inflammatory sites. The particles were characterized by transmission electron microscopy, Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, ζ potential, dynamic light scattering, and time-resolved anisotropy. The photophysical properties and photosensitizing capacity of the particles and their interaction with proteins was dependent on the nature of the attached molecules. While PEG attachment did not alter the photophysical behavior of NH2SiNP, the attachment of Fo diminished particle photoluminescence. Particles retained the capacity for 1O2 generation; however, efficient 1O2 quenching by the attached surface groups may be a drawback when using these particles as 1O2photosensitizers. In addition, Fo attachment provided particles with the capacity to generate the superoxide anion radical （O2-）. The particles were able to bind tryptophan residues of bovine serum albumin （BSA） within quenching distances. NH2SiNP and PEG-NHSLNP ground state complexes with BSA showed binding constants of （3.1 ± 0.3） × 10^4 and （1.3 ±0.4） × 10^3 M-1, respectively. The lower value observed for PEG-NHSiNP complexes indicates that surface PEGylation leads to a reduction in protein adsorption, which is required to prevent opsonization. An increase in particle luminescence upon BSA binding was attributed to the hydrophobic environment generated by the protein. NH2SiNP-BSA complexes were also capable of resonance energy transfer.

Advanced radiative cooling materials with both heating and cooling mode is of pivotal importance for all-season energy-saving in *** this work,we report the design and fabrication of bacterial cellulose-based janus films(j-BC)with radiative cooling and solar heating properties,which were developed by two-step vacuum-assisted filtration of modified MXene-doped bacterial cellulose and modified silicon nitride(Si_(3)N_(4))-doped bacterial cellulose,followed by hot-pressing and drying *** as-prepared j-BC films show a unique janus structure where modified MXene nanosheets and cellulose nanofibers are on the bottom surface,and modified silicon nitride(Si_(3)N_(4))nanoparticles and cellulose nanofibers are on the top *** radiative cooling effect of j-BC films is enabled by the Si_(3)N_(4)-doped bacterial cellulose due to the high mid-infrared emissivity of Si_(3)N_(4) nanoparticles,which shows a high solar reflection of~98.1%and high emissivity of~93.6%in the atmospheric transparency window(8-13μm).Thanks to the enhanced photothermal conversion of the modified MXene nanosheets,a reduced solar reflection(6.6%)and relatively low thermal emissivity in the atmospheric window(71.4%)are achieved,making sure the solar heating effect of j-BC *** the outdoor tests,j-BC films achieve a sub-ambient temperature drop of~3.8°C and an above-ambient temperature rise of~14.2°*** prediction demonstrated that the j-BC films with dual modes have great potential of all-season energy saving for buildings and a corresponding energy-saving map in China is also *** work disclosed herein can provide an avenue for the shaping of advanced radiative cooling materials for emerging applications of personal thermal management,sustainable energy-efficient buildings,and beyond.
