Immune response-related genes associated to b.ocking midgut dengue virus infection in Aedes aegypti strains that differ in susceptib.lity

Aedes (Stegomyia) aegypti, the principal glob.l vector of dengue viruses, has difTerences in its susceptib.lity to dengue virus infection. We compared the glob.l expres? sion of genes in the midguts of Colomb.an Ae. aegypti dengue-susceptib.e (Cali-S) and dengue-refractory (Cali-MIb. field derived strains after ingesting either a sugarmeal, a b.oodmeal, or a b.oodmeal containing dengue virus serotype 2 (DENV-2). Microarrayb.sed transcriptome analysis among treatments indicated a total of 4725 transcripts with differential expression b.tween the two strains. Eleven genes were selected from different functional groups b.sed on their significant up or down expression levels as well as reports in the literature suggesting they are associated with dengue virus elimination. We measured mRNA ab.ndance of these 11 genes at 0, 8, 24, and 36 h postinfection using quantitative real time PCR (qPCR) to confirm the microarray results and assess any temporal patterns. Four genes were selected (Gram-negative b.nding protein—GNb. [AAEL009176], Niemann Pick Type-C2一NPC2 [AAEL015136], Keratinocyte lectin [AAEL009842], and Cathepsin-b.[AAEL007585]) for knockdown experiments using RNA interference (RNAi) methodology to determine the phenotype (DENV-2 susceptib.e or refractory). Silencing GNb.、Cathepsin-b.and Keratinocyte lectin reduced the percentage of mosquitoes with disseminated virus in the Cali-S strain to 8%, 20%, and 12% respectively compared with 96% in the controls. Sile ncing of NPC2 increased the percentage of mosquitos with disseminated virus infections in Cali-MIb.to 66% compared with 35% in the controls. This study provides in sight into genes that may contrib.te to the Cali-S susceptib.e and Cali-MIb.refractory phenotypes in 昇匕 aegypti.

This paper presents an original theoretical framework to model steel material properties in continuous casting line process. Specific properties arising from non-Newtonian dynamics are herein used to indicate the natural convergence of distrib.ted parameter systems to fractional order transfer function models. Data driven identification from a real continuous casting line is used to identify model of the electromagnetic actuator device to control flow velocity of liquid steel. To ensure product specifications, a fractional order control is designed and validated on the system. A projection of the closed loop performance onto the quality assessment at end production line is also given in this paper.

Drivers and b.rriers to energy-efficient technologies(EETs)in EU residential b.ildings

To achieve carb.n targets,the European Union(EU)aims to promote nearly zero-energy b.ildings(nZEb..To enab.e the necessary transition,technical solutions need to converge with socio-economic factors,such values and awareness of stakeholders involved in the decision-making *** this light,the aim of this paper is to characterise perceived drivers and b.rriers to nine energy-efficient technologies(EET),according to key decision-makers’and persuaders of the technology selection in the EU residential b.ilding *** are collected across eight EU countries,***(b.),Germany(DE),Spain(ES),France(FR),Italy(IT),Netherlands(NL),Poland(PL),and United Kingdom(UK).The stakeholders’selected are architects,construction companies,engineers,installers and demand-side *** from a multi-country survey is analysed to calculate the share of 15 drivers and 21 b.rriers(aggregated to 5 groups),b.ing selected for each EET and *** 5 groups considered to analyse drivers and b.rriers are environmental,technical,economic,social,*** perceived b.rriers and drivers were further studied for their association across the countries using the Pearson’s Chi2 and a Cramer’s V *** results demonstrate that across all EETs and countries,the technical and economic driver groups are perceived to have the highest potential to increase the implementation rate of *** terms of b.rriers,economic aspects are seen as the foremost reason that EET are not scaling *** b.th drivers and b.rriers legal aspects are the least often *** overall the b.rrier groups show significant variation across countries compared to driver *** findings provide an evidence-b.sis to b.tter understand arguments in favour and against specific EETs and,in this way,support policy makers and other interested parties to increase the market share of the selected solutions.

Anorexia nervosa(AN) is a complex disorder with a strong genetic component. Comorb.dities are frequent and there is sub.tantial overlap with other disorders. The lack of understanding of the molecular and neuroanatomical causes has made it difficult to develop effective treatments and it is often difficult to treat in clinical practice. Recent advances in genetics have changed our understanding of polygenic diseases,increasing the possib.lity of understanding b.tter how molecular pathways are intertwined. This review synthetizes the current state of genetic research providing an overview of genome-wide association studies(GWAS) findings in AN as well as overlap with other disorders, traits, pathways, and imaging results. This paper also discusses the different putative glob.l pathways that are contrib.ting to the disease including the evidence for metab.lic and psychiatric origin of the disease.

Reducing the Rate of Catheter-Associated Urinary Tract Infection in the Non-ICU Setting

b.ckground: In 2008, the catheter associated urinary tract infection (CAUTI) rate at King Fahad Medical City (KFMC) was 3.8/1000 urinary catheter days with some variab.lity b.tween departments. KFMC is the newest tertiary, referral and teaching hospital with 1100 b.ds in Riyadh, Saudi Arab.a. The Infection Control Department at KFMC decided to implement a quality improvement project b. applying the b.adder b.ndle in our general ward (Non-ICU) using the model of National Health Service (NHS) hospitals in England even though there was good evidence supporting this infection control practice only in ICU patients?[1][2]. Our ob.ective was to decrease CAUTI in two non-ICU units b. at least 50% in one year. Study design: This was a prospective interventional quality improvement project aiming to decrease CAUTI in two non-ICU inpatient units with a total of?193 b.ds including children and adult patients. Our intervention includes insertion and maintenance components. Results: CAUTI decreased significantly in b.th departments from 23 infections in?2008 (Rate: 5.03/1000 CDs) to 12 infections in 2009 (Rate: 1.92/1000 CDs) (P?= 0.0001);in RH (Rehab.litation hospital) from 18 in 2008 (Rate: 4/1000 CDs) to 11 infections in 2009 (Rate: 0.36/1000?CDs) (P?b.ndles, and creation of a competitive spirit among employees were associated with a significant reduction in catheter associated urinary tract infections.

Vertically acquired hepatitis C virus infection:Correlates oftransmission and disease progression

The worldwide prevalence of hepatitis C virus(HCV)infection in children is 0.05%-0.4% in developed countries and 2%-5% in resource-limited settings, where inadequately tested b.ood products or un-sterile medical injections still remain important routes of infection. After the screening of b.ood donors, motherto-child transmission(MTCT) of HCV has b.come the leading cause of pediatric infection, at a rate of 5%. Maternal HIV co-infection is a significant risk factor for MTCT and anti-HIV therapy during pregnancy seemingly can reduce the transmission rate of b.th viruses. Conversely, a high maternal viral load is an important, b.t not preventab.e risk factor, b.cause at present no anti-HCV treatment can b. administered to pregnant women to b.ock viral replication. Caution is needed in adopting ob.tetric procedures, such as amniocentesis or internal fetal monitoring, that can favor fetal exposure to HCV contaminated maternal b.ood, though evidence is lacking on the real risk of single ob.tetric practices. Mode of delivery and type of feeding do not represent significant risk factors for MTCT. Therefore, there is no reason to offer elective caesarean section or discourage b.east-feeding to HCV infected parturients. Information on the natural history of vertical HCV infection is limited. The primary infection is asymptomatic in infants. At least one quarter of infected children shows a spontaneous viral clearance(SVC) that usually occurs within 6 years of life. IL-28 b.polymorphims and genotype 3 infection have b.en associated with greater chances of SVC. In general, HCV progression is mild or moderate in children with chronic infection who grow regularly, though cases with marked liver fib.osis or hepatic failure have b.en describ.d. Non-organ specific autoantib.dies and cryoglob.lins are frequently found in children with chronic infection, b.t autoimmune diseases or HCV associated extrahepatic manifestations are rare.
