Physicochemical and Rheological Characterization of an Acidic Milk Product: Kefir Concentration Effect

The main objective of this study was the analysis of physicochemical and rheological behavior of a drinkable beverage prepared with different concentrations (5%, 10% and 20% w/w) of Kefir. Several rheological models were also evaluated to determine the model that better fit experimental data. Apparent viscosity was measured with a Brookfield Viscometer within shear rates of 0.26 rad/s to 2.09 rad/s; and with an Ar-2000ex TA Instruments Rheometer within shear rates of 0.01 rad/s to 1,000 rad/s. Physicochemical tests, such as titratable acidity, chloride percent, fat concentration, color, humidity, syneresis, and total solids, were determined using laboratory equipment. The main result observed in all samples tested with Brookfield viscometer was that Kefir drink behaved as a Newtonian Fluid within shorter strain rates (0.26 rad/s to 2.09 rad/s). However, when the same samples were tested with Ar-2000ex Rheometer, a different behavior was observed: Over a wider range of shear rates, the fully non-Newtonian behavior of Kefir samples was discovered. Besides, additional variables such as shear and normal stresses, and loss and storage modulus, were analyzed with Ar-2000ex Rheometer. The rheological model that better fitted the experimental data was Cross model, followed by Power Law Model. Statistical analysis of viscosity data acquired with Brookfield Viscometer from all Kefir concentrations was not significant (p values greater than 0.05); which demonstrated that the effect of Kefir concentration over shorter strain rates was not significant, and thus, the samples behave as a Newtonian fluid. On the other hand, p values lower than 0.05 were observed in most of the different Kefir concentration samples tested with Ar-2000ex Rheometer; which indicates that a statistical significant effect was observed and thus a non-Newtonian behavior of Kefir samples. Background Extracellular vesicles(EVs) present in oviductal(OF) and uterine fluid(UF) have been shown to enhance bovine embryo quality during in vitro culture by reducing lipid contents and modulating lipid metabolism-related genes(LMGs), while also influencing cell proliferation, suggesting their involvement on the regulation of different biological pathways. The regulation of signaling pathways related to cell differentiation, proliferation, and metabo-lism is crucial for early embryo development and can determine the success or failure of the pregnancy. Bioactive molecules within EVs in maternal reproductive fluids, such as micro RNAs(miRNAs), may contribute to this regulatory process as they modulate gene expression through post-transcriptional *** This study evaluated miRNA cargo in OF-EVs from the early luteal phase and UF-EVs from the mid-luteal phase, coinciding with embryo transit within oviduct and uterus in vivo, and its possible influence on LMGs and sign-aling pathways crucial for early embryo development. A total of 333 miRNAs were detected, with 11 exclusive to OF, 59 to UF, and 263 were common between both groups. From the 20 differentially expressed miRNAs, 19 up-regulated in UF-EVs(bta-miR-134, bta-miR-151-3p, bta-miR-155, bta-miR-188, bta-miR-181b, bta-miR-181d, bta-miR-224, bta-miR-23b-3p, bta-miR-24-3p, bta-miR-27a-3p, bta-miR-29a, bta-miR-324, bta-miR-326, bta-miR-345-3p, bta-miR-410, bta-miR-652, bta-miR-677, bta-miR-873 and bta-miR-708) and one(bta-miR-148b) in OF-EVs. Shear Rate Effect on the Viscosity of Homemade and Commercial Tomato Sauce

Rheological characterization of tomato products is important not only for design of unit operations, but also to optimizeprocesses and guarantee high quality of food products. Time dependence is related with structural changes due to shear rate. Thus,rheological characterization as a function of time is extremely important to understand the changes that occurred in food productsduring commercial processes. However, these characterizations are rare in the literature for tomato products. Those rheologicalproperties depend of several parameters, such as agronomic, structural and process parameters. In this study, the effect of shear rateon the viscosity at room temperature （22 ~C） of two kinds of tomato sauce （homemade and commercial） is analyzed; using aBrookfield viscometer model 4535, Lab-Line Instruments. Homemade sauce was prepared with fresh saladette tomatoes from a localmarket. Rheological analysis was performed at shear rates from 0.05 to 10.47 sl; using all the spindles available. Statistical analysiswas made using LSD and Duncan tests with a confidence interval of 95% （p-value of 0.05）. It was demonstrated that homemade andcommercial tomato sauce behaved as a thixotropic fluid, due to the typical decrease on viscosity samples observed when testing timewas increased. When tomato sauce is at rest, it has a gelled tridimensional structure; and, as soon as a shear stress is applied, themovement generates an alignment of tomato constituent chains in flow direction, breaking physic entanglements and thus causing adecrease in viscosity as a function of time. Furthermore, it was discovered that the viscosity of homemade sauce was higher than theviscosity of commercial sauce. The latter may be due to the pre-thermal treatment. During this unit operation, polymeric chains oflycopene most likely break into smaller chains of isoprene, causing the observed decrease in viscosity. Compounds with dual action on cyclic adenosine monophosphate （cAMP） and cyclic guanosine monophosphate （cGMP） may be a treatment option for erectile dysfunction, as they not only promote penile erection but also prevent the upregulation of phosphodiesterase-5. In this study, we examined the possible relaxant effect and mechanism of 17-nor-subincanadine E （SEC, 0.2-200 pmol I^-1）, a plant-derived alkaloid, in rabbit corpus cavernosum （RbCC） strips that had been precontracted by exposure to phenylephrine （10 pmol I^- 1） or a high concentration of K＋（60 mmol I^- 1） in vitro. In addition to SEC＇s effect on cAMP and cGMP levels, electrical field stimulation （EFS） in phenylephrine-precontracted RbCC and calcium chloride （1-100 mmol I^-1） evoked responses in depolarized RbCC were analysed. SEC relaxed the phenylephrine-precontracted RbCCs in a concentration-dependent manner. Atropine, guanethidine and N-co-nitro-L-arginine methyl ester （L-NAME） did not have any effect on the relaxation of RBCCs. When 1H-1,2,4oxadiazole[4,3-a] quinoxalin-1-one （ODQ） was added, it effectively blocked the relaxant response of SEC. Although SEC enhanced the maximal relaxation produced by sodium nitroprusside （SNP） and forskolin in phenylephrine-precontracted cavernosal smooth muscle, it caused a decrease in the maximal contractile response induced by calcium chloride in depolarized RbCCs. The relaxant effect of SEC was paralleled by an increase in the tissue levels of the Cyclic nucleotides cAMP and cGMP. We conclude that SEC promotes the relaxation of RbCC, possibly favouring cAMP and cGMP accumulation and calcium blockade. This novel mechanism could be useful for patients who do not benefit from phosphodiesterase inhibitors and for those with endothelial and nitrergic dysfunction, such as patients with diabetes, hypertension and dyslipidaemias.

Acetylsalicylic Acid Administered in Patients with Type 2 Diabetes Mellitus and Its Effect on the Antioxidant Enzyme System

Type 2 diabetes mellitus and its complications are associated with oxidative stress and the depletion of antioxidant defenses. The influence of acetylsalicylic acid on reversing the decrease in antioxidants, insulin resistance, glucose homeostasis, and inflammatory cascade can help prevent diabetes complications. Purpose: The aim of the study was to evaluate the effect of acetylsalicylic acid on the antioxidant enzymatic system in patients with diabetes. Methods: A randomized, double-blind, placebo-controlled clinical trial was carried out in 21 patients of both sexes with Type 2 diabetes for less than five years at the time of diagnosis, without pharmacological treatment, and randomly selected. Acetylsalicylic acid (300 mg) was administered orally for three months to the study group (n = 11) compared to the placebo control group (n = 10). Before and after the intervention, anthropometric and metabolic measurements were taken, fasting plasma glucose, glycated hemoglobin A1c, lipid profile panel, glutathione peroxidase, superoxide dismutase, catalase, and antioxidant capacity/activity were determined;values are presented as mean ± standard deviation. Intra- and intergroup differences were tested by Wilcoxon signed rank and Mann-Whitney U test, respectively;p-value ≤ 0.05 was considered statistically significant. Results: The acetylsalicylic acid group showed a decrease in weight (85.6 ± 19.3 vs. 84.1 ± 19.0 kg p = 0.01), cholesterol (205.9 ± 16.6 vs. 186.0 ± 23.2 mg/dL p = 0.02), and glycated hemoglobin A1c (7.8% ± 0.9% vs. 7.0% ± 0.7% p = 0.02). The placebo group exhibited reduction in weight (76.1 ± 14.9 vs. 74.9 ± 15.0 kg p = 0.04), glycated hemoglobin A1c (6.9% ± 0.6% vs. 6.2% ± 0.4% p = 0.004), and total antioxidant capacity (4.1 ± 0.5 vs. 4.8 ± 0.3 mmol/L p = 0.002). Conclusion: The administration of acetylsalicylic acid did not modify the antioxidant enzyme system.

Clinical Facial Study of the Parents of Children with OAV (Oculo-Auriculo-Vertebral) Syndrome. Cases and Controls

Introduction: The oculo-auriculo-vertebral syndrome is a complex disease of the head and neck. Aim: The purpose of this study was to look for facial asymmetry, measuring both sides of the mandible and external ear in the parents of these patients in a clinical way, to see if it could be considered as a minor manifestation of the disease. Patients and Methods: A clinical, observational study was done with 50 trios and their parents’ controls, at the Instituto Nacional de Rehabilitación LGII in 2019, from January to May. Mandible and ear measurements were done. Results: All of our patients had microtia and facial asymmetry, and in 17 cases, other anomalies. We compared both sides of the mandible in the patients’ parents and in the parents’ controls. The results showed differences only in the fathers, and in the comparison with the controls, we found statistical differences only in women, family and non-family cases. The ears did not show any differences. Discussion: It is very interesting that we found differences in the measurements of the mandible of the patients’ mothers and their controls;in the mothers the mandible was bigger than in the controls, this probably means that the development of the cranial structures is modified by some genetic variants. No differences were found in the measurements of the ears. Conclusion: It is very difficult to know exactly if the differences we found are important or not. The clinical measurements could be a good option to establish if the parents have minor manifestations that could explain some heritability.
