Coherent Raman scattering(CRs)microscopy is widely recognized as a powerful tool for tackling biomedical problems based on its chemically specific label-free contrast,high spatial and spectral resolution,and high ***,the clinical translation of CRs imaging technologies has long been hindered by traditional solid-state lasers with environmentally sensitive operations and large *** fibre lasers can potentially overcome these shortcomings but have not yet been fully exploited for CRs imaging,as previous implementations have suffered from high intensity noise,a narrow tuning range and low power,resulting in low image qualities and slow imaging ***,we present a novel high-power self-synchronized two-colour pulsed fibre laser that achieves excellent performance in terms of intensity stability(improved by 50 dB),timing jitter(24.3 fs),average power fluctuation(20 dB)and pulse width variation(wavenumber range(2700-3550 cm^(−1)).The versatility of the laser source enables,for the first time,high-contrast,fast CRs imaging without complicated noise reduction via balanced detection *** capabilities are demonstrated in this work by imaging a wide range of species such as living human cells and mouse arterial tissues and performing multimodal nonlinear imaging of mouse tail,kidney and brain tissue sections by utilizing second-harmonic generation and twophoton excited fluorescence,which provides multiple optical contrast mechanisms simultaneously and maximizes the gathered information content for biological visualization and medical *** work also establishes a general scenario for remodelling existing lasers into synchronized two-colour lasers and thus promotes a wider popularization and application of CRs imaging technologies.

In two-photon microscopy,low illumination powers on samples and a high signal-to-noise ratio(sNR)of the excitation laser are highly desired for alleviating the problems of photobleaching and phototoxicity,as well as providing clean backgrounds for ***,the high-repetition-rate Ti:sapphire laser and the low-sNR Raman-shift lasers fall short of meeting these demands,especially when used for deep ***,we demonstrate a 937-nm laser frequency-doubled from an all-fiber mode-locked laser at 1.8μm with a low repetition rate of∼9 MHz and a high sNR of 74 *** showcase two-photon excitations with low illumination powers on multiple types of biological tissues,including fluorescence imaging of mouse brain neurons labeled with green and yellow fluorescence proteins(GFP and YFP),DiI-stained and GFP-labeled blood vessels,Alexa Fluor 488/568-stained mouse kidney,and second-harmonic-generation imaging of the mouse skull,leg,and *** achieve a penetration depth in mouse brain tissues up to 620μm with an illumination power as low as∼10 mw,and,even for the DiI dye with an extremely low excitation efficiency of 3.3%,the penetration depth is still up to 530μm,indicating that the low-repetition-rate source works efficiently for a wide range of dyes with a fixed excitation *** low-repetition-rate and high-sNR excitation source holds great potential for biological investigations,such as in vivo deep-tissue imaging.

A magnesium alloy containing essential,non-toxic,biodegradable elements such as Ca and Zn has been fabricated using a novel twin-roll casting process(TRC).Microstructure,mechanical properties,in vivo corrosion and biocompatibility have been assessed and compared to the properties of the rare earth(RE)element containing wE43 *** Mg-0.5 wt% Zn-0.5 wt% Ca exhibited fine grains with an average grain size ranging from 70 to 150μ*** properties of a TRC Mg-0.5Zn-0.5Ca alloy showed an ultimate tensile strength of 220 MPa and ductility of 9.3%.The TRC Mg-0.5Zn-0.5Ca alloy showed a degradation rate of 0.51±0.07 mm/y similar to that of the wE43 alloy(0.47±0.09 mm/y)in the rat model after 1 week of *** week 4 the biodegradation rates of both alloys studied were lowered and stabilized with fewer gas pockets around the *** histological analysis shows that both wE43 and TRC Mg-0.5Zn-0.5Ca alloy triggered comparable tissue healing responses at respective times of *** presence of more organized scarring tissue around the TRC Mg-0.5Zn-0.5Ca alloys suggests that the biodegradation of the RE-free alloy may be more conducive to the tissue proliferation and remodelling process.

In contrast to the cries of human infants, sounds made by non-human infants in different stressful behavioral contexts （hunger or physical discomfort, isolation, capture by humans or predators） are usually treated as distinct types of vocalizations. However, if distress vocalizations produced by different species and in different contexts share a common motivational state and associated neurochemical pathways, we can expect them to share a common acoustic structure and adaptive function, showing only limited variation that corresponds to the infant＇s level of arousal. Based on this premise, we review the acoustic structure and adaptive function of two types of distress calls, those given when infants were isolated from their mothers （isolation caUs） or captured by humans （capture calls）. we conducted a within-context comparison examining the two call types across a diverse se- lection of mammalian species and other vertebrate groups, followed by a comparison of how acoustic structure and function dif- fers between these contexts. In addition, we assessed acoustic traits that are critical to the response of caregivers. Across verte- brate species, distress vocalizations produced in these two behavioral contexts tend to be tonal with a simple chevron, flat or de- scending pattern of frequency modulation. Reports that both isolation and capture calls of vertebrate infants serve to attract care- givers are universal, and the fundamental frequency of infant vocalizations is often critical to this response. The results of our re- view are consistent with the hypothesis that differences in the acoustic structure of isolation and capture distress vocalizations re- flect differences in arousal, and not discrete functions. The similarity in acoustic structure and caregiver response observed across vertebrates adds support to the hypothesis that the production and processing of distress vocalizations are part of a highly-conserved system of social vocal behaviour in vertebrate

BACKGROUND Alpha-defensin has been widely studied for the diagnosis of periprosthetic joint infection(PJI).However,there is a lack of detailed information regarding the proper laboratory technique of the enzyme-linked immunosorbent assay(ELIsA)method,such as sample *** To assess the influence of dilution in the synovial fluid during ELIsA for the diagnosis of knee PJI;and determine which dilution presents a better *** Forty samples of synovial fluid from arthroplasty knees were included,17 in the infected group and 23 in the aseptic group,according to Musculoskeletal Infection society ***,five synovial fluid samples from each group were assessed for quantitative analysis of alpha-defensin using *** dilution ratios(1:10,1:100,1:500,1:1000 and 1:5000)were tested based on the predetermined cutoff value of 5.2 mg/*** dilutions that performed better were used to compare the results of all *** For infected cases,a gradual increase in the dilution of synovial fluid samples led to an equivalent increase in alpha-defensin *** same was not observed in the aseptic *** 1:1000 and 1:5000 dilutions presented satisfactory results to differentiate infected and aseptic *** analyses were performed using 1:1000 and 1:5000 for all 40 *** 1:1000 dilution resulted in a sensitivity of 88.2%(95%CI,66%-98%)and specificity of 95.7%(95%CI,79%-99%),whereas the 1:5000 dilution presented a sensitivity of 94.1%(95%CI,73%-99%)and a specificity of 100%(95%CI,86%-100%).CONCLUsION The synovial fluid dilution had an important influence on the alpha-defensin ELIsA *** of 1:5000 showed the best performance for the diagnosis of knee *** results of this study set the basis for a more reliable and reproducible alpha-defensin ELIsA during the investigation of PJI,contributing to the expansion of this technique in different treatment centers worldwide.
