Pharmacogenetics uses genetic tests to predict the effectiveness of medication treatments for individual patients with drug dependence such as alcohol and opiates. An important shared neurobiological component of the reinforcement and dependence on alcohol and opiates is alcohol’s enhancement of β- endorphin release. The physiological and subjective effects of both alcohol and opiates are associated with β-endorphin and the mu opiate system. A pharmacotherapy that has been effective for both disorders is naltrexone, which increases baseline β-endorphin release blocking further release by alcohol. Naltrexone’s action as an alcohol pharmacotherapy is facilitated by a putative functional single nucleotide polymorphism (SNP) in the opioid mu receptor gene (Al18g) which alters its receptor function. Patients with this SNP have significantly lower relapse rates to alcoholism when treated with naltrexone. This SNP is particularly common among northern Chinese suggesting an important role for naltrexone treatment of alcoholism in China. Another important genetic polymorphism related to stimulant and perhaps other types of dependence frequently occurs (about 40% of the population) in the promoter region of the gene coding for dopamine beta hydroxylase (DβH). This polymorphism leads to 10 to 100 times lower levels of the DβH enzyme, which converts dopamine to norepinephrine in neurons. Stimulant abusers with normal levels of this enzyme show significant reductions in stimulant abuse when treated with DβH inhibitors, while those with the polymorphism for low DβH levels show no treatment efficacy with these medications. Pharmacogenetics has great potential for improving treatment outcome as we identify gene variants that affect pharmacodynamic as well as the more commonly studied pharmacokinetic factors. These mutations can guide pharmacotherapeutic agent choice for optimum treatment of alcohol and stimulant abuse and reduce relapse after successful initial abstinence.

Region of Treatment in Radiotherapy and Second Malignancies in Breast Cancer Patients

A cohort study was conducted based on clinical records for 5248 women treated for breast cancer in Florence (Italy), with continuous follow up from 1965 to 1994. The subjects were categorised into groups such as no radiation treatment;breast dose only;and radiation doses in one, two, three or four of the following fields: namely internal mammary chain, supraclavicular nodes, axillary lymph nodes and chest wall. The Cox proportional hazards model for ungrouped survival data was used to estimate the relative risk for second cancer after radiotherapy delivered to different regions. The relative risk for all second cancers combined was statistically significantly raised if the internal mammary chain and supraclavicular nodes were irradiated. However, we found that the relative risk of the second malignancies could be reduced if all the locoregional lymph nodes (the internal mammary chain, supraclacicular nodes, axillary nodes) and chest wall are irradiated at the same time. If this finding can be verified by other large cohort studies or randomized clinical trials, it may have implications in clinical practice when deciding upon the targeted areas for radiotherapy;partial radiation treatment of the locoregional lymph nodes could raise the risk of second malignancies and should ideally be avoided.

It is likely that the majority of proteins will undergo post-translational modification, be it enzymatic or non-enzymatic. These modified protein(s) regulate activity, localization and interaction with other cellular molecules thereby maintaining cellular hemostasis. Alcohol exposure significantly alters several of these post-translational modifications leading to impairments of many essential physiological processes. Here, we present new insights into novel modifications following ethanol exposure and their role in the initiation and progression of liver injury. This critical review condenses the proceedings of a symposium at the European Society for the Biomedical Research on Alcoholism Meeting held September 12-15, 2015, in Valencia, Spain.

背景与目的肥胖是Ⅱ型糖尿病、心血管病、高血压、中风和多种癌症的高危因素,随着肥胖的全球性流行,对其发生机制的深入研究并最终为肥胖及其相关疾病的防治提供新靶标成为这一领域的重点。本文利用 CYP1B1基因敲除小鼠,研究高脂肪膳食下 CYP1B1在肝脏脂肪代谢中的作用并探讨其可能机制。方法 CYP1B1基因敲除小鼠(KO)由美国 NIH 赠送,C57BL/6J 野生型小鼠(WT)购自杰克逊实验室,各20只。WT 和 KO 雄性小鼠随机分为高(HFD)、低(LFD)脂肪膳食组,即 KO-LFD、 WT-LFD、KO-HFD 和 WT-HFD,每组10只。在小鼠断奶后开始喂养,共11周,每三天换食物及称量体重。实验结束后以 CO2麻醉断头处死,取附睾脂肪组织测定各项指标。结果整体指标显示,KO-HDF 小鼠的体脂分布,体重增量(22.1±1.16 g)和附睾脂肪组织重量(1.04±0.20 g)均显着低于 WT-HFD(分别为29.4±0.87 g,2.37±0.10g)。组化结果显示基因敲除小鼠其脂肪细胞体积明显小于野生型小鼠。血清学指标表明,CYP1B1基因缺失小鼠其肝脏脂肪含量明显降低(KO-HDF 11.8±5.2 vs WT-HFD 25.0±5.6μg/g);低、高脂膳食组 CYP1B1基因敲除小鼠血糖浓度(101±14,106±15 mg/dL)均显着低于相对应的野生型小鼠(159±36,210±27 mg/dL),并抑制高脂膳食对血清胰岛素水平诱导(KO-HDF 1.6±0.6 vs WT-HFD 6.3±5.1 μg/l)。鉴于肝脏在脂肪代谢中的关键作用,我们利用实时定量 RT-PCR 测量 CYP1B1敲除对肝脏基因表达的影响。通过对肝脏中控制脂肪形成(PPARg,CD36,PPARa,SREBP1c 等)、转运(gLUT4,FABP1 等)和脂肪酸代谢(SCD-1,FAS,CPT1a,UCP2,gPAT 等)关键基因表达进行分析,同时利用免疫印迹对 AMPK 的活化状态进行检测。结果表明,CYP1B1敲除可显着抑制 PPARg 与 CD36的表达,并进一步抑制高脂膳食对其的诱导:肝脏中控制脂肪合成的基因如 FAS,SCD-1,Crat 等表达下降;而肝脏中控制脂肪酸氧化的基因如 CPT1a,UCP-2表达显着上升。AMP 蛋白激酶(AMPK)是机体能量状态的探测器,主要通过影响其下游基因的表达发挥作用,如上调 gLUT4和线粒体基因,下调肝脏脂肪酸合成和糖异生的基因表达等。对肝脏 AMPK 免疫印迹结果显示,CYP1B1的敲除可激活 AMPK 的磷酸化,表明 AMPK 可能是 CYP1B1基因敲除抑制机体脂肪堆积的调节器。结论 CYP1B1基因敲除对高脂膳食诱导的肥胖有抵抗作用,并改善肥胖相关的胰岛素抵抗: 而这种保护作用可能是通过 AMPK 的活化,调节机体对脂肪形成及脂肪酸氧化代谢相关基因表达来实现的。

High-density linkage map construction and identification of loci regulating fruit quality traits in blueberry

Fruit quality traits play a significant role in consumer preferences and consumption in blueberry(Vaccinium corymbosum L).The objectives of this study were to construct a high-density linkage map and to identify the underlying genetic basis of fruit quality traits in blueberry.A total of 287 F_(1) individuals derived from a cross between two southern highbush blueberry cultivars,‘Reveille’and‘Arlen’,were phenotyped over three years(2016–2018)for fruit quality-related traits,including titratable acidity,pH,total soluble solids,and fruit weight.A high-density linkage map was constructed using 17k single nucleotide polymorphisms *** linkage map spanned a total of 1397 cM with an average inter-loci distance of 0.08 *** quantitative trait loci interval mapping based on the hidden Markov model identified 18 loci for fruit quality traits,including seven loci for fruit weight,three loci for titratable acidity,five loci for pH,and three loci for total soluble *** of these loci were detected in more than one *** loci explained phenotypic variance ranging from 7 to 28% for titratable acidity and total soluble solid,and 8-13% for ***,the loci identified for fruit weight did not explain more than 10%of the phenotypic *** also reported the association between fruit quality traits and metabolites detected by Proton nuclear magnetic resonance analysis directly responsible for these fruit quality *** acids,citric acid,and quinic acid were significantly(Pg and positive correlation with total soluble ***,the study dissected the genetic basis of fruit quality traits and established an association between these fruit quality traits and metabolites.

裂隙走向的最佳估算

本文评价了将地震试验中分析横波分裂时所用的通用的代数等价技术,双源累积技术(DCT)用于估算不同排列的微裂隙穿过的地层模型的裂隙走向精度。并用另一种分析方法,双分动源-检技术(DIT)进行了辅助分析,研究另一种公式表述的任何益处。该研究所设计的模型是在半空间之上覆盖有两组地层,而这些地层的裂隙走向与深度间的关系有三种情况:裂隙走向单一;上下层之间裂隙的走向突变;裂隙走向在这两组地层范围的连续增大。研究中分析了模型的合成垂直地震剖面(VSP)和反射资料。零偏和近零偏 VSP 以及反射剖面的合成数据是用全波模拟程序包计算的,该程序包用等效各向异性介质来做模拟排列成行的、垂直的、充满水的平行微裂隙的分布状况。这两种技术的估算结果对于裂隙走向不变的模型来说是一致的,但对裂隙走向变化的 VSP 资料则有差异。由 DIT 得到的两个角参量θ_g 和θ_S 的渐近特性看出。这些 VSP 资料在适当的情况下,当研究层的分裂横波间的时间延迟超过子波的峰周期时,可用于确定裂隙走向。在这种限定下,θ_g 趋于追随裂隙走向变化,θ_S 趋于某一常数,而 DCT 给出的是上、下层之间裂隙走向的一个使人误解的值。尽管DIT 的特点就一切情况而论尚未了解,但由 VSP 资料得到的θ_g 和θ_S 值看来总是在裂隙走向发生突变的点的附近分散。这点可用作地层剥层时的定性指示。在记录数据矩阵必定对称的情况下,这两种技术对于反射资料是一致的,但当裂隙走向存在变化时,仍然会对深层给出使人误解的结果。本文认为,为检测储层岩石裂隙性质而设计并分析试验结构时,一定要考虑到分析技术的响应和分辨率极限。这对那些将极化估算值直接解释为裂隙走向的人们提出了警告。
