This study aimed to investigate the effects of arsenic trioxide(As2.3) on the mitochondrial DNA(mtDNA) of acute promyelocytic leukemia(APL) *** NB4 cell line was treated with 2.0 μmol/L As2.3 in vitro,and the primary APL cells were treated with 2.0 μmol/L As2.3 in vitro and 0.16 mg kg-1 d-1 As2.3 in *** mitochondrial DNA of all the cells above was amplified by PCR,directly sequenced and analyzed by Sequence Navigatore and Factura *** apoptosis rates were assayed by flow *** DNA mutation in the D-loop region was found in NB4 and APL cells before As2.3 use,but the mutation spots were remarkably increased after As2.3 treatment,which was positively correlated to the rates of cellular apoptosis,the correlation coefficient:rNB4-As2.3=0.973818,and rAPL-As2.3=*** mutation types include transition,transversion,codon insertion or deletion,and the mutation spots in all samples were not constant and *** is revealed that As2.3 aggravates mtDNA mutation in the D-loop region of acute promyelocytic leukemia cells both in vitro and in *** DNA might be one of the targets of As2.3 in APL treatment.

Phytolacca is the biggest and most original genus in Phytolaccaceae and an important genus in plant systematic *** microscopy results show that the Phytolacca americana *** arises from the caulis(floral receptacle).The perisperm and hypostase are simultaneously initiated from the top several layers of cells of chalaza after fertilization,and the perisperm is located between the nucellus and *** the early stages of development,the hypostase cells are thin-walled with dense cytoplasm,clear nuclei,and some reserve ***,at the heart-shaped embryo stage,the hypostase cells are dead and *** main functions of the hypostase may be to maintain cellular division and perisperm growth without delivering nutrient materials to the *** evolutionary picture of placentation in Caryophyllales is also presented.

The stimulator of interferon genes(STING),an integral adaptor protein in the DNA-sensing pathway,plays a pivotal role in the innate immune response against ***,it presents a valuable therapeutic target for infectious diseases and *** observed that fangchinoline(Fan),a bis-benzylisoquinoline alkaloid(BBA),effectively impedes the replication of vesicular stomatitis virus(VSV),encephalomyocarditis virus(EMCV),influenza A virus(H1N1),and herpes simplex virus-1(HSV-1)in *** treatment significantly reduced the viral load,attenuated tissue inflammation,and improved survival in a viral sepsis mouse ***,Fan activates the antiviral response in a STING-dependent manner,leading to increased expression of interferon(IFN)and interferon-stimulated genes(ISGs)for potent antiviral effects in vivo and in ***,Fan interacts with STING,preventing its degradation and thereby extending the activation of IFN-based antiviral ***,our findings highlight the potential of Fan,which elicits antiviral immunity by suppressing STING degradation,as a promising candidate for antiviral therapy.

The events of cell death and the expression of nuclear matrix protein (NMP) have been investigated in a promyelocytic leukemic cell line HL-60 induced with etoposide. By means of TUNEL assay, the nuclei displayed a characteristic morphology change, and the amount of apoptotic cells increased early and reached maximun about 39% after treatment with etoposide for 2.h. Nucleosomal DNA fragmentation was observed after treatment for 4 h. The morphological change of HL-60 cells, thus, occurred earlier than the appearance of DNA ladder. Total nuclear matrix proteins were analyzed by 2.dimensional gel electrophoresis. Differential expression of 59 nuclear matrix proteins was found in 4 h etoposide treated cells. Western blotting was then performed on three nuclear matrix acssociated proteins, PML, HSC70 and NuMA. The expression of the suppressor PML protein and heat shock protein HSC70 were significantly upregulated after etoposide treatment, while NuMA, a nuclear mitotic apparatus protein, was down regulated. These results demonstrate that significant biochemical alterations in nuclear matrix proteins take place during the apoptotic process.

A full-length cDNA of GlFtsZ was isolated by screening the cDNA library of Gentiana lutea. Analysis of the deduced amino acid sequence encoded by GIFtsZ indicated that GIFtsZ protein possesses the typical conservative motifs existed in all FtsZ proteins. The existence of putative plastid transit peptide in its N-terminus suggested that GIFtsZ might function inside of plastids. With the developmental process of petals of Gentiana lutea, the expression of plastid division gene GIFtsZ declined gradually, whereas the expression of carotenoids biosynthesis gene Zds increased obviously; meanwhile, in contrast to the increment of carotenoids, the content of chlorophyll in petals decreased sharply. The chloroplasts turned into chromoplasts, and the color of petals also turned from green to golden. All of these results suggested that the expression of GIFtsZ is accompanied with the development and differentiation of plastids.

Two cDNAs of plastid division gene NtFtsZ1-1 and NtFtsZl-2.are isolated from Nicotiana tabacum by RT-PCR and rapid amplification cDNA ends (RACE) method. Analysis of the deduced amino acid sequences encoded by NtFtsZl-1 and NtFtsZl -2.indicate that these two proteins possess the typical conservative motifs and GTP binding sites existing in all FtsZ proteins. The existence of putative plastid transit peptide in their N-terminal suggests that there are at least two transit-peptide containing FtsZ proteins in higher plants. Phylogenetic analysis based on amino acid sequences of FtsZ proteins also supports this interference. These two NtFtsZ genes demonstrate a similar expression pattern during the plant development detected by Northern blot. Expression of NtFtsZl-1 and NtFtsZl-2.in E. coli interrupts the normal division process of host cells. These results suggest the diverse functions of FtsZ proteins in higher plants.

Suppression of roots and/or their symbiotic microorganisms,such as mycorrhizal fungi and rhizobia,is an effective way for alien plants to outcompete native ***,little is known about how invasive and native plants interact with the quantity and activity of nutrient-acquisition *** a pot experiment was conducted with monoculture and mixed plantings of an invasive plant,Xanthium strumarium,and a common native legume,Glycine *** measured traits related to root and nodule quantity and activity and mycorrhizal *** to the monoculture,fine root quantity(biomass,surface area)and activity(root nitrogen(N)concentration,acid phosphatase activity)of *** decreased in mixed plantings;nodule quantity(biomass)decreased by 45%,while nodule activity in Nfixing via rhizobium increased by 106%;mycorrhizal colonization was *** of N fixation to leaf N content in *** increased in the mixed plantings,and this increase was attributed to a decrease in the rhizosphere soil N of *** in the mixed *** root quantity and activity,along with a higher mycorrhizal association was observed in *** in the mixed compared to ***,the invasive plant did not directly scavenge N from nodule-fixed N,but rather depleted the rhizosphere soil N of the legume,thereby stimulating the activity of N-fixation and increasing the dependence of the native legume on this N *** quantity-activity framework holds promise for future studies on how native legumes respond to alien plant invasions.
